Loss of GATA6-mediated up-regulation of UTX promotes pancreatic tumorigenesis and progression

Ubiquitously transcribed tetratricopeptide repeat on chromosome X(UTX),also known as lysine(K)-specific demethylase 6A(KDM6A),functions as a tumor suppressor gene or oncogene depending on the tumor type and context.However,its tumor-suppressive mechanisms remain largely unknown.Here,we investigated the clinical significance and biological effects of UTX expression in pancreatic ductal adenocarcinoma(PDA)and determined the potential mechanisms of its dysregulation.UTX expression and its association with clinicopathologic characteristics of PDA patients were analyzed using immunohistochemistry.UTX mRNA and protein expression and their regulation in PDA cell lines were measured using quantitative polymerase chain reaction and Western blot analyses.The biological functions of UTX in PDA cell growth,migration,and invasion were determined using gain-and loss-of-function assays with both in vitro and in vivo animal models.UTX expression was reduced in human PDA cell lines and specimens.Low UTX expression was associated with poor differentiation and prognosis in PDA.Forced UTX expression inhibited PDA proliferation,migration,and invasion in vitro and PDA growth and metastasis in vivo,whereas knockdown of UTX expression did the opposite.Mechanistically,UTX expression was trans-activated by GATA6 activation.GATA6-mediated PDA progression could be blocked,at least partially,by silencing UTX expression.In conclusion,loss of GATA6-mediated UTX expression was evident in human PDA and restored UTX expression suppressed PDA growth and metastasis.Thus,UTX is a tumor suppressor in PDA and may serve as a prognostic biomarker and therapeutic target.

Electrocatalytic performance of CNTs/graphene composited rare earth phthalocyanines(M=La,Y,Yb,Sc)

In this study,a class of rare earth composite sandwich phthalocyanines(MPcs,M=La,Y,Yb,Sc) were prepared and compounded with graphene and carbon nanotubes to obtain MPc/Gr and MPc/CNTs composites.The electrocatalytic behaviors of MPc/Gr and MPc/CNTs electrodes were further investigated.The results show that the central rare earth metal has a large influence on the electrocatalytic performance.For the MPcs/Gr samples,ScPc with the smallest ionic radius and molecular size can be more uniformly dispersed in graphene,and the hydrogen precipitation overpotential of ScPc/Gr electrode is514 mV,corresponding to a Tafel slope of 148 mV/dec,with better electrocatalytic performance than other rare earth metal phthalocyanines.As for the MPc/CNTs composites,LaPc,which has the largest ionic radius and molecular size,is more uniformly dispersed on the surface of CNTs,so that the LaPc/CNT electrode exhibits the best LSV performance with the smallest corresponding Tafel slope(176 mV/dec).The main reason is the different binding modes of MPcs molecules in Gr and CNTs.When rare earth phthalocyanine is combined with layered graphene,the smallest size of rare earth phthalocyanine(ScPc)is more easily embedded in the graphene layer,resulting in better homogeneity of the composite,larger effective contact area and better electrocatalytic performance.In contrast,when rare earth phthalocyanine is bound to carbon nanotubes in a tubular structure,it is mainly bound by attaching to the surface or being entangled by the carbon nanotubes.In this case,the rare-earth phthalocyanine molecules(LaPc)with larger layer spacing can provide more contact area with CNTs,forming a more uniform and effective composite,which eventually provides more active sites and better electrocatalytic performance.

Adaptive temporal patterns of echolocation and flight behaviors used to fly through varied-sized windows by 2 species of high duty cycle bats

As actively sensing animals guided by acoustic information, echolocating bats must adapt their vocal–motor behavior to various environmentsand behavioral tasks. Here, we investigated how the temporal patterns of echolocation and flight behavior were adjusted in 2 species of batswith a high duty cycle (HDC) call structure, Rhinolophus ferrumequinum and Hipposideros armiger, when they flew along a straight corridorand then passed through windows of 3 different sizes. We also tested whether divergence existed in the adaptations of the 2 species. Both H.armiger and R. ferrumequinum increased their call rates by shortening the pulse duration and inter-pulse interval for more rapid spatial samplingof the environment when flying through smaller windows. Bats produced more sonar sound groups (SSGs) while maintaining a stable proportion of calls that made up SSGs during approaches to smaller windows. The 2 species showed divergent adjustment in flight behavior across3 different window sizes. Hipposideros armiger reduced its flight speed to pass through smaller windows while R. ferrumequinum increasedits flight speed. Our results suggest that these 2 species of HDC bats adopt similar acoustic timing patterns for different tasks although theyperformed different flight behaviors.

Comparative Study of Linear Variable Flux Reluctance Machine with Linear Wound Field Flux Reversal Machine

As members of doubly salient magnetless linear machines,linear variable flux reluctance(LVFR)and wound field flux reversal(LWFFR)machines inherit the merits of conventional magnetless linear machines such as low cost,high flux adjustment capability and high reliability.Furthermore,like linear switched reluctance machine,they have a very simple and compact long secondary,which are very attractive for long stroke applications.However,low force capability is their major defect.To solve this issue,new LVFR and LWFFR machine topologies were proposed in recent work,while lacking studies on their force improvement mechanism and further force evaluation.In this paper,LVFR and LWFFR machines with improved force performance are comparatively studied with the emphasis on their force capabilities.The operation principle of the two machines is analyzed based on magnetic field harmonics produced by flux modulation.Contributions of air-gap flux density harmonic components to no-load back electromagnetic forces of the two machines are analyzed and the average force equation is derived.Moreover,force capabilities of the both machines are investigated by means of the time-stepping finite-element analysis to verify the theoretical analysis.

Engineering the inter-island plasmonic coupling in homometallic Au-Au_(n)core-satellite structures

We show that through strong ligand mediated interfacial energy control between Au seeds and the deposited Au,the non-wetting growth of Au on Au seeds led to the formation homometallic core-satellite nanostructures.To modulate the intraparticle plasmonic coupling between the core and the satellites,the number and size of the Au satellites,and their inter-island distances were continuously tuned by varying the growth kinetics.As a result of the precise structural control,the plasmonic absorptions of the core-satellite nanostructures were tuned from visible to near-infrared(NIR)spectral range,and the extent of spectral modulation(500-1300 nm)is among the best of the literature methods.This synthetic advance enriches the toolbox for nanosynthesis and points to a new direction in the exploration of sophisticated functional designs.

Highly ordered Janus CdS-Au-TiO_(2) Z-scheme structure with high efficiency in photocatalysis

Precise structural control had attracted tremendous interest in nanosynthesis due to its great importance in tailoring the physical properties of nanomaterials.Here we report the synthesis of highly ordered Cd S-Au-TiO_(2) ternary Janus structure via templateprotected sequential growth and conversion method.Arising from the integration of the rectification effect of Au-CdS and AuTiO_(2) Schottky barriers,the Janus configuration of the Cd S-TiO_(2) domains,the plasmonic effect of Au nanosphere,and the Zscheme charge transportation,the Cd S-Au-TiO_(2) Janus structure showed high efficiency in the model photocatalytic degradation of methyl orange(MO)dye.Importantly,the well-defined structural order allowed the identification of the correlation between the structure and the catalytic performance.We believe that the synthetic control and the mechanism insights would help the design and synthesis of sophisticated nanostructures,and would eventually promote their applications in photocatalysis fields.

Nutritional requirements and precise fertilization of wine grapes in the eastern foothills of Helan Mountain

Cabernet Sauvignon grapes in the wine-producing area of Helan Mountain,East Ningxia,China,were the research object in this study.The dissection of the roots and branching stems method was used to explore the dynamic changes in the nitrogen,phosphorus,and potassium nutrient requirements of wine grapes over a number of growth stages.The results showed that over the whole growth period,the nitrogen content of the roots was the highest during the leaf-expansion stage and lowest during the turning-color stage,and that the nitrogen content of the leaves and fruit showed a downward trend as growth progressed.The nitrogen content of the secondary branches was the lowest during the fruit expansion stage and the highest during the leaf-expansion stage;and the phosphorus content of the roots was the highest during the leaf-expansion stage and lowest during the fruit expansion stage.The phosphorus content of the trunk and primary branches showed a trend of“rising-falling-rising”.The phosphorus content of the leaves and secondary branches was the lowest during the turning-color stage,whereas the phosphorus content of the fruit was at its highest during this stage.The potassium contents of the secondary branches and fruit showed a downward trend,but the potassium content of the leaves was highest during the fruit expansion stage and lowest in the nutrient return stage.Over the whole growth period,the accumulation of nitrogen,phosphorus,and potassium in wine grapes was 129.92 kg/hm2,41.51 kg/hm2,and 189.47 kg/hm2,respectively,the total requirements for N,P2O5,and K2O were 262.38 kg/hm2,288.15 kg/hm2,and 569.04 kg/hm2,respectively,and the reasonable nutrient requirement ratio was 1.00:1.10:2.17.

Combining the Curvature and Ligand Effects for Regioselective Growth on Au Nano-bipyramids

We demonstrate the regioselective growth of Au on Au nanobipyramids,either with etching of the tips,uniform coating except the tips,or forming a single island on the axial tip or an island on the equatorial corner.Importantly,the regioselectivity not only arises from the local curvature,as suggested by the recent literature,but also critically depends on the extent of ligand coverage on the seed surface.It is important to consider the competitive growth together with the curvature−ligand interplay:when there are insufficient ligands,they bind preferentially to the sharp tip for high surface energy,so that the remaining growth materials are diverted elsewhere;when the bipyramid is fully covered by ligands,the growth then selectively occurs at the tips because of the larger gaps among the ligands there.Our results demonstrate the great potentials in the rational design and synthesis when constructing sophisticated hybrid structures for functional nanomaterials.
