Salvadora persica extract attenuates cyclophosphamide-induced hepatorenal damage by modulating oxidative stress, inflammation and apoptosis in rats

Objective: Salvadora persica(SP) is used as a food additive and is a common ingredient in folk *** study investigates the antioxidant, anti-inflammatory, and beneficial effects of SP against cyclophosphamide(CYP) toxicity in ***: In a 10-day study, 32 male rats were equally allocated into 4 groups(8 rats/group) as follows:the normal control(NC group), normal rats that only received oral aqueous extract of SP(100 mg/[kg·d];SP group), animals treated with intraperitoneal CYP injections(30 mg/[kg·d];CYP group), and the CYP + SP group that concurrently received CYP with SP aqueous extract. Serum samples were collected to measure the liver and renal biochemical profiles, as well as antioxidant and oxidative stress markers and the concentrations of interleukin-1β(IL-1β), IL-6, IL-10, tumor necrosis factor-a(TNF-a), nuclear factor-κB(NF-κB) and adenosine 5’-monophosphate-activated protein kinase(AMPK). Hepatic and renal tissues were also harvested for histopathology and to measure apoptosis using the terminal deoxynucleotidyl transferase-mediated dUTP nick end labeling technique, alongside tissue levels of oxidative stress ***: Liver enzymes, total bilirubin, creatinine and urea, as well as serum IL-1β, IL-6, TNF-a and NF-κB increased significantly, whilst total protein, albumin, calcium, IL-10 and AMPK declined in serum of the CYP group relative to the NC group. The hepatorenal concentrations of glutathione, glutathione peroxidase and catalase declined markedly in the CYP group, whereas malondialdehyde, protein adducts, and apoptosis index increased compared with the NC group. By contrast, the hepatorenal biochemistry and apoptosis index of the SP group were comparable to the NC group. Interestingly, the CYP + SP group had significant improvements in the liver and renal biochemical parameters, enhanced anti-oxidative and anti-inflammatory effects, and marked declines in hepatic and renal apoptosis relative to the CYP group. Simulations of a Novel All-Optical Flip-Flop Based on a Nonlinear DFB Laser Cavity Using GPGPU Computing

A new all-optical flip-flop based on a nonlinear Distributed feedback (DFB) structure is proposed. The device does not require a holding beam. A nonlinear part of the grating is detuned from the remaining part of the grating and has negative nonlinear coefficient. Optical gain is provided by an injected electrical current into an active layer. In the OFF state, due to the detuned section, no laser light is generated in the device. An injected optical pulse reduces the detuning of the nonlinear section, and the optical feedback provided by the DFB structure generates a laser light in the structure that sustains the change in the detuned section. The device is switched “OFF” by detuning another section of the grating by a Reset pulse. The Reset pulse reduces the refractive index of that section by the generation of electron-hole pairs. The Reset pulse wavelength is adjusted such that the optical gain provided by the active layer at that wavelength is zero. The Reset pulse is prevented from reaching the nonlinear detuned section by introducing an optical absorber in the laser cavity to attenuate the pulse. The device is simulated in time domain using General Purpose Graphics Processing Unit (GPGPU) computing. Set-Reset operations are in nanosecond time scale.

Thermodynamic Calculation for Silicon ModifiedAISI M2 High Speed Tool Steel

During high speed tool steel production up to 0.2 wt % silicon is added, primarily to react with oxygen e.g. silicon acts as a de-oxidizer. If more than 0.2 wt % silicon is added, it serves to improve the deep hardening properties. An addition up to ~1 wt % silicon provides hardness and improves temper-stability but reduces the ductility. At high concentration, silicon causes embrittlement. Alloying with silicon raises the solubility of carbon in the matrix and hence the as-quenched hardness. It has virtually no influence on the carbide distribution, but it promotes the formation of M6C type carbides. The many essential alloy additions to iron (C, W, Mo, V, Cr, Si) make the high speed tool steel, HSS a complex multi-component system. Its complete experimental investigation would require enormous time and effort. Instead, the CALPHAD method has been successfully used for computation of phase equilibrium the multi-component HSS system. In the present work, the Thermo-Calc program has been applied to the system Fe-C-Cr-W-Mo-V-Si with the thermodynamic information contained in the solid-solution-database of the TCFE. In the present work, some temperature-concentration diagrams for silicon modified AISI M2 steel are presented by calculated quantities (melting and transformation temperatures, amount and compositions of phases). Calculated data are compared with standard AISI M2 high speed tool steel.

Recent Trends in Producing Ultrafine Grained Steels

Ultrafine grained steels with grain sizes below about 1 μm offer the prospect of high strength and high toughness with traditional steel compositions. These materials are currently the subject of extensive research efforts worldwide. Alloy design is one of the first considered issues, while designing new steel with targeted mechanical properties. However, the alloying content of steel does not fully determine the mechanical properties, but manufacturing procedure, hot rolling and cooling parameters, heat treatment parameters etc. are also of vital importance. For instance, same steel with different processing conditions can exhibit rather large variations in properties. To be precise, chemical composition with the processing parameters determines the microstructure, which in turn determines the properties of the steel. Steel is defined as an iron alloy containing C, Mn and Si that are generally used as alloying elements in steel. Micro-alloying elements such as Nb, Ti V, and B, are considered to be effective, causing strengthening as well as microstructural refinement in small quantities below 0.1 wt% (therefore these are called micro-alloy elements) and are quite generally used in ultrafine grain steel. Substitution alloying elements, such as Mo, Ni, Cr and Cu are alloyed to suppress phase transformation temperatures, i.e. for reaching certain level of strengthening, since the strength of steel structures strongly depends on the phase transformation temperature. Accordingly, the alloy design of ultrafine grains steels with different structures generally relies on: i) carbon levels, ii) sufficient alloying to obtain the desired transformation temperature and iii) micro-alloying technology in conjunction with Thermo Mechanical Controlled Processes (TMCP). Malignant melanoma is characterized by both genetic and molecular alterations that activate phosphoinositide 3-kinase(PI3K),and RAS/BRAF *** this work,through diversity-based high-throughput virtual screening we identified a lead molecule that selectively targets PI3K and BRAF^(V600E) *** screening,Molecular dynamics simulation and MMPBSA calculations were ***3K and BRAF^(V600E) kinase inhibition was done.A375 and G-361 cells were used for in vitro cellular analysis to determine antiproliferative effects,annexin V binding,nuclear fragmentation and cell cycle *** screening of small molecules indicates compound CB-006-3 selectively targets PI3KCG(gamma subunit),PI3KCD(delta subunit)and BRAF^(V600E).Molecular dynamics simulation and MMPBSA bases binding free energy calculations predict a stable binding of CB-006-3 to the active sites of PI3K and BRAF^(V600E).The compound effectively inhibited PI3KCG,PI3KCD and BRAF^(V600E)kinases with respective IC50 values of 75.80,160.10 and 70.84 ***-006-3 controlled the proliferation of A375 and G-361 cells with GI50 values of 223.3 and 143.6 nM,respectively.A dose dependent increase in apoptotic cell population and sub G0/G1 phase of cell cycle were also observed with the compound treatment in addition to observed nuclear fragmentation in these ***,CB-006-3 inhibited BRAF^(V600E),PI3KCD and PI3KCG in both melanoma ***,based on the computational modeling and in vitro validations,we propose CB-006-3 as a lead candidate for selectively targeting PI3K and mutant BRAF^(V600E) to inhibit melanoma cell *** experimental validations,including pharmacokinetic evaluations in mouse models will identify the druggability of the proposed lead candidate for further development as a therapeutic agent for treating melanoma.

Evaluation of infertile women: Mini-review

Evaluation of infertility indicated for women failed to conceive after one year of unprotected intercourse and for women over 35 years failed to conceive after 6 mo of unprotected intercourse, because the fertility decline as women approach 40 years. Evaluation of infertile couple should begin with semen analysis of male partner. If the semen analysis is within normal range, evaluation then move on to female partner. Evaluation of the infertile woman should be carried in cost-effective manner to identify the causes of infertility using the least invasive methods. Methods of evaluation of infertile women include: 1) history and physical examination;2) evaluation of the male partner;3) documentation of ovulation;4) evaluation of the ovarian reserve;5) evaluation of cervical causes of infertility;6) evaluation of uterine causes of infertility;7) documentation of the tubal patency;and 8) exclusion of peritoneal causes of infertility. Evaluation of infertile couple should include evaluation of both partners. Semen analysis for the male partner, followed by documentation of ovulation and evaluation of genital tract patency. PCT is not used as routine evaluation of infertile women. Laparoscopy is not used as routine evaluation of infertile women unless there is suspected peritoneal factors of infertility or endometriosis or tubal occlusion. Ovarian reserve should only be done for infertile women with diminished response to external gonadotropins (not routine).

INRA82 extender enhances semen quality in ram under cooled and cryopreserved stages

Objective:To investigate the effect of INRA82 extender on ram semen quality preserved in cooled and frozen conditions in comparison with Tris-based extenders and to select the suitable extender for ram semen ***:Semen samples were collected and pooled from Awassi rams(n=5)and divided into three *** aliquot was diluted with one of the following extenders:(1)Tris-citrate glucose,(2)Triscitrate fructose,and (3)*** liquid storage,diluted samples were kept at 5℃ for 72 *** motility was measured at 0,24,48 and 72 h after ***,viability,morphology,membrane,acrosome and DNA integrities were evaluated at 24 h *** cryopreservation,diluted samples were cooled,equilibrated,loaded and frozen in liquid *** samples were evaluated in the same manner as cooled ***:Seminal characteristics were improved after cooling and thawing in samples diluted with INRA-82 compared to those diluted with Tris-citrate glucose or Tris-citrate fructose(P<0.05).Conclusions:Dilution of ram semen with INRA-82 improves semen ***,INRA-82,as a stimulating diluent,can be successfully used to reserve viability and sustainability of cooled and cryopreserved ram semen.

Impact of topical nonsteroidal anti-inflammatory drugs in prevention of macular edema following cataract surgery in diabetic patients

AIM： To evaluate the efficacy of prophylactic administration of topical non-steroidal anti-inflammatory drugs（NSAIDs） on macular edema following cataract surgery in diabetic patients, and to compare between types of NSAIDs（ketorolac tromethamine 0.4% and nepafenac 0.1%）. METHODS： Group 1（control） received artificial tears substitute as a placebo group, group 2（nepafenac） received topical nepafenac 0.1%, and group 3（ketorolac） received topical ketorolac tromethamine 0.4%. Patients were examined postoperatively after completing one week, one month, two months and three months＇ intervals for evaluating cystoid macular edema（CME） development. The main study outcomes were achieving the best corrected visual acuity（BCVA） and change in the central macular thickness（CMT） measured with optical coherence topography（OCT）.RESULTS： Eighty eyes of 76 patients were included in this study. BCVA showed a statistically significant difference at the third month postoperative follow up between the control group and the NSAIDs groups（P=0.04）. There was an increase in the CMT in all cases starting from postoperative first week until third month. CMT showed a statistically significant difference between control group and NSAIDs groups from postoperative first month until third month（P=0.008, 0.027, 0.004）. There was no statistically significant difference between nepafenac and ketorolac groups in BCVA and OCT CMT. CONCLUSION： Prophylactic preoperative and postoperative NSAIDs may have a role in reducing the frequency and severity of CME in diabetic eyes following cataract surgery.

Objective:To investigate the role of zinc in reducing the deleterious effects of cadmium on male ***:Rats were injected subcutaneously with CdCl_2 and ZnCl_2 at dose level of2.2 mg/kg(1/40 of LD50 of cadmium per day).Results:The rats treated with cadmium exhibited a significant increase in levels of testicular malondialdehyde,tumor necrosis factor-alpha,nitrogen oxide and inducible nitrogen oxide synthase immunostaining reaction,as well as an elevation of blood hydroperoxide and follicle stimulating *** addition,a significant decrease in testicular ascorbic acid,zinc,reduced glutathione,catalase,superoxide dismutase,sex organ weight,plasma testosterone and luteinizing hormone were observed in the cadmium *** motility and count were decreased with cadmium treatment,while sperm abnormalities elevated *** treatment was found to mitigate the toxic effects of cadmium on oxidative stress,spermatogenesis,sex hormones,and inflammatory *** injected with cadmium showed intense histopathological *** manifested protective role and markedly reduced tissues damage induced by ***:The protective effect of zinc can be attributed to its antioxidant and antiinflammatory properties.
