Impact of tertiary Gleason pattern 5 on prostate cancer aggressiveness:Lessons from a contemporary single institution radical prostatectomy series

Objective:To better evaluate tertiary Gleason pattern reporting and to evaluate the impact of tertiary Gleason pattern 5(TP5)on prostate cancer pathological features and biochemical recurrence at our large single ***:We retrospectively reviewed 1962 patients who underwent radical prostatectomy(RP)for prostate cancer;TP5 was reported in 159 cases(8.1%).men with Gleason score(GS)7 and GS 8 disease were divided into subgroups with and without TP5,and histopathological features were *** analyses were conducted to assess the impact on TP5 on biochemical-free survival(BFS).Results:Tumors possessing GS 3+4 with TP5 were more likely to exhibit extraprostatic extension(EPE)and had a larger tumor diameter(TD)than GS 3+4 *** 3+4 with TP5 was also associated with positive surgical margins(Sm),seminal vesicle involvement(SVI),and higher pre-operative prostate-specific antigen(PSA)values,but without statistical *** 4+3 with TP5 more commonly presented with EPE,positive Sm,SVI,and greater TD and pre-operative PSA level than GS 4+3 *** multivariate analysis,Gleason score,EPE,and TP5 were overall independent risk factors for PSA recurrence in this ***,GS 4+3 with TP5 was associated with shorter time to recurrence versus GS 4+3 ***:Our results emphasize the importance of TP5 and suggest that criteria for tertiary pattern reporting in prostate cancer should be *** studies are needed to evaluate the role of tertiary patterns in prognostic models.

molecular Docking Studies on Streptomycin Antileishmanial Activity

Resistance to pentavalent antimonial drugs and the lack of vaccines make it urgent to find novel therapeutic options to treat Leishmaniasis, a tropical disease caused by the Leishmania protozoan parasite. The study reported here is to investigate if Streptomycin, an aminoglycoside, and Amphotericin B, the second-line treatment drug, exhibit antileishmanial activity through a similar mechanism. By using mOE (molecular Operating Environment), we performed molecular docking studies on these drugs binding to a range of targets including ribosome targets in Leishmania and H. sapiens. Our study shows that the two drugs do not bind to the same pockets in Leishmania targets but to the same pockets in the human ribosome, with some differences in interactions. moreover, our 2D maps indicated that Amphotericin B binds to the A-site in the human cytoplasmic ribosome, whereas streptomycin does not.

Impact on Soil Organic C and Total Soil N from Cool- and Warm-Season Legumes Used in a Green manure-Forage Cropping System

Annual forage legumes are important components of livestock production systems in East Texas and the southeastern US. Forage legumes contribute nitrogen (N) to cropping systems through biological N fixation, and their seasonal biomass production can be managed to complement forage grasses. Our research objectives were to evaluate both warm- and cool-season annual forage legumes as green manure for biomass, N content, ability to enhance soil organic carbon (SOC) and soil N, and impact on post season forage grass crops. Nine warm-season forage legumes (WSL) were spring planted and incorporated as green manure in the fall. Forage rye (Secale cereale L.) was planted following the incorporation of WSL treatments. Eight cool-season forage legumes (CSL) were fall planted in previously fallow plots and incorporated as green manure in late spring. Sorghum-sudangrass (Sorghum bicolor x Sorghum bicolor var. sudanense) was planted over all treatments in early summer after forage rye harvest and incorporation of CSL treatments. Sorghum-sudangrass was harvested in June, August and September, and treatments were evaluated for dry matter and N concentration. Soil cores were taken from each plot, split into depths of 0 to 15, 15 to 30 and 30 to 60 cm, and soil C and N were measured using combustion analysis. Nylon mesh bags containing plant samples were buried at 15 cm and used to evaluate decomposition rate of above ground legume biomass, including change in C and N concentrations. mungbean (Vigna radiata L. [Wilczek]) had the highest shoot biomass yield (6.24 t Dm ha-1) and contributed the most total N (167 kg∙ha-1) and total C (3043 kg∙ha-1) of the WSL tested. Decomposition rate of WSL biomass was rapid in the first 10 weeks and very slow afterward. Legume Green manure and Intercropping for High Biomass Sorghum Production

Before the advent of cheap, synthetic fertilizers, legumes were commonly used as green manure crops for their ability to fix atmospheric nitrogen (N). A three-year study at Overton, TX examined legume integration into high-biomass sorghum (Sorghum bicolor L.) production systems on a Lilbert loamy fine sand recently cultivated after a fertilized bermudagrass [Cynodon dactylon (L.) Pers.] pasture. In this split-split plot design, ‘Dixie’ crimson clover (Trifolium incarnatum L.) and ‘Iron and Clay’ cowpea (Vigna unguiculata L.) were integrated into a high-biomass sorghum production system to evaluate impacts on N concentration, C concentration, and yield of high-biomass sorghum and their impacts on soil total N and soil organic carbon (SOC). main plots were split into crimson clover green manure (CLGm) and winter fallow (FALL) followed by three sub-plots split into warm-season crop rotations: cowpea green manure (CPGm), cowpea-sorghum intercrop (CPSR), and sorghum monocrop (SORG). Three N fertilizer treatments (0, 45, 90 kg N∙ha−1) were randomized and applied as sub-sub plots. The CLGm increased (P sorghum biomass yield (16.5 t Dm∙ha−1) 28% in year three but had no effect in the first two years. The CPSR treatment reduced sorghum yield up to 62% compared to SORG;whereas CPGm increased sorghum yield 56% and 18% the two years following cowpea incorporation. Rate of N fertilizer had no effect on sorghum biomass yield. Decrease in SOC and soil N over time indicated mineralization of organic N and may explain why no N fertilizer response was observed in sorghum biomass yield. Cowpea showed strong potential as a green manure crop but proved to be too competitive for successful intercropping in high-biomass sorghum production systems.

青藏高原及其周缘褶皱-冲断带变形方式和时限是验证高原形成众多大陆岩石圈变形动力学端员模型的关键。近年发展起来的断层泥自生伊利石K-Ar定年技术为精确限定褶皱-冲断带变形时限提供了有效手段。鲜水河—安宁河—小江断裂系作为伴随印度-欧亚板块碰撞造山和高原物质侧向挤出过程形成的大型左旋走滑断裂系,其变形过程可为解译印度-欧亚板块碰撞远程应力向东传递方式提供关键证据。本文选取安宁河断裂冕宁—西昌段作为研究对象,在详细构造解析以明确其构造运动学特征的基础上,开展了断层泥自生伊利石K-Ar定年研究,以期精确限定安宁河断裂脆性变形的时间。构造解析表明安宁河断裂在近EW向挤压作用下经历了冲断变形,断层带发育碎斑岩和断层泥,指示为脆性变形。安宁河断裂不同粒级断层泥样品的黏土矿物学和伊利石K-Ar定年分析揭示,随着样品粒级减小,高温2m1型伊利石含量相对低温1m/1md型伊利石含量逐渐减少,其K-Ar年龄总体呈变年轻趋势,表明不同粒级伊利石K-Ar年龄是由碎屑2m1型和自生1m/1md型两个多型端员组成的混合年龄。伊利石年龄分析获得自生1m/1md型伊利石的年龄为(42.6±9.4)ma,表明安宁河断裂经历了始新世中期的冲断变形。结合前人构造变形、沉积学、低温热年代学和古地磁等研究,我们认为始新世中期青藏高原腹地及其周缘褶皱-冲断带发生准同期的构造挤压变形,其动力学机制可能与印度-欧亚板块硬碰撞以及青藏高原地块的陆内俯冲所导致的先存构造带活化有关。始新世中期安宁河断裂发生冲断变形直接证明印度-欧亚板块碰撞初期的远程应力已传递至青藏高原东南缘地区。

细石叶技术产品是青藏高原发现最丰富的史前人类文化遗物,代表了早期人类在高原的大规模迁徙.然而,经过系统发掘、具有可靠年代数据的细石叶技术遗址在青藏高原发现较少,高原腹地更为缺乏,这对理解细石叶技术人群在高原大规模迁徙扩散的时间和过程带来了诸多障碍.本文研究报道了青藏高原腹地海拔4600m处新发现的细石叶技术遗址——尼阿底遗址第3地点.技术-类型学分析结果显示,尼阿底遗址第3地点细石叶的生产主要包括楔形石核和半锥形石核两种技术,与旧石器时代晚期中国华北地区的细石叶技术特征接近.光释光测年(OSL)和碳十四测年(^(14)C)的结果表明,尼阿底遗址第3地点的年代为11000~10000a BP.尼阿底遗址第3地点作为目前青藏高原腹地发现的最早的细石叶技术遗址,为了解拥有细石叶技术的狩猎采集者到达高原腹地的时间及其可能的扩散路线提供了重要信息.另外,通过对整个高原细石叶技术发展的分析,结合分子生物学的成果对比显示,细石叶技术人群迁移进入高原可能与人类对高原的长期占领以及现代藏族人的形成有密切联系.

The development of fully solution-processed,biodegradable piezoelectrics is a critical step in the development of green electronics towards the worldwide reduction of harmful electronic ***,recent printing processes for piezoelectrics are hindered by the high sintering temperatures required for conventional perovskite fabrication ***,a process was developed to manufacture lead-free printed piezoelectric devices at low temperatures to enable integration with eco-friendly substrates and electrodes.A printable ink was developed for screen printing potassium niobate(KNbO3)piezoelectric layers in microns of thickness at a maximum processing temperature of 120℃ with high *** parallel plate capacitor and cantilever devices were designed and manufactured to assess the quality of this ink and evaluate its physical,dielectric,and piezoelectric characteristics;including a comparison of behaviour between conventional silicon and biodegradable paper *** printed layers were 10.7–11.2μm thick,with acceptable surface roughness values in the range of 0.4-1.1μ*** relative permittivity of the piezoelectric layer was *** poling parameters were optimised for the piezoelectric response,with an average longitudinal piezoelectric coefficient for samples printed on paper substrates measured as d33,eff,paper=13.57±2.84 pC/N;the largest measured value was 18.37 pC/N on paper *** approach to printable biodegradable piezoelectrics opens the way forward for fully solution-processed green piezoelectric devices.

Cost of treatment support for multidrug-resistant tuberculosis using patient-centred approaches in Ethiopia: a model-based method

Background Patient and health system costs for treating multidrug-resistant tuberculosis (mDR-TB) remain high even after treatment duration was shortened. many patients do not finish treatment, contributing to increased transmission and antimicrobial resistance. A restructure of health services, that is more patient-centred has the potential to reduce costs and increase trust and patient satisfaction. The aim of the study is to investigate how costs would change in the delivery of mDR-TB care in Ethiopia under patient-centred and hybrid approaches compared to the current *** We used published data, collected from 2017 to 2020 as part of the Standard Treatment Regimen of Anti-Tuberculosis Drugs for Patients with mDR-TB (STREAm) trial, to populate a discrete event simulation (DES) model. The model was developed to represent the key characteristics of patients’ clinical pathways following each of the three treatment delivery strategies. To the pathways of 1000 patients generated by the DES model we applied relevant patient cost data derived from the STREAm trial. Costs are calculated for treating patients using a 9-month mDR-TB treatment and are presented in 2021 United States dollars (USD).Results The patient-centred and hybrid strategies are less costly than the standard-of-care, from both a health system (by USD 219 for patient-centred and USD 276 for the hybrid strategy) and patient perspective when patients do not have a guardian (by USD 389 for patient-centred and USD 152 for the hybrid strategy). Changes in indirect costs, staff costs, transport costs, inpatient stay costs or changes in directly-observed-treatment frequency or hospitalisation duration for standard-of-care did not change our *** Our findings show that patient-centred and hybrid strategies for delivering mDR-TB treatment cost less than standard-of-care and provide critical evidence that there is scope for such strategies to be implemented in routine care.
