目的:基于Toll样受体4(TLR4)/髓样分化因子88(MyD88)/核因子κB(NF-κB)信号通路介导的巨噬细胞焦亡探讨黄芩黄连药对(芩连药对)治疗动脉粥样硬化(AS)的作用机制。方法:体外培养J774A.1细胞,SD大鼠灌胃芩连药对后制备含药血清,MTT法筛选芩连药对含药血清最佳干预浓度。采用不同浓度的含药血清预处理J774A.1细胞后,给予脂多糖(LPS)+三磷酸腺苷(ATP)诱导焦亡。光学显微镜下观察细胞形态变化,MTT法检测细胞活力,乳酸脱氢酶(LDH)释放法及Hoechst 33342/碘化丙锭(PI)染色用于检测细胞的损伤情况,酶联免疫吸附测定(ELISA)法测定细胞上清液中白细胞介素(IL)-1β、IL-18的水平。Western Blot法检测通路及焦亡相关蛋白TLR4、MyD88、NF-κB p65、p-NF-κB p65、NLRP3、ASC、Caspase-1、Cleave-Caspase-1、GSDMD、GSDMD-NT的表达。TLR4基因过表达后,检测通路相关蛋白表达水平及细胞焦亡情况。结果:芩连药对含药血清可改善造模后J774A.1的形态;提高造模后J774A.1细胞的活力、降低LDH释放率及PI细胞阳性率(P<0.05);降低细胞上清液中IL-1β及IL-18的水平(P<0.05);显著下调TLR4/MyD88/NF-κB信号通路及焦亡相关蛋白的表达(P<0.05);同时过表达TLR4可逆转芩连药对对J774A.1细胞焦亡的保护作用(P<0.05)。结论:芩连药对可通过抑制TLR4/MyD88/NF-κB信号通路降低巨噬细胞焦亡水平,从而发挥抗AS的作用。

目的:探讨核苷酸结合寡聚化结构域样受体蛋白3(NLRP3)炎症小体通过JAK/STAT3信号通路对脓毒症大鼠的肠道炎症反应和肠粘膜损伤的作用机制。方法:选择健康清洁级雄性Wistar大鼠30只,随机分为三组:假手术组、模型组与NLRP3抑制剂组,各10只,模型组和抑制剂组采用盲肠结扎穿孔术进行造模,抑制剂组于造模前30 min腹腔注射NLRP3抑制剂MCC950(10 mg/kg),模型组注射等量生理盐水。观察各组大鼠24 h的生存情况,HE染色进行回肠病理评分,ELISA法检测血清肿瘤坏死因子-α(TNF-α)和白细胞介素-1β(IL-1β),Western blot法检测小肠组织NF-κB p65、NLRP3、凋亡相关斑点蛋白(ASC)、天冬氨酸特异性半胱氨酸蛋白酶1(Caspase-1)、p-JAK和p-STAT3蛋白表达量。结果:(1)与假手术组相比,模型组大鼠的死亡率和病理评分显著增加,血清TNF-α和IL-1β水平升高,组织NF-κB p65、NLRP3、ASC、Caspase-1、p-JAK和p-STAT3蛋白的相对表达量均显著增加(P<0.05)。(2)与模型组相比,抑制剂组大鼠的死亡率和病理评分显著降低,血清TNF-α和IL-1β水平,组织NF-κB p65、NLRP3、ASC、Caspase-1、p-JAK和p-STAT3蛋白的表达量显著下降(P<0.05)。抑制剂组与假手术组相比,上述指标间仍存在显著的统计学差异(P<0.05)。结论:NLRP3炎症小体可能通过活化JAK/STAT3信号通路增强脓毒症大鼠的肠道炎症反应,损伤肠道黏膜,靶向干预NLRP3能够逆转肠道损伤。

In this study,a natural cotton thread immunoassay device combined with gold nanorod（GNR） reporter probe is developed for the rapid,sensitive and quantitative electrochemical determination of human ferritin,a lung cancer related *** ferritin as an analyte and a pair of monoclonal antibodies are used to demonstrate the proof-of-concept on the cotton thread immunoassay *** enhancement of the sensitivity is achieved by using gold nanorod as an electroactive report probe compared with a traditional gold nanoparticle（GNP） report *** device was capable of measuring 1.58 ng/mL ferritin in 30 min by anodic stripping voltammetry（ASV） testing,which meet the requirement for clinical diagnosis.

无人机集群协同区域搜索能够有效地获取任务区域地面信息,降低环境不确定度。基于区域划分、机群均衡分配以及启发式算法的传统集群协同区域搜索方法依赖于事前设计规则且计算量大,属于不可生成规则算法。考虑任务环境不确定性,算法须满足快速性、智能性和鲁棒性,基于涌现理论的无人机集群协同搜索方法因信息融合能力强、具有高度的智能性而被采用。演化学习算法和强化学习算法是涌现理论中主要组成部分,这两类算法可根据不同的环境和任务生成新的集群行为规则。将系统分析和总结当前无人机集群协同搜索方法研究现状和进展,并据此指出现有研究中的不足以及未来的发展方向。

Roll profile electromagnetic control characteristics under roll heterogeneity

The electromagnetic control roll(ECR)and electromagnetic stick(ES)are the core elements and the main driving parts of roll profile electromagnetic control technology(RPECT).To prolong the service life,it is necessary to treat ECR and ES *** to the heterogeneous characteristics of surface treatments,the roll profile electromagnetic control characteristics were analyzed for different *** electromagnetic-thermal-force coupled axisymmetric finite element model was built to explore the differences in performance as a result of several treatment strategies,and the model was verified by experimental results measured with a roll profile electromagnetic control experimental *** model was used to analyze the influence of the heterogeneity of ECR inner hole and ES on the roll crown,the roll profile,the average contact pressure,and the stress state during RPECT *** results indicate that the heterogeneous layer at ECR inner hole has a restrictive effect on RPECT and that the heterogeneous layer of ES can enhance the profile control ability of RPECT.A reasonable configuration scheme between the heterogeneity of ECR inner hole and the ES can increase the life of ECR and maintain the control ability of RPECT.

Open-shell oligomers and polymers have exhibited intriguing electronic and magnetic properties, making them highly desirable for a wide range of applications, including ambipolar organic field-effect transistors (OFETs), photodetectors, organic thermoelectrics, and spintronics. Although open-shell ground states have been observed in certain small molecules and doped organic semiconductors, the exploration of open-shell ground-state conjugated polymers is still limited, and the strategies for designing these polymers remain obscure. This review aims to briefly introduce the theory and characterization methods of open-shell conjugated polymers, along with an overview of recent progress and applications. The objective is to stimulate further advancements and investigations in this promising area by shedding light on the potential of open-shell conjugated polymers and the challenges that lie ahead.

The crystallization process of Fe78ZrTBls （at%） amorphous ribbon was investigated by Xray diffraction （XRD）, differential scanning calorimetry and scanning electron microscopy （SEM）. The fully amorphous structure of asquenched （Aq） ribbons was confirmed by XRD pattern. The saturation magnetization （Ms） and Curie tem perature of the Aq ribbon were measured as 124.3 （***）＆g and 305 ℃ with vibrating sample magnetometer （VSM）, respectively. When the ribbons was annealed at 550 ℃ near the first onset temperature （Txl = 564.9 ℃）, the Ms was increased by 17 %, which was caused by the formation of a dual phase structure. The isothermal crystallization kinetics and crystallization mechanism of primary ctFe phase in the dual phase structure were studied by Arrhenius and JohnsonMehlAvramiKolmogorov equations respectively. The results showed that the crystallization of Fe phase was a diffusioncontrolled surface nucleation growth process, and the nucleation rate decreased with longer crystallization time.

The objective of this study is to propose a more accurate and faster MTT 3-(4,5-dimethyl thiazol-2-yl)-2,5-diphenyl tetrazolium bromide colorimetric assay (MCA) for quantitative measurement of polypeptide bacteriocins in solutions with nisin as an example. After an initial incubation of nisin and indicator bacterium Micrococcus luteus NCIB 8166 in tubes, MTT was added for another incubation period. After that, nisin was quantified by estimating the number of viable bacteria based on measuring the amount of purple formazan produced by cleavage of yellow tetrazolium salt MTT. Then MCA was compared to a standard agar diffusion assay (ADA). The results suggested a high correlation coefficient (r2=0.975±0.004) between optical density (OD) and the inhibitory effect of nisin on a bacterial strain Micrococcus luteus NCIB 8166 at a range of 0.125～32 IU/ml. The MCA described in this study was very quick. Quantification of nisin took only 7～8 h and the detection limit was at the level of 0.125 IU/ml when compared to 12 IU/ml and 24～28 h for ADA. The MCA provides an accurate and rapid method for quantifi-cation of nisin in solutions and is expected to be used for quantification of other antimicrobial substances.

By means of circular dichroism（CD） spectrum coupled with UV-Vis and fluorescence spectra,the binding model of DNA intercalator A1{4-（2-diethylamino-ethylamino）-8-oxo-8H-acenaphtho[1,2-b]pyrrole-9-carbonitrile} to calf thymus（CT） DNA was investigated,depending on the values of R（R is defined as the ratio of the concentration of A1 to CT DNA base pairs） and different outer *** A1 were intercalated into the CT DNA base pairs in different orientations in the intercalation pocket at a lower R value（R≤0.20）,while A1 molecules aggregated on the surface of the helix of the CT DNA as the R value *** influence of NaCl on the binding was smaller because the electrostatic interaction only provided approximately 16% of the overall free energy of *** protonated diethylamine substitution would influence the binding geometry greatly at a low pH value via forming hydrogen bonds with the exposed C=O group on DNA surface.
