A polyoxometalate based electrochemical sensor for efficient detection of L-cysteine

L-cysteine(L-cys),as an important sulfur-containing amino acid,plays an indispensable role in biological systems.Too low and excessively high ratio of L-cys will cause harm to the function of human organs.Therefore,it is very necessary to develop efficient methods to detect it in multifarious samples.This paper has built an electrochemical sensor by combining Keggin-type polyoxometalate(PMo9V3)and cobalt tetrasulfonate(Ⅱ)phthalocyanine(CoTsPc)on indium tin oxide electrodes using the layer-level self-assembly technology for efficiently detection of L-cys.The assembly process and surface morphology of the modified electrode was characterized by ultraviolet–visible spectroscopy,X-ray photoelectron spectroscopy,scanning electron microscope,and atomic force microscopy.The conditions of electro-catalysed oxidation of L-cys were optimized by cyclic voltammetry,and the kinetic electrochemical parameters were also evaluated by electrochemical impedance spectra.Furthermore,the sensing performance of the modified electrode was explored using amperometry.The proposed electrochemical L-cys sensor was found to have superior sensing performance with a range of linear response of 2.5×10^(−7) to 1.7×10^(−4) mol·L^(−1) and 1.7×10^(−4) to 39.5×10^(−4) mol·L^(−1),the detection limit of 1.0×10^(−7) mol·L^(−1)(signal/noise=3),and satisfactory anti-interference ability.Consequently,the fabricated sensor has the potential to be applied in laboratory practices for L-cys deterimination in commercial drinks.

Transmissive-detected laser speckle contrast imaging for blood flow monitoring in thick tissue: from Monte Carlo simulation to experimental demonstration

Laser speckle contrast imaging(LSCI)is a powerful tool to monitor blood flow distribution and has been widely used in studies of microcirculation,both for animal and clinical applications.Conventionally,LSCI usually works on reflectivedetected mode.However,it could provide promising temporal and spatial resolution for in vivo applications only with the assistance of various tissue windows,otherwise,the overlarge superficial static speckle would extremely limit its contrast and resolution.Here,we systematically investigated the capability of transmissive-detected LSCI(TR-LSCI)for blood flow monitoring in thick tissue.Using Monte Carlo simulation,we theoretically compared the performance of transmissive and reflective detection.It was found that the reflective-detected mode was better when the target layer was at the very surface,but the imaging quality would rapidly decrease with imaging depth,while the transmissivedetected mode could obtain a much stronger signal-to-background ratio(SBR)for thick tissue.We further proved by tissue phantom,animal,and human experiments that in a certain thickness of tissue,TR-LSCI showed remarkably better performance for thick-tissue imaging,and the imaging quality would be further improved if the use of longer wavelengths of near-infrared light.Therefore,both theoretical and experimental results demonstrate that TR-LSCI is capable of obtaining thick-tissue blood flow information and holds great potential in the field of microcirculation research.

Seven-year follow-up of the nonsurgical expansion of maxillary and mandibular arches in a young adult: A case report

BACKGROUND Palatal expansion treatment has been used to expand the constricted maxillary arch and has become a routine procedure in orthodontic practice over the past decades.However,the long-term stability of expansion in the permanent dentition without a surgical approach is uncertain.CASE SUMMARY We present the case of a 15-year-old boy with Class II malocclusion and constricted arches.The patient was treated with rapid palatal expansion(RPE)followed by a fixed orthodontic appliance.A 7-year follow-up evaluation was performed by analyzing cephalometric radiographs,plaster models,and photographs.The patient’s constricted maxillary and mandibular arches were relived after the expansion treatment.A Class I occlusion and normal arch form were established and maintained in the long-term.CONCLUSION RPE treatment is successful in solving constricted dental arch in the permanent dentition without a surgical approach.Permanent retention and even occlusal contact help prevent long-term relapse.

DNA methylation and demethylation link the properties of mesenchymal stem cells: Regeneration and immunomodulation

Mesenchymal stem cells(MSCs)are a heterogeneous population that can be isolated from various tissues,including bone marrow,adipose tissue,umbilical cord blood,and craniofacial tissue.MSCs have attracted increasingly more attention over the years due to their regenerative capacity and function in immunomodulation.The foundation of tissue regeneration is the potential of cells to differentiate into multiple cell lineages and give rise to multiple tissue types.In addition,the immunoregulatory function of MSCs has provided insights into therapeutic treatments for immune-mediated diseases.DNA methylation and demethylation are important epigenetic mechanisms that have been shown to modulate embryonic stem cell maintenance,proliferation,differentiation and apoptosis by activating or suppressing a number of genes.In most studies,DNA hypermethylation is associated with gene suppression,while hypomethylation or demethylation is associated with gene activation.The dynamic balance of DNA methylation and demethylation is required for normal mammalian development and inhibits the onset of abnormal phenotypes.However,the exact role of DNA methylation and demethylation in MSC-based tissue regeneration and immunomodulation requires further investigation.In this review,we discuss how DNA methylation and demethylation function in multi-lineage cell differentiation and immunomodulation of MSCs based on previously published work.Furthermore,we discuss the implications of the role of DNA methylation and demethylation in MSCs for the treatment of metabolic or immune-related diseases.

Durable response to pulsatile icotinib for central nervous system metastases from EGFR-mutated non-small cell lung cancer: A case report

BACKGROUND Central nervous system(CNS) metastases are a catastrophic complication of nonsmall cell lung cancer(NSCLC), including brain and leptomeningeal carcinomatosis, and are always accompanied by a poor prognosis. Despite the continuous development of existing treatments, the therapy of CNS metastases remains challenging.CASE SUMMARY We report a patient who was definitively diagnosed with brain and leptomeningeal metastases from NSCLC with a targeted mutation in epidermal growth factor receptor(EGFR). A standard dosage of icotinib(125 mg three times daily) was implemented but ineffective. CNS lesions developed despite stable systemic control, so pulsatile icotinib(1125 mg every 3 d) was administered. This new strategy for administration has lasted 25 mo so far, and resulted in complete remission of neurological symptoms, almost vanished lesions, and longer survival with no notable side effects.CONCLUSION This is the first successful example of pulsatile icotinib for treating isolated CNS progression from EGFR mutation-positive NSCLC, providing a new alternative for the local treatment of CNS metastases.

BACKGROUND Myxomas are benign tumors of mesenchymal origin that rarely occur in the larynx.CASE SUMMARY We report a case of a laryngeal myxoma that presented as a right vocal cord mass in a 54-year-old man.CONCLUSION Laryngeal myxoma is a rare benign tumor in the larynx.It is difficult to distinguish glottis myxoma from vocal cord polyps on laryngoscopy.We recommend that otolaryngologists acquire a better understanding of this disease.If a laryngeal myxoma is suspected,dynamic laryngoscopy,acoustic voice analysis,and pathological biopsy should be performed.

Fecal microbial dysbiosis in Chinese patients with inflammatory bowel disease

AIM To analyze the alterations of fecal microbiota in Chinese patients with inflammatory bowel disease(IBD).METHODS Fecal samples from 15 patients with Crohn's disease(CD)(11 active CD, 4 inactive CD), 14 patients with active ulcerative colitis(UC) and 13 healthy individuals were collected and subjected to 16 S ribosomal DNA(rDNA) gene sequencing. The V4 hypervariable regions of 16 S rDNA gene were amplified from all samples and sequenced by the Illumina Mi Seq platform. Quality control and operational taxonomic units classification of reads were calculated with QIIME software. Alpha diversity and beta diversity were displayed with R software.RESULTS Community richness(chao) and microbial structure in both CD and UC were significantly different from those in normal controls. At the phyla level, analysis of the microbial compositions revealed a significantly greater abundance of Proteobacteria in IBD as compared to that in controls. At the genera level, 8 genera in CD and 23 genera in UC(in particular, the Escherichia genus) showed significantly greater abundance as compared to that in normal controls. The relative abundance of Bacteroidetes in the active CD group was markedly lower than that in the inactive CD group. The abundance of Proteobacteria in patients with active CD was nominally higher than that in patients with inactive CD; however, the difference was not statistically significant after correction. Furthermore, the relative abundance of Bacteroidetes showed a negative correlation with the CD activity index scores.CONCLUSION Our study profiles specific characteristics and microbial dysbiosis in the gut of Chinese patients with IBD. Bacteroidetes may have a negative impact on inflammatory development.

Effect of liraglutide on endoplasmic reticulum stress in the renal tissue of type 2 diabetic rats

BACKGROUND Liraglutide is a glucagon-like peptide 1 receptor agonist analog that has been found to have a therapeutic effect in diabetes.In addition to its ability to treat diabetes,liraglutide has beneficial effects on the cardiovascular system and kidney as well as other beneficial effects,but its specific mechanism is not clear.In this study,a rat model of type 2 diabetes was established by administration of a high-sugar,high-fat diet combined with low-dose streptozotocin(STZ)to observe the effect of liraglutide on the kidneys of type 2 diabetes rats and the possible underlying mechanisms.AIM To explore whether liraglutide has a protective effect on type 2 diabetic rat kidneys and the underlying mechanisms.METHODS Eight-week-old male Sprague-Dawley rats were randomly divided into a control group,model group,low-dose liraglutide group,and high-dose liraglutide group.Control rats were fed a standard diet,while model group and intervention group rats were fed high-sugar,high-fat feed for 1 mo and then intraperitoneally injected with 40 mg/kg STZ to induce type 2 diabetes.The low-dose and highdose intervention groups received 100μg/kg and 200μg/kg liraglutide,respectively,once daily by subcutaneous injection.The control and model groups were given an equivalent volume of physiological saline for 8 wk.Pathological changes in renal tissues were observed by hematoxylin and eosin staining and periodic acid-Schiff staining,and GRP78 and caspase-12 expression was detected by Western blot and reverse transcription-polymerase chain reaction(RT-PCR).RESULTS Western blot analysis showed that GRP78 and caspase-12 protein expression in kidney tissue was significantly higher in model rats than in normal rats and lowerin the liraglutide-treated groups than in the model group,with a more significant decrease being observed in the high-dose group than in the low-dose group.RTPCR showed that the mRNA expression of GRP78 and caspase-12 was higher in model rats than in control rats and lower in the liraglutide-treated groups than in

Long noncoding RNAs in hepatitis B virus-related hepatocellular carcinoma

AIM: To study the expression of long noncoding RNAs(lncRNAs) in hepatitis B virus(HBV)-related hepatocellular carcinoma(HCC).METHODS: The lncRNA profiles between HBV-related HCC tissues and corresponding normal liver tissues were generated using microarray analysis. Datasets were analyzed using multiple algorithms to depict alterations in gene expression on the basis of gene ontology(GO), pathway analysis, and lncRNA levels.RESULTS: The microarray revealed that 1772 lncRNAs and 2508 mRNAs were differently expressed. The pathway analysis demonstrated that the cell cycle, cytokinecytokine receptor interaction, chemokine signaling pathway, and phosphoinositide 3-kinase-protein kinase B signaling pathway may play important roles in HCC.Several GO terms, such as cell cycle, DNA replication,immune response, and signal transduction, were enriched in gene lists, suggesting a potential correlation with HBVrelated HCC. The upregulated large intergenic noncoding RNA ULK4P2 was physically combined with enhancer of zeste homolog 2. Therefore, the lncRNAs may participate in regulating HBV-related HCC.CONCLUSION: lncRNAs play important roles in HCC,future studies should verify whether large intergenic noncoding ULK4P2 functions by combining with enhancer of zeste homolog 2 in HCC.
