A versatile Tn7 transposon-based bioluminescence tagging tool for quantitative and spatial detection of bacteria in plants

Investigation of plant-bacteria interactions requires quantification of in planta bacterial titers by means of cumbersome and time-consuming colony-counting assays.Here,we devised a broadly applicable tool for bioluminescence-based quantitative and spatial detection of bacteria in plants.We developed vectors that enable Tn7 transposon-mediated integration of the luxCDABEluciferase operon into a specific genomic location found ubiquitously across bacterial phyla.These vectors allowed for the generation of bioluminescent transformants of various plant pathogenic bacteria from the genera Pseudomonas,Rhizobium(Agrobacterium),and Ralstonia.Direct luminescence measurements of plant tissues inoculated with bioluminescent Pseudomonas syringae pv.tomato DC3000(Pto-lux)reported bacterial titers as accurately as conventional colony-counting assays in Arabidopsis thaliana,Solanum lycopersicum,nicotiana benthamiana,and Marchantia polymorpha.We further showed the usefulness of our vectors in converting previously generated Pto derivatives to isogenic bioluminescent strains.Importantly,quantitative bioluminescence assays using these Pto-lux strains accurately reported the effects of plant immunity and bacterial effectors on bacterial growth,with a dynamic range of four orders of magnitude.Moreover,macroscopic bioluminescence imaging illuminated the spatial patterns of Pto-lux growth in/on inoculated plant tissues.In conclusion,our vectors offer untapped opportunities to develop bioluminescence-based assays for a variety of plant-bacteria interactions.

The whitefly,Bemisia tabaci,is a major pest of cassava,particularly in Africa where it is responsible both for the transmission of plant viruses and,increasingly,for direct damage due to feeding by high populations.To date,there have been no practical solutions to combat this emerging problem,due to the inability of the subsistence farmers that grow cassava to afford expensive inputs such as insecticides.A programme of research was carried out linking institutes in Africa,the UK and South America,to identify possible resistance sources in cassava to the whitefly,Bemisia tabaci.The South American genotype MEcu 72 and several Ugandan cassava landraces including Ofumba Chai,nabwire 1 and Mercury showed good levels of resistance to B.tabaci.Field and screen-house experiments showed that all of the improved,high-yielding cassava mosaic disease（CMD）resistant cassava genotypes assessed were highly susceptible to B.tabaci and supported high populations of all life stages.These data support the hypothesis that the continuing high populations of cassava B.tabaci in Uganda are due,in part,to the widespread adoption of CMD-resistant cassava varieties during the CMD pandemic.They also show that the whitefly,Aleurotrachelus socialis,resistance present in the South American cassava genotypes could have broader applicability in the Old World.

Age calibrated deformation histories established by detailed mapping and dating of key magmatic time markers are correlated across all tectono-metamorphic provinces in the Damara Orogenic System.Correlations across structural belts result in an internally consistent deformation framework with evidence of stress field rotations with similar timing,and switches between different deformation events.Horizontal principle compressive stress rotated clockwise ~180°in total during Kaoko Belt evolution,and^135° during Damara Belt evolution.At most stages,stress field variation is progressive and can be attributed to events within the Damara Orogenic System,caused by changes in relative trajectories of the interacting Rio De La Plata,Congo,and Kalahari Cratons.Kaokoan orogenesis occurred earliest and evolved from collision and obduction at ~590 Ma,involving E-W directed shortening,progressing through different transpressional states with ~45° rotation of the stress field to strike-slip shear under nW-SE shortening at ~550-530 Ma.Damaran orogenesis evolved from collision at ~555-550 Ma with nW-SE directed shortening in common with the Kaoko Belt,and subsequently evolved through ~90°rotation of the stress field to nE-SW shortening at ~512-508 Ma.Both Kaoko and Damara orogenic fronts were operating at the same time,with all three cratons being coaxially convergent during the 550-530 Ma period;Rio De La Plata directed SE against the Congo Craton margin,and both together over-riding the Kalahari Craton margin also towards the SE.Progressive stress field rotation was punctuated by rapid and significant switches at ~530-525 Ma,~508 Ma and ~505 Ma.These three events included:(1)Culmination of main phase orogenesis in the Damara Belt,coinciding with maximum burial and peak metamorphism at 530-525 Ma.This occurred at the same time as termination of transpression and initiation of transtensional reactivation of shear zones in the Kaoko Belt.Principle compressive stress switched from nW-SE to nnW-SSE shortening in both Kaoko and Damar

For most particle-based applications, formulation in the liquid phase is a decisive step, and thus, particle interactions and stability in liquid media are of major importance. The concept of Hansen solubility parameters (HSP) was initially invented to describe the interactions of (polymer) molecules and their solubility in different liquids and is increasingly being used in particle technology to describe dispersibility. Because dispersions are not thermodynamically stable, the term Hansen dispersibility parameters (HDP) is used instead of HSP (SiiE, Sobisch, Peukert, Lerche,& Segets, 2018). Herein, we extend a previously developed standardized and non-subjective method for determination of Hansen parameters based on analytical centrifugation to the important class of quantum materials. As a technically relevant model system, zinc oxide quantum dots (QDs) were used to transfer our methodology to nanoparticles (nPs) with sizes below lOnm. The results obtained using the standard procedure starting from a dried powder were compared with those obtained through redispersion from the wet sediment produced during the typical washing procedure of QDs, and drying was observed to play an important role. In conclusion, our study reveals the high potential of HDP for quantifying the interfacial properties of nPs as well as their link to dispersibility.

Volatile oil was isolated from the seed of Treculia africana (Moraceae) by solvent extraction and the chemical constituents were identified by means of GC and GC-MS. A total of twelve (12) components were characterized in the volatile oil. Introduction: People in rural communities are exposed to the risk factors for non-communicable disease as those in urban areas. Correct interpretation and management of symptoms are important in primary care especially in rural communities where diagnostic facilities are not readily available. Materials and Methods: This was a cross-sectional study carried out in a village in Enugu South Local Government area of Enugu State. Thorough medical history was taken from all eligible participants who gave their consent. Data were analyzed using SPSS version 26. Results: Data from a total of 1019 (64.7%) out of 1576 participants were analysed. Fever was by far the most common presenting complaint 580 (56.9%) of all participants and 67.9% of medical complaints seen in the clinic. This was followed remotely by musculoskeletal problems and neurological problems 15.6% and 3.8% of the participants respectively. Across all age groups, febrile illness was more than non-febrile illness except in those 65 years. Generalized body pains 74 (7.3%), back pains 55 (5.1), headache 33 (3.2%) and abdominal pains 24 (2.4%) were the commonest symptoms presented by the participants. About 15 (1.5%) of the population surveyed had had trauma/surgery in the past. This was twice the number who presented to the clinic with symptoms related to injury and trauma 7 (0.7%). Conclusions: Fever and musculoskeletal pains are the dominant medical complaints presented by adults in a rural community clinic. This may be related to the season of the year and the occupation of the population studied.

Antibacterial activity of methanol, n-hexane and water extracts of seeds and bark of Moringa oleifera and Moringa ovalifolia was conducted. The causative agents of common bacterial infections that are thought to cause water-borne diseases, namely, Escherichia coli, Enterococcus faecalis, and Bacillus cereus were chosen for the study. The paper-disc diffusion method was used with treatments arranged in a completely randomized design and replicated four times. The M. oleifera extracts showed more inhibitory effect than those of M. ovolifolia. The conventional antibiotic Ampicilin generally showed higher inhibitory effect than the extracts of both M. oleifera and M. ovalifolia. The inhibitory effect varied depending on the solvent used. The n-hexane extracts of both seeds and bark of M. ovalifolia and M. oleifera had almost the same inhibition activities (6 ± 1 mm mean inhibition zones) on B. cereus, E. coli and E. faecalis. The n-hexane extract generally gave lower antibacterial activities than those of the other solvents for seeds and back. The results of the study showed that M. oleifera and M. ovalifolia had a degree of antibacterial properties against the selected test organisms that cause water borne diseases.

印刷废水具有化学组分复杂、油墨悬浮物质含量高等特点。通常采用的沉淀/厌氧/好氧组合处理工艺存在污泥产量大、占地面积大、运行管理复杂等问题。采用An MBR-SBR组合工艺处理印刷废水,设计处理规模为5 m3/d,出水COD均值为(304±30)mg/L,氨氮均值为(45±11)mg/L,优于北京市《水污染物排放标准》(DB 11/307—2005)中"排入城镇污水处理厂的水污染物排放限值"。与传统印刷废水处理工艺相比,该处理工艺提高了出水水质,缩短了工艺流程,简化了运行管理。
