FGFR3 alterations in bladder cancer:Sensitivity and resistance to targeted therapies

＜正＞ Objective: Cellular drug resistance measured at initial diagnosis is associated with an increased relapse risk and unfavorable clinical outcome in childhood ALL. In addition, the presence of adverse clinical prognostic factors such as age. and pro - B and T - lineage immunophenotype have been shown to be associated with cellular resistance to drugs in children with ALL. P - glycoprotein （Pgp）, the gene product of MDR1, confers multidrug resistance to a number of antineoplastic agents. A silent mutation in the exon 26 （C3435T） has been associated with altered expression and function of Pgp in tissues. Methods: MIT cytotoxicity assay, PCR alnalysis of C3435T polymorphism in MDR1 gene. Results: We compared the impact of C3435T polymorphism on in vitro chemosensitivity of leukemia cells to methylprednisolone. CC genotype carriers showed higher IC50 values in comparison with the carriers of CT or TT genotype. Statistical analysis Mann- Whitney U test showed P = 0.035. Conclusions: The group of patients with CC genotype seems to be more resistant to glucocorticoids, or at least methyprednisolone.

In this review,we revisit the pivotal role of fibroblast growth factor receptor 3(FGFR3)in bladder cancer(BLCA),underscoring its prevalence in both nonmuscle-invasive and muscle-invasive forms of the ***3 mutations in up to half of BLCAs play a well-established role in tumorigenesis,shaping distinct tumor initiation patterns and impacting the tumor microenvironment(TME).Emphasizing the importance of considering epithelial-mesenchymal transition profile and TME status,we revisit their relevance in predicting responses to immune checkpoint inhibitors in FGFR3-mutated *** writing highlights the initially promising yet transient efficacy of the FGFR inhibitor Erdafitinib on FGFR3-mutated BLCA,stressing the pressing need to unravel resistance mechanisms and identify co-targets for future combinatorial studies.A thorough analysis of recent preclinical and clinical evidence reveals resistance mechanisms,including secondarymutations,epigenetic alterations in pathway effectors,phenotypic heterogeneity,and population-specific variations within FGFR3 mutational ***,we discuss the potential of combinatorial treatments and concepts like synthetic lethality for discovering more effective targeted therapies against FGFR3-mutated BLCA.

Studying the membrane physiology of filamentous fungi is key to understanding their interactions with the environment and crucial for developing new therapeutic strategies for disease-causing ***,their plasma membrane has been inaccessible for a micron-sized patch-clamp pipette for pA current recordings due to the rigid chitinous cell ***,we report the first femtosecond IR laser nanosurgery of the cell wall of the filamentous fungi,which enabled patch-clamp measurements on protoplasts released from hyphae.A reproducible and highly precise(diffraction-limited,submicron resolution)method for obtaining viable released protoplasts was *** release from the nanosurgery-generated incisions in the cell wall was achieved from different regions of the *** plasma membrane of the obtained protoplasts formed tight and high-resistance(GΩ)contacts with the recording *** entire nanosurgical procedure followed by the patch-clamp technique could be completed in less than 1 *** to previous studies using heterologously expressed channels,this technique provides the opportunity to identify new ionic currents and to study the properties of the ion channels in the protoplasts of filamentous fungi in their native environment.

This study aims to apply a methodology to analyze the regulatory obstacles for the development of thermal power plants in a country. The study uses Brazil as an application case, although mostly of the findings can be useful to other countries. It also discusses other policies and actions by the government that may contribute to the growth of natural gas-fired power plants. Using deductive and comparative research methods, the first part of this method focuses on the stage of thermoelectric power generation in Brazil. The subsequent sections present the role of the integration of gas and electricity sectors, the main obstacles to the expansion of natural-gas power generation, how the country has deal with the expansion of natural-gas power generation, as well as how other obstacles has been faced. In raising the research question, when comparing the research results of selected countries, it should be noticed the impact that the experience and lessons elsewhere have had on policies formulation on Brazil. Our conclusions pointed out that adequate policies for the natural gas-electricity Brazilian market integration make advisable the unification of electricity and natural gas agencies. We recommended developing a mathematical model to support decision-making on natural gas and electricity integration.

The main goal of this paper is to establish necessary and sufficient conditions for a matrix A ∈（（lp）T,l∞）, where T is an arbitrary triangle, 1 ≤ p ≤∞, to be a compact operator. In the past,only sufficient conditions were established in almost all of those cases, by using the Hausdorff measure of noncompactness. We improve those results by applying another method for the characterizations of compact linear operators between BK spaces.

The present study was set up to examine the effect of alloying elements （including harmful elements） on metallurgical features （material properties and qualitative parameters） of tin bronzes, with particular reference to church bells from Middle Ages to Current times. A driving force of this study was to identify and demonstrate features related to the quality of church bells made in different centuries. The findings have been derived via metallographic and chemical analysis of specimens of bells from various parts of Australasia and Europe. The bell materials consisted of a mixture of the a phase and the （α＋β） eutectoid essentially, in proportions determined by tin content and mould materials during casting. The samples from the 15th century to the one from the 20th century showed a progressive increase in hardness, ranging from the minimum of -280 VHM20g to a maximum of -470 VHM20g for the （α＋β） eutectoid, and -160 VHM20g to -230 VHM20g for the a phase. The investigation also shows that the sound decay of the bell decreased with lowering the wt.% of tin and increasing the wt.% of lead and silver. This information is expected to provide an additional interesting knowledge into manufacturing practices and their significance in the quality of church bells over past centuries.

The general impact of“Eastern”(***,Egyptian and Levantine)cosmological ideas and astronomical methods on the emergence of early Greek philosophy and science is still *** this context,it is often stated that the Greeks were inspired by“foreign”ideas that helped them to free their minds from the determinations of their own culture of wisdom and knowledge.1 However,it is extremely hard to determine how this intellectual exchange worked in *** Greeks themselves seem to have assumed that they were highly indebted to older,more advanced Eastern cultures,as numerous stories about the fruitful encounters between early thinkers and typical places and agents of“oriental wisdom”illustrate.2 Most of these stories are connected to Ionia,where journeys to Phoenicia and Egypt began and where intercultural contacts must have been a daily experience:in agreement with Christoph Ulf,we may describe Ionia as a zone of intense,intimate contact between various different cultures.3 The fact that all the earliest Greek natural philosophers and investigators,from Thales to Heraclitus,were Ionian perfectly fits into this picture of Ionia as a stimulating meeting place and a centre of trade and exchange of ideas.

Adaptability of language-related brain network in a low-grade glioma patient

Because functional magnetic resonance imaging can be used for dynamic observation of functional cortical changes after brain injuries, we followed up functional magnetic resonance imaging manifestations of a language-related brain network in a low-grade glioma patient. Disease progression and therapy during a 3-year period were followed up at different time points： before and after reoperation, after radiation therapy, and 1 year after irradiation. During the whole 3-year follow-up period, the patient exhibited no neurological deficits while functional magnetic resonance imaging revealed different topologies of the language-related brain network. During disease progression and after irradiation, the language-related brain network was extended or completely transferred to the nondominant （right） hemisphere. In addition, after reoperation and 1 year after irradiation, language areas were primarily found in the language dominant （left） hemisphere. Our results suggest a high level of adaptability of the language-related cortical network of the bilateral hemispheres in this low-grade glioma patient.
