Recent advances in cancer-associated fibroblast:Biomarkers,signaling pathways,and therapeutic opportunities

Anti-cancer therapies usually focus on tumor cells,but non-tumor stromal components in the tumor microenvironment also play vital roles in tumor initiation and progression,which may be the prognostic factors and potential therapeutic targets.Cancer-associated fibroblasts(CAFs)are the essential component in the tumor environment,exhibiting high heterogeneity in their cell origin and phenotype with diverse functions that influence tumor angiogenesis,immune systems,and metabolism.Single-cell RNA sequencing and genetically engineered mouse models have increased our understanding of CAF diversity,and many subtypes have been defined.However,the precise functions of these subtypes need to be studied and validated.Studies of signaling pathways and epigenetic changes in CAFs facilitate understanding of the phenotypes of CAFs and the crosstalk between tumor cells and CAFs to provide potential therapeutic targets.Some clinical trials,including phase III trials targeting CAFs,have been performed recently.However,few of these trials have generated promising results,which indicates that the complexity of CAFs in the tumor microenvironment remains largely unknown,and in-depth investigations of CAFs should be performed.This review summarizes the research on CAFs,focusing on the heterogeneity of their phenotypes and functions,specific signaling pathways,and the therapeutic strategies involving CAFs.Additionally,we briefly discuss the current technologies commonly used in CAF studies and describe the challenges and future perspectives of CAF research.

Phase field modeling of ferroelastic variant switching in yttria-stabilized t'zirconia with strain gradient elasticity and interface tension

The 6–8 wt%yttria-stabilized zirconia with a tetragonal structure(t’-YSZ)is extensively employed in thermal barrier coatings.The exceptional fracture toughness of t’-YSZ can be attributed to its distinctive ferroelastic toughening mechanism.Microstructure and interface tension play a critical role in ferroelastic variant switching at the micro-and nano-scale.This paper presents an original thermodynamically consistent phase field(PF)theory for analyzing ferroelastic variant switching at the micro-and nano-scale of t’-YSZ.The theory incorporates strain gradient elasticity using higher-order elastic energy and interface tension tensor via geometric nonlinearity to represent biaxial tension resulting from interface energy.Subsequently,a mixed-type formulation is employed to implement the higher-order theory through the finite element method.For an interface in equilibrium,the effects of strain gradient elasticity result in a more uniform distribution of stresses,whereas the presence of interface tension tensor significantly amplifies the stress magnitude at the interface.The introduction of an interface tension tensor increases the maximum value of stress at the interface by a factor of 4 to 10.The nucleation and evolution of variants at a pre-existing crack tip in a mono-phase t’-YSZ have also been studied.The strain gradient elasticity is capable of capturing the size effect of ferroelastic variant switching associated with microstructures in experiments.Specifically,when the grain size approaches that of the specimen,the critical load required for variant switching at the crack tip increases,resulting in greater dissipation of elastic energy during ferroelastic variant switching.Moreover,the interface tension accelerates the evolution of variants.The presented framework exhibits significant potential in modeling ferroelastic variant switching at the micro-and nano-scale.

Surface wave inversion with unknown number of soil layers based on a hybrid learning procedure of deep learning and genetic algorithm

Surface wave inversion is a key step in the application of surface waves to soil velocity profiling.Currently,a common practice for the process of inversion is that the number of soil layers is assumed to be known before using heuristic search algorithms to compute the shear wave velocity profile or the number of soil layers is considered as an optimization variable.However,an improper selection of the number of layers may lead to an incorrect shear wave velocity profile.In this study,a deep learning and genetic algorithm hybrid learning procedure is proposed to perform the surface wave inversion without the need to assume the number of soil layers.First,a deep neural network is adapted to learn from a large number of synthetic dispersion curves for inferring the layer number.Then,the shear-wave velocity profile is determined by a genetic algorithm with the known layer number.By applying this procedure to both simulated and real-world cases,the results indicate that the proposed method is reliable and efficient for surface wave inversion.

Multiplexed manipulation of orbital angular momentum and wavelength in metasurfaces based on arbitrary complex-amplitude control

Due to its unbounded and orthogonal modes,the orbital angular momentum(OAM)is regarded as a key optical degree of freedom(DoF)for future information processing with ultra-high capacity and speed.Although the manipulation of OAM based on metasurfaces has brought about great achievements in various fields,such manipulation currently remains at single-DoF level,which means the multiplexed manipulation of OAM with other optical DoFs is still lacking,greatly hampering the application of OAM beams and advancement of metasurfaces.In order to overcome this challenge,we propose the idea of multiplexed coherent pixel(MCP)for metasurfaces.This approach enables the manipulation of arbitrary complex-amplitude under incident lights of both plane and OAM waves,on the basis of which we have realized the multiplexed DoF control of OAM and wavelength.As a result,the MCP method expands the types of incident lights which can be simultaneously responded by metasurfaces,enriches the information processing capability of metasurfaces,and creates applications of information encryption and OAM demultiplexer.Our findings not only provide means for the design of high-security and high-capacity metasurfaces,but also raise the control and application level of OAM,offering great potential for multifunctional nanophotonic devices in the future.

Programmable synthetic receptors:the next-generation of cell and gene therapies

Cell and gene therapies hold tremendous promise for treating a range of difficult-to-treat diseases.However,concerns over the safety and efficacy require to be further addressed in order to realize their full potential.Synthetic receptors,a synthetic biology tool that can precisely control the function of therapeutic cells and genetic modules,have been rapidly developed and applied as a powerful solution.Delicately designed and engineered,they can be applied to finetune the therapeutic activities,i.e.,to regulate production of dosed,bioactive payloads by sensing and processing user-defined signals or biomarkers.This review provides an overview of diverse synthetic receptor systems being used to reprogram therapeutic cells and their wide applications in biomedical research.With a special focus on four synthetic receptor systems at the forefront,including chimeric antigen receptors(CARs)and synthetic Notch(synNotch)receptors,we address the generalized strategies to design,construct and improve synthetic receptors.Meanwhile,we also highlight the expanding landscape of therapeutic applications of the synthetic receptor systems as well as current challenges in their clinical translation.

Molecular mechanism of Xuebijing in treating pyogenic liver abscess complicated with sepsis

BACKGROUND:Xuebijing(XBJ)can alleviate the inflammatory response,improve organ function,and shorten the intensive care unit(ICU)stay in patients with pyogenic liver abscess(PLA)complicated with sepsis,but the molecular mechanisms have not been elucidated.This study aimed to explore the molecular mechanism of XBJ in treating PLA complicated with sepsis using a network pharmacology approach.METHODS:The active ingredients and targets of XBJ were retrieved from the ETCM database.Potential targets related to PLA and sepsis were retrieved from the GeneCards,PharmGKB,DisGeNet,Online Mendelian Inheritance in Man(OMIM),Therapeutic Targets Database(TTD),and DrugBank databases.The targets of PLA complicated with sepsis were mapped to the targets of XBJ to identify potential treatment targets.Protein-protein interaction networks were analyzed using the STRING database.Potential treatment targets were imported into the Metascape platform for Gene Ontology(GO)functional enrichment and Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway enrichment analyses.Molecular docking was performed to validate the interactions between active ingredients and core targets.RESULTS:XBJ was found to have 54 potential treatment targets for PLA complicated with sepsis.Interleukin-1β(IL-1β),interleukin-6(IL-6),and tumor necrosis factor(TNF)were identifi ed as core targets.KEGG enrichment analysis revealed important pathways,including the interleukin-17(IL-17)signaling pathway,the TNF signaling pathway,the nuclear factor-kappa B(NF-κB)signaling pathway,and the Toll-like receptor(TLR)signaling pathway.Molecular docking experiments indicated stable binding between XBJ active ingredients and core targets.CONCLUSION:XBJ may exert therapeutic eff ects on PLA complicated with sepsis by modulating signaling pathways,such as the IL-17,TNF,NF-κB,and TLR pathways,and targeting IL-1β,IL-6,and TNF.

Critical layer in liquid-solid system influencing the remediation of chromium using zeolite-supported sulfide nano zero-valent iron

Sulfidated nano zero-valent iron particles were immobilized on ZSM-5 zeolite(Z/S-nZⅥ)and used for hexavalent chromium(Cr(Ⅵ))remediation.The performance of Z/S-nZⅥimproved with the increase in Cr(Ⅵ)concentration(0.6).Moreover,the removal mechanisms of Cr(Ⅵ)were elucidated through XRD,transmission electron microscopy(TEM)and XPS techniques.This results demonstrate that the species of chromium in the critical layer changed from Cr(Ⅲ)to Cr(Ⅵ)as the concentration of chromium increased from low to high.Furthermore,the critical layer was composed of Cr(Ⅵ),Fe(Ⅱ),O and H elements.Additionally,the experiments of coexisting ions and aging time confirmed that Z/S-nZⅥpossessed high selectivity and stability to ensure efficiency and cost-effectiveness in practical applications.

Track-before-detect for bistatic radar based on pseudo-spectrum

Track-Before-Detect(TBD) is an efficient method to detect dim targets for radars. Conventional TBD usually follows an approximate motion model of the target, which may cause an inaccurate integration of the target energy. A TBD technique on basis of pseudo-spectrum in mixed coordinates adopting an accurate motion model for bistatic radar system is developed in this paper.The predicted position in bistatic polar plane is derived according to a nonlinear function that exactly describes the constant Cartesian velocity motion. Then around the predicted position, a pseudo-spectrum is formulated and its samples are accumulated to the integration frame for energy integration. The evolution of the state and the procedure of accumulation of the target energy are derived elaborately. The superior performance of the proposed method is demonstrated by some simulations.

Effect of CO_(2) Opposing Multiple Jets on Thermoacoustic Instability and NO_(x) Emissions in a Lean-Premixed Model Combustor

This paper experimentally studied the effect of CO_(2) opposing multiple jets on the thermoacoustic instability and NO_(x) emissions in a lean-premixed model combustor.The feasibility was verified from three variables:the CO_(2) jet flow rate,hole numbers,and hole diameters of the nozzles.Results indicate that the control effect of thermoacoustic instability and NO_x emissions show a reverse trend with the increase of open area ratio on the whole,and the optimal jet flow rate range is 1-4 L/min with CO_(2) opposing multiple jets.In this flow rate range,the amplitude and frequency of the dynamic pressure and heat release signals CH~* basically decrease as the CO_(2) flow rate increases,which avoids high-frequency and high-amplitude thermoacoustic instability.The amplitude-damped ratio of dynamic pressure and CH*can reach as high as 98.75% and 93.64% with an optimal open area ratio of 3.72%.NO_(x) emissions also decrease as the jet flow rate increases,and the maximum suppression ratio can reach 68.14%.Besides,the flame shape changes from a steep inverted "V" to a more flat "M",and the flame length will become shorter with CO_(2) opposing multiple jets.This research achieved the synchronous control of thermoacoustic instability and NO_(x) emissions,which could be a design reference for constructing a safer and cleaner combustor.
