Auxin directs plant ontogenesis via differential accumulation within tissues depending largely on the activity of PIN proteins that mediate auxin efflux from cells and its directional cell-to-cell transport. Regard- less of the developmental importance of PINs, the structure of these transporters is poorly characterized. Here, we present experimental data concerning protein topology of plasma membrane-localized PINs. Utilizing approaches based on pH-dependent quenching of fluorescent reporters combined with immuno- localization techniques, we mapped the membrane topology of PINs and further cross-validated our results using available topology modeling software. We delineated the topology of PIN1 with two transmembrane （TM） bundles of five m-helices linked by a large intracellular loop and a C-terminus positioned outside the cytoplasm. Using constraints derived from our experimental data, we also provide an updated position of helical regions generating a verisimilitude model of PIN1. Since the canonical long PINs show a high degree of conservation in TM domains and auxin transport capacity has been demonstrated for Arabidopsis representatives of this group, this empirically enhanced topological model of PIN1 will be an important starting point for further studies on PIN structure-function relationships. In addition, we have established protocols that can be used to probe the topology of other plasma membrane proteins in plants.

Versatile optimization of the synthesis method and composition of Yb^3＋ and Tm^3＋ co-doped CaF2 nanoparticles as well as a novel biofunctionalization method were developed and evaluated. Through multistep synthesis, the luminescence intensity of the Tm3. activator was enhanced by more than 10-fold compared to standard one-step synthesis. The proposed methods were used to homogenously distribute the doping ions within the nanoparticle＇s volume and thus reduce luminescence quenching. Optimization of dopant ions concentration led to the selection of the most efficient visible and near-infrared up-converting nanoparticles, which were CaF2 doped with 10% Yb^3＋ 0.05% Tm^3＋ and 20% Yb3. 0.5% Tm^3＋, respectively. To illustrate the suitability of the synthesized nanoparticles as bio-labels, a dedicated biofunctionalization method was used, and the nanoparticles were applied for labeling and imaging of Candida albicans cells. This method shows great promise because of extremely low background and high specificity because of the presence of the attached molecules.

In this context,four specimens,i.e.(i)circumferentially notched cylindrical torsion(CNCT),(ii)circum-ferentially notched cylindrical direct tension(CNCDT),(iii)edge notch disc bend(ENDB)and(iv)three-point bend beam(3PBB),were utilized to measure the modesⅠandⅢfracture toughness values of *** the CNCT specimen provides pure modeⅢloading in a direct manner,this pure mode condition is indirectly produced by the ENDB *** ENDB specimen provided lower KⅢc and a non-coplanar(***)fracture surface compared with the CNCT specimen,which showed a planar modeⅢfracture *** ENDB specimen is also employed for conducting pure modeⅠ(with different crack depths)and mixed modeⅠ/Ⅲ*** value was independent of the notch depth,and it was consistent with the RILEM and ASTM standard *** the modeⅢfracture results were highly sensitive to the notch *** the fracture resistance against modeⅢwas significantly lower than that of modeⅠ,the greater work of fracture under modeⅢwas noticeable.

The endocannabinoid system(ECS),particularly its signaling pathways and ligands,has garnered considerable interest in recent *** with clinical work investigating the ECS’functions,including its role in the development of neurological and inflammatory conditions,much research has focused on developing analytical protocols enabling the precise monitoring of the levels and metabolism of the most potent ECS ligands:exogenous phytocannabinoids(PCs)and endogenous cannabinoids(endocannabinoids,ECs).Solid-phase microextraction(SPME)is an advanced,non-exhaustive sample-preparation technique that facilitates the precise and efficient isolation of trace amounts of analytes,thus making it appealing for the analysis of PCs and ECs in complex matrices of plant and animal/human *** this paper,we review recent forensic medicine and toxicological studies wherein SPME has been applied to monitor levels of PCs and ECs in complex matrices,determine their effects on organism physiology,and assess their role in the development of several diseases.

AIM: To evaluate the diagnostic efficiency of endoscopic ultrasound (EUS) as the main imaging modality in patients with moderate suspicion of common bile duct stones (CBDS).METHODS: 55 patients with moderate clinical suspicion of CBDS were prospectively included to the study and evaluated with EUS. This study was done in single blind method in the clinical and biochemical data of patients. EUS was done with echo-endoscope Pentax FG 32-UA (f=5-7,5 MHz) and Hitachi EUB 405 ultrasound machine. Patients diagnosed with CBDS by EUS were excluded from this study and treated with ERC. All the other patients were included to the follow up study obtained by mail every 6 months for clinical evaluation (need of ERC or surgery).RESULTS: CBDS was found in 4 patients by EUS. Diagnosis was confirmed in all cases on ERC. The remaining 51 patients without CBDS on EUS were followed up for 6-26 months (meanly 13 months) There were: 40 women, 42cholecystectomized patients, aged: 55 (mean). Biochemical values (mean values) were as follows: bilirubin: 14,9 μmol.L-1,alkaline phosphatase: 95 IU.L-,1 γ-GTP: 131 IU.L-1, ALT: 50IU.L-1, AST: 49 IU.L-1, Only 1 patient was lost for follow *** the remaining 50 patients with follow up, there was only 1 (2 %) patient with persistent biliary symptoms in whom CBDS was finally diagnosed by ERC with ES. All other patients remained symptoms free on follow up and did not require ERC or biliary ***: Vast majority of patients with moderate suspicion of CBDS and no stones on EUS with linear array can avoid invasive evaluation of biliary tree with ERC.

Uncovering microstructure evolution mechanisms that accompany the long-term operation of solid oxide fuel cells is a fundamental challenge in designing a more durable energy system for the *** date,the study of fuel cell stack degradation has focused mainly on electrochemical performance and,more rarely,on averaged microstructural *** we show an alternative approach in which an evolution of three-dimensional microstructural features is studied using electron tomography coupled with topological data *** latter produces persistent diagrams of microstructure before and after long-term operation of *** diagrams unveil a new insight into the degradation process of three involved phases:Nickel,pores,and yttrium-stabilized zirconium.

Effect of the C:N:P ratio on the denitrifying dephosphatation in a sequencing batch biofilm reactor(SBBR)

A series of investigations were conducted using sequencing batch biofilm reactor（SBBR） to explore the influence of C：N：P ratio on biological dephosphatation including the denitrifying dephosphatation and the denitrification *** in the reactor occurred mainly in the form of a biofilm attached to completely submerged *** acid was used as the source of organic carbon.C：N：P ratios have had a significant effect on the profiles of phosphate release and phosphate uptake and nitrogen *** highest rates of phosphate release and phosphate uptake were recorded at the C：N：P ratio of 140：70：*** C：N ratio of 2.5：1 ensured complete *** highest rate of denitrification was achieved at the C：N：P ratio of 140：35：*** increase of nitrogen load caused an increase in phosphates removal until a ratio C：N：P of 140：140：*** of the biofilm exposed to alternate conditions of mixing and aeration exhibited enhanced intracellular accumulation of ***,the structure of the biofilm encouraged anaerobic-aerobic as well as anoxic-anaerobic and absolutely anaerobic conditions in a *** heterogeneous conditions in the presence of nitrates may be a significant factor determining the promotion of denitrifying polyphosphate accumulating organism（DNPAO） development.

To evaluate blood concentration of melatonin and urinary excretion of its metabolite, 6-sulfatoxymelatonin （6-OHMS）, in functional dyspepsia （FD）. METHODS： Ninety individuals were enrolled in the study： 30 in each study group： patients with postprandial distress syndrome （PDS）, epigastric pain syndrome （EPS）, and controls. Blood samples were drawn at 02：00 and 09：00 h and 24-h urine collection was performed. Serum melatonin and urinary 6-OHMS concentrations were measured by enzyme-linked immunosorbent assay. RESULTS： Serum melatonin concentration at night and in the morning was significantly （P 〈 0.001） higher inPDS patients [at 02：00 h-93.3 pg/mL, quartile range （QR）： 79.8-116.2; at 09.00 h-14.3 pg/mL, QR： 7.06-19.0] than in EPS （57.2 pg/mL, QR： 42.6-73.1, 8.1 pg/mL, QR： 4.2-9.3） and control patients （57.7 pg/mL, QR： 51.2-62.5; 8.1 pg/mL, QR： 5.4-10.3）. A similar relationship was observed for urinary 6-OHMS excretion. Patients with severe PDS symptoms had a higher melatonin concentration than these with moderate syndromes, whereas patients with severe EPS had a lower urinary 6-OHMS excretion than patients with moderate ***： Evaluation of melatonin serum concentrations and 24-h urinary 6-OHMS excretion are useful methods for differential diagnosis of various clinical forms of FD.
