Transcriptional regulatory network during axonal regeneration of dorsal root ganglion neurons:laser-capture microdissection and deep sequencing

The key regulators and regeneration-associated genes involved in axonal regeneration of neurons after injury have not been clarified.In high-throughput sequencing,various factors influence the final sequencing results,including the number and size of cells,the depth of sequencing,and the method of cell separation.There is still a lack of research on the detailed molecular expression profile during the regeneration of dorsal root ganglion neuron axon.In this study,we performed lase r-capture microdissection coupled with RNA sequencing on dorsal root ganglion neurons at 0,3,6,and 12 hours and 1,3,and 7 days after sciatic nerve crush in rats.We identified three stages after dorsal root ganglion injury:early(3-12 hours),pre-regeneration(1 day),and regeneration(3-7 days).Gene expression patterns and related function enrichment res ults showed that one module of genes was highly related to axonal regeneration.We verified the up-regulation of activating transcription factor 3(Atf3),Kruppel like factor 6(Klf6),AT-rich inte raction domain 5A(Arid5α),CAMP responsive element modulator(Crem),and FOS like 1,AP-1 transcription factor Subunit(Fosl1) in dorsal root ganglion neurons after injury.Suppressing these transcription factors(Crem,Arid5o,Fosl1 and Klf6) reduced axonal regrowth in vitro.As the hub transcription factor,Atf3 showed higher expression and activity at the preregeneration and regeneration stages.G protein-coupled estrogen receptor 1(Gper1),inte rleukin 12a(Il12α),estrogen receptor 1(ESR1),and interleukin 6(IL6) may be upstream factors that trigger the activation of Atf3 during the repair of axon injury in the early stage.Our study presents the detailed molecular expression profile during axonal regeneration of dorsal root ganglion neurons after peripheral nerve injury.These findings may provide reference for the clinical screening of molecular targets for the treatment of peripheral nerve injury.

Least square method based on Haar wavelet to solve multi-dimensional stochastic Ito-Volterra integral equations

This paper proposes a method combining blue the Haar wavelet and the least square to solve the multi-dimensional stochastic Ito-Volterra integral equation.This approach is to transform stochastic integral equations into a system of algebraic equations.Meanwhile,the error analysis is proven.Finally,the effectiveness of the approach is verified by two numerical examples.

Recovery from injury to the peripheral nervous system is different from that of the central nervous system in that it can lead to gene reprogramming that can induce the expression of a series of regeneration-associated genes.This eventually leads to axonal regeneration of injured neurons.Although some regeneration-related genes have been identified,the regulatory network underlying axon regeneration remains largely unknown.To explore the regulator of axon regeneration,we performed RNA sequencing of lumbar L4 and L5 dorsal root ganglion(DRG)neurons at different time points(0,3,6,12 hours,1,3 and 7 days)after rat sciatic nerve crush.The isolation of neurons was carried out by laser capture microscopy combined with NeuN immunofluorescence staining.We found 1228 differentially expressed genes in the injured sciatic nerve tissue.The hub genes within these differentially expressed genes include Atf3,Jun,Myc,Ngf,Fgf2,Ezh2,Gfap and Il6.We verified that the expression of the enhancer of zeste homologue 2 gene(Ezh2)was up-regulated in DRG neurons after injury,and this up-regulation differed between large-and small-sized dorsal root ganglion neurons.To investigate whether the up-regulation of Ezh2 impacts axonal regeneration,we silenced Ezh2 with siRNA in cultured DRG neurons and found that the growth of the newborn axons was repressed.In our investigation into the regulatory network of Ezh2 by interpretive phenomenal analysis,we found some regulators of Ezh2(including Erk,Il6 and Hif1a)and targets(including Atf3,Cdkn1a and Smad1).Our findings suggest that Ezh2,as a nerve regeneration-related gene,participates in the repair of the injured DRG neurons,and knocking down the Ezh2 in vitro inhibits the axonal growth of DRG neurons.All the experimental procedures approved by the Administration Committee of Experimental Animals of Jiangsu Province of China(approval No.S20191201-201)on March 21,2019.

Socialized care services for the aged population:System construction and support measures

Given the aging society,an increase in social demand,information-and communication technology-driven culture,and government policy support emerges to enable the development of the socialized care services system for the aged(SCSSA).The development of the SCSSA would be a significant step toward addressing China’s aging population.However,the construction of the SCSSA challenges the theories and methods of traditional elderly care service system construction.Specifically,the implementation path for such elderly care service policies is unclear,the necessary technological support is insufficient,and the mechanism for integrating intelligent information technology remains underexplored.Thus,this paper focuses on the needs of the elderly,grounded in the context of the changing elderly care service policies in China,and proposes a research paradigm that integrates system construction and support measure embedding.We then construct the original SCSSA,which includes“material+spirit+medical treatment+healthcare”and propose a method of optimization and iteration.Finally,we build the research framework of systematic support measures from the perspectives of policy reconstruction,institutional embeddedness,and technical support.Our work provides theoretical support and practical guidance for the construction and dynamic optimization of the SCSSA,thus making a significant contribution that will help China effectively cope with its aging society.

Quantitative attribution of Northern Hemisphere temperatures over the past 2000 years

Quantitative assessment of natural internal variability and externally forced responses of Northern Hemisphere(NH)temperatures is necessary for understanding and attributing climate change signals during past warm and cold periods.However,it remains a challenge to distinguish the robust internally generated variability from the observed variability.Here,largeensemble(70 member)simulations,Energy Balance Model simulation,temperature ensemble reconstruction,and three dominant external forcings(volcanic,solar,and greenhouse gas)were combined to estimate the internal variability of NH summer(June–August)temperatures over the past 2000 years(1–2000 CE).Results indicate that the Medieval Climate Anomaly was predominantly attributed to centennial-scale internal oscillation,accounting for an estimated 104%of the warming anomaly.In contrast,the Current Warm Period is influenced mainly by external forcing,contributing up to 90%of the warming anomaly.Internal temperature variability offsets cooling by volcanic eruptions during the Late Antique Little Ice Age.These findings have important implications for the attribution of past climate variability and improvement of future climate projections.

Covert communication based on transmission antenna selection in the downlink communication link

A downlink covert communication model that consists of a base station and two legitimate users was considered. In addition to the general signals shared by the two users, the base station will send the covert signals only to one user in a certain time without wanting the other to detect this covert communication behavior. In order to achieve covert communication, two information transmission schemes are designed based on transmission antenna selection(TAS) with the help of artificial noise(AN) transmitted by the user receiving the covert signals, denoted as TAS-Ⅰ and TAS-Ⅱ respectively. Considering the best detection performance of the user only receiving the general signals, under the two schemes, the detection error probabilities and their average values, the connection probabilities, the system covert throughputs are separately calculated. In addition, on the premise of meeting the system's covert conditions, an optimization scheme is proposed to maximize the covert system throughput. Finally, the simulation results show that the proposed system can realize covert communication successfully, and the system covert performance under TAS-Ⅰ is better than that under TAS-Ⅱ.

Telomerase-mediated immortalization of human vaginal wall fibroblasts derived from patients with pelvic organ prolapse

Background: Extracellular matrix (ECM) remodeling is the most important pathomechanism of pelvic organ prolapse (POP). Fibroblasts are the key to ECM regulation. The passaging capacity of human vaginal wall fibroblasts (hVWFs) is limited in vitro . Here, we aimed to immortalize hVWFs through the introduction of human telomerase reverse transcriptase (hTERT). Methods: Primary cells were derived from the vaginal wall tissue of patients with POP. Cellular senescence was detected via senescence-associated β-galactosidase staining. We employed a lentiviral transfection vector to stably express hTERT in hVWFs at passage 3, generating immortalized hVWFs (i-hVWFs). We then assessed cellular proliferation via the CCK-8 and EdU assays as well as cellular migration via wound healing assays. G-banded chromosome karyotypic analysis was performed to evaluate chromosomal karyotype stability. Finally, cellular tumorigenesis capacity was assessed in nude mice. A two-tailed Student’s t test was used to compare differences between the two groups. Results: Our results showed that senescence of primary hVWFs significantly increased from passage seven. From passage 11, hVWFs showed a significantly higher senescence percentage than i-hVWFs. During the continuous passage, i-hVWFs presented stability in proliferation, migration capacity, expression of ECM regulation-related genes, and chromosome karyotype. In vivo tumorigenesis was absent in i-hVWFs. Conclusions: The senescence of hVWFs significantly increased from the seventh passage, and we successfully used hTERT to immortalize hVWFs derived from patients with POP. Studies on POP that require a long-lived hVWF line will benefit from our technique.
