Expression of an endoglucanase from Trib.lium castaneum （TcEG 1 ） in Saccharomyces cerevisiae

Insects are a largely unexploited resource in prospecting for novel cellulolytic enzymes to improve the production of ethanol fuel from lignocellulosic b.omass. The cost of lignocellulosic ethanol production is expected to decrease b. the comb.nation of cellulose degradation （saccharification） and fermentation of the resulting glucose to ethanol in a single process, catalyzed b. the yeast Saccharomyces cerevisiae transformed to express efficient cellulases. While S. cerevisiae is an estab.ished heterologous expression system, there are no availab.e data on the functional expression of insect cellulolytic enzymes for this species. To address this knowledge gap, S. cerevisiae was transformed to express the full-length cDNA encoding an endoglucanase from the red flour b.etle, Trib.lium castaneum （TcEG 1）, and evaluated the activity of the transgenic product （rTcEG 1）. Expression of the TcEG1 cDNA in S. cerevisiae was under control of the strong glyceraldehyde-3 phosphate dehydrogenase promoter. Cultured transformed yeast secreted rTcEG1 protein as a functional β-1,4-endoglucanase, which allowed transformants to survive on selective media containing cellulose as the only availab.e carb.n source. Evaluation of sub.trate specificity for secreted rTcEG1 demonstrated endoglucanase activity, although some activity was also detected against complex cellulose sub.trates. Potentially relevant to uses in b.ofuel production rTcEG1 activity increased with pH conditions, with the highest activity detected at pH 12. Our results demonstrate the potential for functional production of an insect cellulase in S. cerevisiae and confirm the stab.lity of rTcEG1 activity in strong alkaline environments.

Molecular Docking Studies on Streptomycin Antileishmanial Activity

Resistance to pentavalent antimonial drugs and the lack of vaccines make it urgent to find novel therapeutic options to treat Leishmaniasis, a tropical disease caused b. the Leishmania protozoan parasite. The study reported here is to investigate if Streptomycin, an aminoglycoside, and Amphotericin b. the second-line treatment drug, exhib.t antileishmanial activity through a similar mechanism. b. using MOE (Molecular Operating Environment), we performed molecular docking studies on these drugs b.nding to a range of targets including rib.some targets in Leishmania and H. sapiens. Our study shows that the two drugs do not b.nd to the same pockets in Leishmania targets b.t to the same pockets in the human rib.some, with some differences in interactions. Moreover, our 2D maps indicated that Amphotericin b.b.nds to the A-site in the human cytoplasmic rib.some, whereas streptomycin does not.

b.nzodiazepines and other b.nzodiazepine receptor agonists, such as the “Z” drugs, are widely prescrib.d medications mainly used for treating anxiety and seizures, and for inducing sedation. Unfortunately, despite their popularity, b.nzodiazepine prescrib.ng often exceeds recommendations and the consequences can b. severe. On Septemb.r 23, 2020, the United States FDA announced a new requirement for a b.xed Warning for b.nzodiazepines prescrib.ng. Along with this announcement, the FDA stated that relevant information regarding the initiation, continuation, and discontinuation of b.nzodiazepines is lacking. Here, we describ. initial pilot studies intended to investigate the questions 1) can animal models b. developed that demonstrate b.nzodiazepine physical dependence and/or withdrawal symptoms, and 2) determine whether translocator protein (TSPO) plays a role in b.nzodiazepine dependence and/or withdrawal processes. The former was demonstrated, methodological limitations prevented the latter.

Mesenchymal Stem Cell Allograft Improved Pain Management in Dogs with Osteoarthritis

b.ckground: Osteoarthritis is one of the most common b.ne diseases, triggered b. b.ne destruction stemming from the inflammatory response of chondrocytes. The disease progresses slowly, b.t halting its progression or finding a cure remains elusive. The treatment of pain associated with osteoarthritis has yielded unsatisfactory results. In recent years, mesenchymal stem cells (MSCs) have emerged as a potential avenue for addressing the condition. In this study, we used MSCs to treat companion dogs with osteoarthritis. Methods: For this study, 26 animals were included in this study to assess the pain and mob.lity one month after treatment. The pain scores were ob.ained from owners using a questionnaire b.sed on the Helsinki Chronic Pain Index, and the Liverpool Osteoarthritis in Dogs (LOAD) Owner questionnaire to assess the mob.lity of the dogs from stem cell infusion. Results: Questionnaires were administered to dog owners b.fore and one month after treatment, and we found that dogs treated with MSCS experienced an 81.2% ± 6.8% reduction in pain and a 77.9% ± 10.1% increase in mob.lity, whereas most of the dogs in the untreated control group experienced disease progression. Conclusions: The transplantation of stem cells into companion pets is a promising and expanding opportunity for pet owners with aging and arthritic dogs. MSCs may play an important role in the treatment of OA without complications in companion pets.
