Parasitism-induced effects on host growth and metabolic efficiency in Plutella xylostella larvae parasitized by Cotesia vestalis or Diadegma semiclausum

The nutritional physiology of the diamondback moth, Plutella xylostella, larvae was examined after parasitization by the solitary endoparasitoids Cotesia vestalis or Diadegma semiclausum. Examinations were performed in two phases, one was examined at the time point of 24 h post-parasitization, and the other was done at the end of the 4th instar larvae of host. Rates of growth, food consumption, assimilation, excretion, and respiration were calculated as well as approximate digestibility and the rate ratios ECI （percent efficiency of conversion of ingested food to body substance）, and ECD （percent efficiency of conversion of digested food to body substance）. Parasitization by C. vestalis resulted in significant decrease in the rates of growth, feeding, excretion, assimilation, and respiration, but the final dry rate of respiration at the end of last larval stadium was elevated. The ECI and ECD were also reduced as the result of parasitization, but digestibility was increased. All these parameters in the larvae parasitized by D. semiclausum at 24 h post-parasitization were also significantly changed compared to the control; however, these differences were quantitatively, but not qualitatively before pupation, similar to those resulted from parasitization by C. vestalis. In spite of the similarities of the parasitism-induced effects caused by these endoparasitoids, the final metabolic rate, that is, the rate of intake of nutrients required to compensate for metabolism, was much lower in the larvae parasitized by C. vestalis than that of the larvae parasitized by D. semiclausum. All of the results discussed here will contribute toward explaining the different ways these two wasps regulate the parasitoid-host relationship.

Human umbilical cord mesenchymal stem cells(hUC-MSCs)are a promising candidate for spinal cord injury(SCI)repair owing to their advantages of low immunogenicity and easy accessibility over other MSC ***,modest clinical efficacy hampered the progression of these cells to clinical *** discrepancy may be due to many variables,such as cell source,timing of implantation,route of administration,and relevant efficacious cell dose,which are critical factors that affect the efficacy of treatment of patients with ***,we have evaluated the safety and efficacy of 4×10^(6) hUC-MSCs/kg in the treatment of subacute SCI by intrathecal implantation in rat *** search for a more accurate dose range for clinical translation,we compared the effects of three different doses of hUC-MSCs-low(0.25×10^(6) cells/kg),medium(1×10^(6) cells/kg)and high(4×10^(6) cells/kg)-on subacute SCI repair through an elaborate combination of behavioral analyses,anatomical analyses,magnetic resonance imaging-diffusion tensor imaging(MRI-DTI),biotinylated dextran amine(BDA)tracing,electrophysiology,and quantification of mRNA levels of ion channels and neurotransmitter *** study demonstrated that the medium dose,but not the low dose,is as efficient as the high dose in producing the desired therapeutic ***,partial restoration of theγ-aminobutyric acid type A(GABAA)receptor expression by the effective doses indicates that GABAA receptors are possible candidates for therapeutic targeting of dormant relay pathways in injured spinal ***,this study revealed that intrathecal implantation of 1×10^(6) hUC-MSCs/kg is an alternative approach for treating subacute SCI.

Porcine Small Intestinal Submucosa Mesh for Treatment of Pelvic Organ Prolapsed

Down-regulation of peroxisome proliferator-activated receptor γ coactivator-1α expression in fatty acid-induced pancreatic betacell apoptosis involves nuclear factor-κB pathway

Background Pancreatic beta-cell apoptosis induced by lipotoxicity, to a large extent, contributes to the progression of type 2 diabetes. To investigate the mechanism of free fatty acid induced apoptosis, we aimed to study the effects of palmitic acid （PA） on the apoptosis and peroxisome proliferator-activated receptor y coactivator-1α （PGC-1α） expression in βTC3 cells as well as the possible role of nuclear factor-KB （NF-KB） in this process. Methods Hoechst 33258 was used to detect βTC3 cell apoptosis, which was induced by PA stimulation for 12 hours. PGC-1α expression was analyzed by reverse transcription polymerase chain reaction, IκB kinase β （IKKβ）, IκBα NF-KB-inducing kinase （NIK） and ReI-B expressions were analyzed by Western blotting. MGβ2 was employed to block the endogenous IκBαdegradation before PA administration, and then its effect on PA-inducing cell apoptosis and PGC-1α mRNA expression was analyzed. Results Significant increased cell apoptosis was found at the concentration of 0.5 mmol/L and 1.0 mmol/L PA administration. PA （0.5 mmol/L） could extensively reduced the expression of PGC-1α mRNA. After exposing βTC3 cells to 0.5 mmol/L PA for different time periods （0, 4, 6, 8, 10 and 12 hours）, IKKβ protein expression increased while IκBα NIK and ReI-B protein expression declined in a time-dependent manner. Pretreatment with MGβ2 to inhibit the degradation of IκBα partially prevented the down-regulation of PGC-1α mRNA expression after 12-hour PA treatment in accordance with the decrease of PA induced apoptosis. Conclusions NF-KB canonical pathway was activated in PA-mediated βTC3 cell apoptosis, whereas non-canonical pathway was inhibited. Reduced PGC-1α expression by PA in βTC3 cells could involve the activation of canonical NF-KB pathway, so as to deteriorate the PA induced apoptosis.

The silk sericin hydrolysate（SSH） from the waste of silk processing as a substitute of fetal bovine serum（FBS）was used for the culture of Chinese hamster ovary（CHO） cells and Henrietta Lacks（Hela） strain of human cervical cancer *** survival ratio of these cells cultured in SSH media were similar to or higher than those in FBS *** after the serum was replaced by low concentration of SSH at 15.0 μg/ml for 5 d,the proliferation of both cells was also similar to or higher than that of FBS group;the percentages of CHO and Hela cells in S-phase were 28.9 and 28.0%,*** former is nearly two times that of FBS group,the latter is also higher than the control *** transcription-polymerase chain reaction（RT-PCR） revealed that among the differentially expressed genes,the relative expression of CXCL12 gene of CHO cells in SSH group increased,was three times that of serum group,and the relative expression of LCN2 gene of Hela cells increased 2.8 times,indicating that these related genes were activated to promote cell growth and *** results fully illustrated the hydrolysated sericin has a potential use as serum substitutes in cell culture.

Quantitative and HPLC fingerprint analysis combined with chemometrics for quality evaluation of Codonopsis Radix processed with different methods

Objective: To observe the effect of different processing methods on the quality of Codonopsis Radix and provide data reference for the reasonable processing in the producing ***: An analytical strategy that combined quantitative and HPLC fingerprint analysis with chemometrics was developed for the quality evaluation of Codonopsis Radix. Lobetyolin, polysaccharide, sucrose,glucose, and fructose were detected simultaneously in Codonopsis Radix samples treated with different processing methods including dryer-drying, sun-drying, shade-drying, sulfur fumigation, and kneading,***: The results showed that different processing methods had an obvious impact on the quality of Codonopsis Radix. Sun-drying or shade-drying was recommended, and sulfur fumigation should be avoided. And dryer-drying at 40 °C-50 °C was suggested to meet the requirement for large scale processing. In addition, based on the content of polysaccharide, glucose, and fructose, it was advised to knead for three times. As for lobetyolin and sucrose, kneading should be ***: Our findings demonstrated that processing method had a big impact on the quality of Codonopsis Radix. Based on the results, the appropriate processing methods directed at different requirements were recommended. The study could lay a foundation for the reasonable processing of Codonopsis Radix in producing areas.

目的观察未累及黄斑区的缺血型视网膜分支静脉阻塞(BRVO)患者眼底激光治疗前后对比敏感度(CS)的变化情况。方法收集38眼未累及黄斑区的缺血型BRVO患者进行眼底激光治疗,并观察3个月,分别测试激光治疗前及治疗后1和3个月的矫正视力及空... 详细信息

目的观察未累及黄斑区的缺血型视网膜分支静脉阻塞(BRVO)患者眼底激光治疗前后对比敏感度(CS)的变化情况。方法收集38眼未累及黄斑区的缺血型BRVO患者进行眼底激光治疗,并观察3个月,分别测试激光治疗前及治疗后1和3个月的矫正视力及空间频率为1.5、3、6、12、18周/度(c/d)处的CS。CS两组间的比较采用两独立样本t检验,CS及矫正视力的多组间比较采用单因素方差分析,组间多重比较方差齐性时采用LSD检验,方差不齐性时采用Dunnett’s T3检验。结果所有患者激光治疗后3个月通过眼底荧光血管造影(FFA)检查证实显效的20眼(52.63%),有效13眼(34.21%),无效的5眼(13.16%),总共治疗有效33眼(86.84%)。所有患者激光治疗前及激光治疗后1和3个月与对照组比较,差异有统计学意义(P0.05)。将BRVO按阻塞部位、范围及非灌注区面积分组比较激光治疗后CS降低值,差异有统计学意义(P<0.05),显示阻塞部位在颞侧及阻塞范围较大、非灌注区面积较大的患者在激光治疗后在高频区(12c/d、18c/d)CS降低较多。随访3个月,BRVO患者激光治疗后与未进行激光治疗的患者比较,差异有统计学意义(P<0.05),各空间频率下的CS降低,特别是12c/d、18c/d空间频率下CS降低明显。结论眼底激光治疗是治疗缺血型BRVO的有效方法,但是激光治疗对于患者视网膜功能也是一种损伤,可造成未累及黄斑区的缺血型BRVO患者CS下降,尤其是高频区CS。
