Oncolytic viruses(OVs)have attracted growing awareness in the twenty-first century,as they are generally considered to have direct oncolysis and cancer immune effects.With the progress in genetic engineering technology,OVs have been adopted as versatile platforms for developing novel antitumor strategies,used alone or in combination with other therapies.Recent studies have yielded eye-catching results that delineate the promising clinical outcomes that OVs would bring about in the future.In this review,we summarized the basic principles of OVs in terms of their classifications,as well as the recent advances in OV-modification strategies based on their characteristics,biofunctions,and cancer hallmarks.Candidate OVs are expected to be designed as“qualified soldiers”first by improving target fidelity and safety,and then equipped with“cold weapons”for a proper cytocidal effect,“hot weapons”capable of activating cancer immunotherapy,or“auxiliary weapons”by harnessing tactics such as anti-angiogenesis,reversed metabolic reprogramming and decomposing extracellular matrix around tumors.Combinations with other cancer therapeutic agents have also been elaborated to show encouraging antitumor effects.Robust results from clinical trials using OV as a treatment congruously suggested its significance in future application directions and challenges in developing OVs as novel weapons for tactical decisions in cancer treatment.

Dear Editor,An increasing body of evidence suggests that USP28 could be a target for cancer treatment^(1-4).To identify its inhibitors,we screened a 100-thousand synthetic compound library and found 3 lead compounds,CT1001-1003,that showed significant inhibitory activity(Supplementary Fig.1a).The optimization of these compounds led to CT1018,a much stronger inhibitor(Supplementary Fig.1b).

The reliability of a network is an important indicator for maintaining communication and ensuring its stable operation. Therefore, the assessment of reliability in underlying interconnection networks has become an increasingly important research issue. However, at present, the reliability assessment of many interconnected networks is not yet accurate,which inevitably weakens their fault tolerance and diagnostic capabilities. To improve network reliability,researchers have proposed various methods and strategies for precise assessment. This paper introduces a novel family of interconnection networks called general matching composed networks(gMCNs), which is based on the common characteristics of network topology structure. After analyzing the topological properties of gMCNs, we establish a relationship between super connectivity and conditional diagnosability of gMCNs. Furthermore, we assess the reliability of g MCNs, and determine the conditional diagnosability of many interconnection networks.

Review and comparison of methods and benchmarks for automatic modal identification based on stabilization diagram

Automatic modal identification via automatically interpreting the stabilization diagram provides key technique in bridge structural health monitoring.This paper reviews the progress in the area of automatic modal identification based on interpreting the stabilization diagram.The whole identification process is divided into four steps from establishing the stabilization diagram to removing the outliers in the identification results.The criteria and algorithms used in each step in the existing studies are carefully summarized and classified.Comparisons between typical methods in cleaning and interpreting the stabilization diagram are also conducted.Real structure benchmarks used in the existing studies to validate the proposed automatic modal identification methods are also summarized.Based on the review and comparison,the specific ratio method for cleaning the stabilization diagram,the hierarchical clustering method for interpreting the stabilization diagram and the adjusted boxplot for removing the outliers in the identification results are the most suitable methods for each step.The key point of automatic modal identification based on interpreting the stabilization diagram has also discussed,and it is recommended to pay more attention to cleaning the stabilization diagram.Future study about automatic modal identification under situation with very few sensors deployed should be more concerned.This review aims to help researchers and practitioners in implementing existing automatic modal identification algorithms effectively and developing more suitable and practical methods for civil engineering structures in the future.

Analysis and experimental study on resistance-increasing behavior of composite high efficiency autonomous inflow control device

Bottom water coning is the main reason to reduce the recovery of horizontal bottom water reservoir. By water coning, we mean the oil-water interface changes from a horizontal state to a mound-shaped cone and breaks through to the wellbore. Autonomous inflow control device(AICD) is an important instrument maintain normal production after bottom water coning, however, the resistance increasing ability of the swirl type AICD is insufficient at present, which seriously affects the water control effect. Aiming this problem, this paper designs a multi-stage resistance-increasing and composite type AICD. The separation mechanism of oil-water two phases in this structure, the resistance form of oil-water single phase and the resistance-increasing principle of water phase are analyzed. Establishing the dual-phase multi-stage separation and resistance-increasing model, and verified by measuring the throttling pressure drop and oil-water volume fraction of the AICD, it is found that the composite type AICD has the effect of ICD and AICD at the same time, which can balance the production rate of each well section at the initial stage of production, delay the occurrence of bottom water coning. In the middle and later stages of production, water-blocking can be effectively increased to achieve water control and stable production.After structural sensitivity analysis, the influence law of various structural parameters on the water control performance of composite AICD was obtained. The simulation calculation results show that,compared with the existing swirl type AICD, composite AICD has higher sensitivity to moisture content,the water phase throttling pressure drop is increased by 4.5 times on average. The composite AICD is suitable for the entire stage of horizontal well production.

MULTIPLE LEVELS OF PD-L1 REGULATION IN CANCER IMMUNOTHERAPY To date,the PD-1/PD-L1 blockade strategy has been tested in a variety of cancer types.However,only bladder cancer,melanoma,mismatch repair-deficient colorectal cancer,and certain hematopoietic malignancies have been highly responsive in terms of clinical outcomes.1,2 It is presumed that the expression level of the PD-L1 protein largely determines the therapeutic efficacy of PD-1/PD-L1-targeted cancer immunotherapy.3,4 Previous studies have shown that several transcriptional factors are involved in the upregulation of PD-L1 by directly binding to its promoter region.5 For instance,individual activation of STAT1,NF-κB,HIF-1α,c-Myc,or MAPK increased PD-L1 transcription and immune evasion of tumor cells.On the other hand,oncogenic RAS signaling reportedly stabilizes PD-L1 mRNA to promote tumor immunoresistance.
