Hereditary complement deficiencies are usually associated with increased susceptibility to infections and/ or autoimmune diseases. Generally, they are rare： the majority have been detected in no more than a few dozen individuals. However, due to lack of population-based studies, such cases are usually detected by chance, e.g., in severely ill persons and their family members. An exception is mannan-binding lectin （MBL） deficiency, affecting perhaps 5%-10% of the population. MBL, like collectin-10, -11 and the ficolins （M-, L-, H-）, is a pattern-recognition molecule, cooperating with MBL- associated serine proteases （MASPs） in the initiation of complement activation via the lectin pathway. Root and Foot Rot Diseases of Winter Wheat Grown in Conventional and Organic Systems

The object of the study was fungous diseases occurring on roots, leave sheaths and stem base of winter wheat in the two opposing cropping systems (organic and conventional). The observations were made in vegetation periods (2007-2009) in the fields of winter wheat in northern Poland. Every year on each plot of compared farming systems root rot occurred (Fusarium spp., Gaeumannomyces graminis and other fungi). For the period of 3 years the degree of disease injury on the roots of winter wheat grown in the conventional system in the vegetation period increased, while in the organic one remained on pretty the same level. On average a lot more affected roots, especially in the flowering stage, occurred on the winter wheat grown in the conventional system. Fusarium foot rot (Fusarium spp.) developed on the wheat during the entire vegetation period. It was the most dangerous root and foot rot disease (the highest indexes of injury). The mean degree of disease injury on leave sheath was on pretty the same level in the two farming systems, although in investigated vegetation periods differed a lot, whereas at the bases of stems the pathogen was on the higher level on the wheat in the conventional system. Also eyespot (Tapesia yallude) developed in the entire vegetation period of the winter wheat, but its intensity was much lower than in case of fusarium foot rot. Leave sheaths of the wheat grown in the conventional system were slightly stronger affected than those grown in the organic system. In the flowering stage the intensity of the disease in both farming systems became equal, while in the wax maturity it was considerably higher in the conventional system. Sharp eyespot (Rhizoctonia spp.) appeared relatively late and occurred only in two years of investigation. The intensiveness of the disease was definitely higher on the organic plots. AIM： To screen for genes related to metabotropic re- ceptors that might be involved in the development of chronic hepatitis. METHODS： Assessment of 20 genes associated with metabotropic receptors was performed in liver speci- mens obtained by punch biopsy from 12 patients with autoimmune and chronic hepatitis type B and C. For this purpose, a microarray with low integrity grade and with oligonucleotide DNA probes complementary to target transcripts was used. Evaluation of gene expression was performed in relation to transcript level, correlation between samples and grouping of clinical parameters used in chronic hepatitis assessment. Clini- cal markers of chronic hepatitis included alanine and aspartate aminotransferase, ~,-glutamyltranspeptidase, alkaline phosphatase and cholinesterase activity, levels of iron ions, total cholesterol, triglycerides, albumin, glucose, hemoglobin, platelets, histological analysis of inflammatory and necrotic status, fibrosis according to METAVIR score, steatosis, as well as anthropometric body mass index, waist/hip index, percentage of adi- pose tissue and liver size in ultrasound examination. Gender, age, concomitant diseases and drugs were also taken into account. Validation of oligonucleotide microarray gene expression results was done with the use of quantitative real-time polymerase chain reaction （qRT-PCR）. RESULTS： The highest （0.002 〈 P 〈 0.046） expres- sion among genes encoding main components of metabotropic receptor pathways, such as the a subunit of G-coupled protein, phosphoinositol-dependent pro- tein kinase or arrestin was comparable to that of an- giotensinogen synthesized in the liver. Carcinogenesis suppressor genes, such as chemokine ligand 4, tran- scription factor early growth response protein 1 and lysophosphatidic acid receptor, were characterized by the lowest expression （0.002 〈 P 〈 0.046）, while the factor potentially triggering hepatic cancer, transcrip- tion factor JUN-B, had a 20-fold higher expression. Endothelial dysfunction is considered one of the etiological factors of inflammatory bowel disease(IBD). An inflammatory process leads to functional and structural changes in the vascular endothelium. An increase of leukocyte adhesiveness and leukocyte diapedesis, as well as an increased vascular smooth muscle tone and procoagulant activity is observed. Structural changes of the vascular endothelium comprise as well capillary and venule remodeling and proliferation of endothelial cells. Hypoxia in the inflammatory area stimulates angiogenesis by upregulation of vascular endothelial growth factor, fibroblast growth factor and tumor necrosis factor-α. Inflammatory mediators also alter the lymphatic vessel function and impair lymph flow, exacerbating tissue edema and accumulation of dead cells and bacteria. The endothelial dysfunction might be diagnosed by the use of two main methods: physical and biochemical. Physical methods are based on the assessment of large arteries vasodilatation in response to an increased flow and receptors stimulation. Flowmediated vasodilatation(FMD) is the method that is the most widely used; however, it is less sensitive in detecting early changes of the endothelium function. Most of the studies demonstrated a decrease of FMD in IBD patients but no changes in the carotic intima-media thickness. Biochemical methods of detecting the endothelial dysfunction are based on the assessment of the synthesis of compounds produced both by the normal and damaged endothelium. The endothelial dysfunction is considered an initial step in the pathogenesis of atherosclerosis in the general population. In IBD patients, the risk of cardiovascular diseases is controversial. Large, prospective studies are needed to establish the role of particular medications or dietary elements in the endothelial dysfunction as well to determine the real risk of cardiovascular diseases.

AIM:To assess the mucosa-associated bacterial microflora and mucus layer in adolescents with inflammatory bowel disease(IBD) .METHODS:Sixty-one adolescents(mean age 15 years,SD ± 4.13) were included in the *** biopsies from inflamed and non-inflamed mucosa of IBD patients and from controls with functional abdominal pain were cultured under aerobic and anaerobic *** number of microbes belonging to the same group was calculated per weight of collected *** mucus thickness in frozen samples was measured under a fluorescent ***:The ratios of different bacterial groups in inflamed and non-inflamed mucosa of IBD patients and controls were specific for particular *** *** predominant in the inflamed mucosa of Crohn's disease(CD) patients(80% of all bacteria) ,and Lactobacillus *** predominant in ulcerative colitis patients(90%) .The differences were statistically significant(P = 0.01-0.001) .Lower number of bifidobacteria was observed in the whole IBD group.A relation was also found between clinical and endoscopic severity and decreased numbers of Lactobacillus and,to a lesser extent,of Streptococcus in biopsies from CD *** mucus layer in the inflamed sites was significantly thinner as compared to controls(P = 0.0033) and to non-inflamed areas in IBD patients(P = 0.031) .CONCLUSION:The significantly thinner mucosa of IBD patients showed a predominance of some aerobes specific for particular diseases,their numbers decreased in relation to higher clinical and endoscopic activity of the disease.

Intracellular protein routing is mediated by vesicular transport which is tightly regulated in eukaryotes. The protein and lipid homeostasis depends on coordinated delivery of de novo synthesized or recycled cargoes to the plasma membrane by exocytosis and their subsequent removal by rerouting them for recycling or degradation. Here, we report the characterization of protein affected trafficking 3 （pat3） mutant that we identified by an epifluorescence-based for- ward genetic screen for mutants defective in subcellular distribution of Arabidopsis auxin transporter PIN1-GFR While pat3 displays largely normal plant morphology and development in nutrient-rich conditions, it shows strong ectopic intracellular accumulations of different plasma membrane cargoes in structures that resemble prevacuolar compart- ments （PVC） with an aberrant morphology. Genetic mapping revealed that pat3 is defective in vacuolar protein sorting 35A （VPS35A）, a putative subunit of the retromer complex that mediates retrograde trafficking between the PVC and trans-Golgi network. Similarly, a mutant defective in another retromer subunit, vps29, shows comparable subcellular defects in PVC morphology and protein accumulation. Thus, our data provide evidence that the retromer components VPS35A and VPS29 are essential for normal PVC morphology and normal trafficking of plasma membrane proteins in plants. In addition, we show that, out of the three VPS35 retromer subunits present in Arabidopsis thaliana genome, the VPS35 homolog A plays a prevailing role in trafficking to the lyric vacuole, presenting another level of complexity in the retromer-dependent vacuolar sorting.

目的:探讨遗传算法的局限性和实用性,并分析基于相互作用产生的上位效应对遗传算法可靠性的影响。创新点:1.指出遗传算法缺陷的根源;2.基于测试样本函数定义目标函数,以判断遗传算法的适用性。方法:1.基于非上位效应函数(表1)和上位效应函数(表2),以及非上位效应函数F4和上位效应函数F6的结构图来验证遗传算法可靠性;2.通过计算样本函数(公式(1))和遗传算法流程(图3)表达遗传算法的工作原理。3.利用克洛弗函数(公式(2))和计算不同结构角下的函数分布(图4),进一步判断匹配度(表3)和计算效率(表4);定义新的目标函数(公式(9))和一组新的变量(公式(10))来实现变量相关性解离。结论:1.对当前遗传算法存在的不足给出了独到见解,并认为正定性的假设并非可以保证遗传算法实际的有效性和优化性。2.定义成本代价函数用以判断遗传算法可靠性,并分别考虑上位性和非上位性效应两种情形。当成本代价函数在非上位性效应下时,遗传算法是有效的;否则,可以把N维函数降级为N个一维函数,从而采用更简单的算法来判断。基于一些通用的基准,进一步设计三类样本函数来证实以上判断,且这些样本函数适合于上位性效应情形和非上位效应情形。3.遗传算法的瓶颈在于主算子和相干匹配性;可以通过破坏某些结构来实现变量关系的解离,从而抑制相干匹配性对遗传算法的影响。希望相关读者在处理实际优化问题时能验证作者关于上位效应的定性结论,并给出更可靠的方法来表征这种效应。

The Use of Liquid Crystal Thermography in Selected Technical and Medical Applications—Recent Development

Thermochromic liquid crystals (TLC) and true-colour digital image processing have been successfully used in non-intrusive technical, industrial and biomedical studies and applications. Thin coatings of TLC at surfaces are utilized to obtain detailed temperature distributions and heat transfer rates for steady or transient processes. Liquid crystals also can be used to make the temperature and velocity fields in liquids visible by the simple expedient of directly mixing the liquid crystal material into the liquid (water, glycerol, glycol, and silicone oils) in very small quantities to use as thermal and hydrodynamic tracers. In biomedical situations, e.g., skin diseases, breast cancer, blood circulation and other medical application, TLC and image processing are successfully used as an additional non-invasive diagnostic method especially useful for screening large groups of potential patients. The history of this technique is reviewed, principal methods and tools are described and some examples are presented. Also steady-state and transient liquid crystal thermography (LCT) is used to measure local heat transfer on a plate equipped with transverse vortex generators. Automated evaluation allows determining the heat transfer coefficient without arbitrary influence of human interpretation.
