DICER-like Proteins and Their Role in Plant-herbivore Interactions in Nicotiana attenuata

DICER-like （DCL） proteins produce small RNAs that silence genes involved in development and defenses against viruses and pathogens. Which DCLs participate in plant-herbivore interactions remains unstudied. We identified and stably silenced four distinct DCL genes by RNAi in Nicotiana attenuata （Torrey ex. Watson）, a model for the study of plant-herbivore interactions. Silencing DCL1 expression was lethal. Manduca sexta larvae performed significantly better on ir-dcl3 and ir-dcl4 plants, but not on ir-dcl2 plants compared to wild type plants. Phytohormones, defense metabolites and microarray analyses revealed that when DCL3 and DCL4 were silenced separately, herbivore resistance traits were regulated in distinctly different ways. Crossing of the lines revealed complex interactions in the patterns of regulation. Single ir-dcl4 and double ir-dcl2 ir-dcl3 plants were impaired in JA accumulation, while JA- Ile was increased in ir-dcl3 plants. Ir-dcl3 and ir-dcl4 plants were impaired in nicotine accumulation; silencing DCL2 in combination with either DCL3 or DCL4 restored nicotine levels to those of WT. Trypsin proteinase inhibitor activity and transcripts were only silenced in ir-dcl3 plants. We conclude that DCL2/3/4 interact in a complex manner to regulate anti-herbivore defenses and that these interactions significantly complicate the already challenging task of understanding smRNA function in the regulation of biotic interactions.

Effect of water deficiency on relationships between metabolism,physiology,biomass,and yield of upland cotton(Gossypium hirsutum L.)

Drought is a common abiotic stress that considerably limits crop production. The objective of this study is to explore the influence of water deficiency on the yield, physiologic and metabolomic attributes in upland cotton cultivars（Gossypium hirsutum L）. Cotton cultivars, ＇Ishonch＇ and ＇Tashkent-6＇ were selected to study the relationships among their physiologic, metabolomic and yield attributes during water deficiency. Deficit irrigation was designed by modifying the traditional watering protocol to reduce water use. Results indicate that cotton cultivars respond differently to water deficit stress. Water deficit significantly influenced plant height, the number of internodes, and sympodial branches in both cultivars. However, yield components such as the number of bolls, boll seed, lint mass, and individual plant yield were significantly reduced only in ＇Tashkent-6＇. The leaf area decreased and the specific leaf weight increased in ＇Ishonch＇ under deficit irrigation conditions. However, ＇Tashkent-6＇ demonstrated significant water loss compared to ＇Ishonch＇, and both cultivars showed reduced transpiration rates. Untargeted metabolite profiles of leaves showed clear separation in ＇Ishonch＇, but not in ＇Tashkent-6＇ under deficit irrigation compared to full irrigation. The individual metabolites such as proline and galactinol showed strong association with yield under water deficit stress. Moreover, this study indicates that leaf area and transpiration intensity influence yield during water deficiency. In summary, the correlation among morpho-physiologic, metabolic, and yield components significantly varied between the two cultivars under water deficiency. The flowering stage was sensitive to water stress for both cultivars. The direct relationship between physiology, metabolism, and yield may be a useful selection criterion for determining candidate parents for cotton drought tolerance breeding.

Multivariate Cluster and Principle Component Analyses of Selected Yield Traits in Uzbek Bread Wheat Cultivars

Investigation of genetic diversity of geographically distant wheat genotypes is a useful approach in wheat breeding providing efficient crop varieties. This article presents multivariate cluster and principal component analyses (PCA) of some yield traits of wheat, such as thousand-kernel weight (TKW), grain number, grain yield and plant height. Based on the results, an evaluation of economically valuable attributes by eigenvalues made it possible to determine the components that significantly contribute to the yield of common wheat genotypes. Twenty-five genotypes were grouped into four clusters on the basis of average linkage. The PCA showed four principal components (PC) with eigenvalues > 1, explaining approximately 90.8% of the total variability. According to PC analysis, the variance in the eigenvalues was the greatest (4.33) for PC-1, PC-2 (1.86) and PC-3 (1.01). The cluster analysis revealed the classification of 25 accessions into four diverse groups. Averages, standard deviations and variances for clusters based on morpho-physiological traits showed that the maximum average values for grain yield (742.2), biomass (1756.7), grains square meter (18,373.7), and grains per spike (45.3) were higher in cluster C compared to other clusters. Cluster D exhibited the maximum thousand-kernel weight (TKW) (46.6).

Water scarcity is a major limiting factor for oasis-desert agricultural production,for which cotton is the most economically important *** drought tolerance and minimizing drought-related crop losses so that crop production during a drought can be maintained are critical targets for cotton production in a desert-oasis *** approach could be a way for cotton improvement for oasis-desert *** have focused on an aldehyde dehydrogenase gene,ScALDH21,from the desiccation tolerant moss Syntrichia caninervis as a target gene for cotton drought tolerance *** of ScALDH21 in cotton was field-tested under six different irrigation treatments:root zone model simulated forecast irrigation(FFI) and deficit irrigation(FDI),soil moisture sensor-based irrigation(SFI) and deficit irrigation(SDI),and flood irrigation based evapotranspiration estimate irrigation(EFI) and deficit irrigation(EDI) to evaluate growth performance and *** results demonstrated that ScALDH21-transgenic cotton had increased plant height and leaf area in all *** transgenic lines exhibited slight increased chlorophyll contents,and net photosynthesis rate,and increased water use in nearly all *** transgenic cotton lines tested had increased yields and superior fiber quality than non-transgenic(NT)plants regardless of irrigation *** results demonstrate that ScALDH21-transgenic cotton lines are more drought tolerant than NT plants and exhibit higher yields and fiber quality under water deficit *** study also provides guidelines for optimal irrigation modes and minimum water requirements for the use of the ScALDH21-transgenic cotton lines for improved cotton yields and quality in arid zones.
