Versatile multiplexed super-resolution imaging of nanostructures by Quencher-Exchange-PAINT

The optical super-resolution technique DNA-PAINT (Point Accumulation Imaging in Nanoscale Topography) provides a flexible way to achieve imaging of nanoscale structures at ∼10-nanometer resolution. In DNA-PAINT, fluorescently labeled DNA “imager” strands bind transiently and with high specificity to complementary target “docking” strands anchored to the structure of interest. The localization of single binding events enables the assembly of a super-resolution image, and this approach effectively circumvents photobleaching. The solution exchange of imager strands is the basis of Exchange-PAINT, which enables multiplexed imaging that avoids chromatic aberrations. Fluid exchange during imaging typically requires specialized chambers or washes, which can disturb the sample. Additionally, diffusional washout of imager strands is slow in thick samples such as biological tissue slices. Here, we introduce Quencher-Exchange-PAINT—a new approach to Exchange-PAINT in regular open-top imaging chambers—which overcomes the comparatively slow imager strand switching via diffusional imager washout. Quencher-Exchange-PAINT uses “quencher” strands, i.e., oligonucleotides that prevent the imager from binding to the targets, to rapidly reduce unwanted single-stranded imager concentrations to negligible levels, decoupled from the absolute imager concentration. The quencher strands contain an effective dye quencher that reduces the fluorescence of quenched imager strands to negligible levels. We characterized Quencher-Exchange-PAINT when applied to synthetic, cellular, and thick tissue samples. Quencher-Exchange-PAINT opens the way for efficient multiplexed imaging of complex nanostructures, e.g., in thick tissues, without the need for washing steps.

A recent report has uncovered a fascinating aspect of how neurons utilize alternative splicing of transcripts of an adhesion G protein-coupled receptor(aGPCR)gene to serve specific functions in the life of a synapse.1 The work provides significant insight into the ability of cells to support distinct biochemical and physiological operations through post-transcriptional processing of genetic information that is encoded in a single aGPCR locus.

Extension of Flow Behaviour and Damage Models for Cast Iron Alloys with Strain Rate Effect

Cast iron alloys with low production cost and quite good mechanical properties are widely used in the automotive *** study the mechanical behavior of a typical ductile cast iron(GJS-450)with nodular graphite,uni-axial quasi-static and dynamic tensile tests at strain rates of 10^(-4),1,10,100,and 250 s^(-1)were carried *** order to investigate the influence of stress state on the deformation and fracture parameters,specimens with various geometries were used in the *** strain curves and fracture strains of the GJS-450 alloy in the strain rate range of 10^(-4)to 250 s^(-1)were obtained.A strain rate-dependent plastic flow model was proposed to describe the mechanical behavior in the corresponding strain-rate *** available damage model was extended to take the strain rate into account and calibrated based on the analysis of local fracture *** with the proposed plastic flow model and the damage model were conducted to observe the deformation and fracture *** results show that the strain rate has obviously nonlinear effects on the yield stress and fracture strain of GJS-450 *** predictions with the proposed plastic flow and damage models at various strain rates agree well with the experimental results,which illustrates that the rate-dependent plastic flow and damage models can be used to describe the mechanical behavior of cast iron alloys at elevated strain *** proposed plastic flow and damage models can be used to describe the deformation and fracture analysis of materials with similar properties.

The Eocene-Oligocene transition(EOT)marked a rapid global cooling event,often considered as the beginning of the modern icehouse *** by various factors,including tectonic activity and paleogeographic settings,the terrestrial records indicate a diverse response of fauna and vegetation to this global *** examined nine macrofossil assemblages from seven fossil localities on the southeastern margin of the Tibetan Plateau and from the mid-latitudinal Europe ranging from the latest Bartonian and Priabonian(37.71-33.9 Ma)to the Rupelian(33.9-27.82 Ma).Our aims were to trace and compare the vegetation history of both regions in the late Eocene and early *** results show that both regions experienced changes in vegetation composition in response to climate change,characterized by a decrease in the percentages of broad-leaved evergreen elements and distinctive changes in general vegetation types.A general change in the overall vegetation type from subtropical broad-leaved evergreen forests in the late Eocene to temperate broad-leaved mixed deciduous evergreen forests,or mixed mesophytic forests,in the early Oligocene is recognized in both *** results indicate a clear change in leaf architecture,leaf margin states,and secondary venation types in the mid-latitudinal Europe,while the results from the southeastern margin of the Tibetan Plateau show a distinct reduction in leaf *** data suggest that both global and regional factors played key roles in shaping the vegetation in the two regions.

Exploring the interplay between climate change and schistosomiasis transmission dynamics

Schistosomiasis, a neglected tropical disease caused by parasitic worms, poses a major public health challenge in economically disadvantaged regions, especially in Sub-Saharan Africa. Climate factors, such as temperature and rainfall patterns, play a crucial role in the transmission dynamics of the disease. This study presents a deterministic model that aims to evaluate the temporal and seasonal transmission dynamics of schistosomiasis by examining the influence of temperature and rainfall over time. Equilibrium states are examined to ascertain their existence and stability employing the center manifold theory, while the basic reproduction number is calculated using the next-generation technique. To validate the model's applicability, demographic and climatological data from Uganda, Kenya, and Tanzania, which are endemic East African countries situated in the tropical region, are utilized as a case study region. The findings of this study provide evidence that the transmission of schistosomiasis in human populations is significantly influenced by seasonal and monthly variations, with incidence rates varying across countries depending on the frequency of temperature and rainfall. Consequently, the region is marked by both schistosomiasis emergencies and re-emergences. Specifically, it is observed that monthly mean temperatures within the range of 22–27 °C create favorable conditions for the development of schistosomiasis and have a positive impact on the reproduction numbers. On the other hand, monthly maximum temperatures ranging from 27 to 33 °C have an adverse effect on transmission. Furthermore, through sensitivity analysis, it is projected that by the year 2050, factors such as the recruitment rate of snails, the presence of parasite egg-containing stools, and the rate of miracidia shedding per parasite egg will contribute significantly to the occurrence and control of schistosomiasis infections. Ab initio density response and local field factor of warm dense hydrogen

We present quasi-exact ab initio path integral Monte Carlo(PIMC)results for the partial static density responses and local field factors of hydrogen in the warm dense matter regime,from solid density conditions to the strongly compressed *** full dynamic treatment of electrons and protons on the same footing allows us to rigorously quantify both electronic and ionic exchange–correlation effects in the system,and to compare the results with those of earlier incomplete models such as the archetypal uniform electron gas or electrons in a fixed ion snapshot potential that do not take into account the interplay between the two *** full electronic density response is highly sensitive to electronic localization around the ions,and our results constitute unambiguous predictions for upcoming X-ray Thomson scattering experiments with hydrogen jets and fusion *** PIMC results are made freely available and can be used directly for a gamut of applications,including inertial confinement fusion calculations and the modeling of dense astrophysical ***,they constitute invaluable benchmark data for approximate but computationally less demanding approaches such as density functional theory or PIMC within the fixed-node approximation.

Transfer learning from synthetic data for open-circuit voltage curve reconstruction and state of health estimation of lithium-ion batteries from partial charging segments

Data-driven models for battery state estimation require extensive experimental training data,which may not be available or suitable for specific tasks like open-circuit voltage(OCV)reconstruction and subsequent state of health(SOH)*** study addresses this issue by developing a transfer-learning-based OCV reconstruction model using a temporal convolutional long short-term memory(TCN-LSTM)network trained on synthetic data from an automotive nickel cobalt aluminium oxide(NCA)cell generated through a mechanistic model *** data consists of voltage curves at constant temperature,C-rates between C/30 to 1C,and a SOH-range from 70%to 100%.The model is refined via Bayesian optimization and then applied to four use cases with reduced experimental nickel manganese cobalt oxide(NMC)cell training data for higher use *** TL models’performances are compared with models trained solely on experimental data,focusing on different C-rates and voltage *** results demonstrate that the OCV reconstruction mean absolute error(MAE)within the average battery electric vehicle(BEV)home charging window(30%to 85%state of charge(SOC))is less than 22 mV for the first three use cases across all *** SOH estimated from the reconstructed OCV exhibits an mean absolute percentage error(MAPE)below 2.2%for these *** study further investigates the impact of the source domain on TL by incorporating two additional synthetic datasets,a lithium iron phosphate(LFP)cell and an entirely artificial,non-existing,cell,showing that solely the shifting and scaling of gradient changes in the charging curve suffice to transfer knowledge,even between different cell chemistries.A key limitation with respect to extrapolation capability is identified and evidenced in our fourth use case,where the absence of such comprehensive data hindered the TL process.

Comparison of Air Pollution−Mortality Associations Using Observed Particulate Matter Concentrations and Reanalysis Data in 33 Spanish Cities

We present a novel approach for Stimulated Raman Scattering(SRS)spectroscopy in which a hyper spectral resolution and high-speed spectral acquisition are achieved by employing amplified offset-phase controlled fs-pulse *** investigate the method by solving the coupled non-linear Schrödinger equations and validate it by numerically characterizing SRS in molecular nitrogen as a model *** spectral resolution of the method is found to be determined by the inverse product of the number of pulses in the burst and the intraburst pulse *** SRS spectrum is obtained through a motion-free scanning of the offset phase that results in a sweep of the Raman-shift *** to high spectral resolution and fast motion-free scanning the technique is beneficial for a number SRS-based applications such as gas sensing and chemical analysis.
