Axonal regeneration in the central nervous system is an energy-intensive *** contrast to mammals,adult zebrafish can functionally recover from neuronal *** raises the question of how zebrafish can cope with this high energy *** previously showed that in adult zebrafish,subjected to an optic nerve crush,an antagonistic axon-dendrite interplay exists wherein the retraction of retinal ganglion cell dendrites is a prerequisite for effective axonal *** postulate a‘dendrites for regeneration’paradigm that might be linked to intraneuronal mitochondrial reshuffling,as ganglion cells likely have insufficient resources to maintain dendrites and restore axons ***,we characterized both mitochondrial distribution and mitochondrial dynamics within the different ganglion cell compartments(dendrites,somas,and axons)during the regenerative *** nerve crush resulted in a reduction of mitochondria in the dendrites during dendritic retraction,whereafter enlarged mitochondria appeared in the optic nerve/tract during axonal *** dendritic regrowth in the retina,mitochondrial density inside the retinal dendrites returned to baseline ***,a transient increase in mitochondrial fission and biogenesis was observed in retinal ganglion cell somas after optic nerve *** together,these findings suggest that during optic nerve injury-induced regeneration,mitochondria shift from the dendrites to the axons and back again and that temporary changes in mitochondrial dynamics support axonal and dendritic regrowth after optic nerve crush.

Activity of a lipid synthesis inhibitor(spiromesifen) in Culiseta longiareolata(Diptera:Culicidae)

Objective: To evaluate the activity of spiromesifen against the most abundant and investigated mosquito species, Culiseta longiareolata Aitken, 1954(Diptera, Culicidae).Methods: Culiseta longiareolata larvae were collected from untreated areas located at Te′bessa(Northeast Algeria). A commercial formulation of spiromesifen(Oberon?240 SC) was tested at different concentrations ranging between 238 and 1 428 mg/L on newly molted fourth-instar larvae under standard laboratory conditions according to Word Health Organization recommendations. The effects were examined on the mortality, the morphometric measurements, two biomarkers(catalase and malondialdehyde), and the biochemical composition of larvae, ***: The compound exhibited insecticidal activity. Moreover, it disturbed growth and several morphological aberrations were observed. It also affected body volume,biomarkers and contents of carbohydrates, lipids and proteins. A marked effect on lipids and malondialdehyde was noted, confirming its primary mode of action on lipid ***: Spiromesifen appears less potent than other insecticides tested such as the insect growth disruptors.

Has the risk of colorectal cancer in inflammatory bowel disease decreased?

The association between inflammatory bowel disease(IBD)and colorectal cancer(CRC)has been acknowledged for almost a century and is assumedly promoted by a chronic inflammation-driven carcinogenic process in the intestine in combination with a genetic *** magnitude of the risk of CRC in IBD remains a continuing subject of *** early,high risk estimates for CRC in IBD were most likely overestimated due to selected patient populations originating from tertiary referral centers with a disproportional high percentage of patients with severe *** population-based studies calculating risk estimates from a broad spectrum of IBD patients have found the risk to be significantly *** present,there is evidence that IBD patients with longstanding and extensive disease with uncontrolled inflammation are those at increased ***,other recognized risk factors include early age at onset,family history of CRC,and concomitant primary sclerosing cholangitis.A significant amount of effort is put into identifying potential preventive factors of CRC in IBD,including surveillance programs and chemopreventive agents but the individual effect of these remains ***,recent studies have reported a decline in risk of CRC over *** programs and the new treatment strategies,particular biological treatment might be part of the reason for the observed decline in risk of CRC in IBD over time but future studies will have investigate this assumption.

Risk of infections associated with biological treatment in inflammatory bowel disease

Tumor necrosis factor-α (TNF-α) inhibitors are biological agents introduced in the late 1990s for the treatment of different immune-mediated diseases as inflammatory bowel disease, rheumatoid arthritis and psoriasis. The most commonly used TNF-α antagonists are infliximab, adalimumab, and certolizumab pegol, and though highly effective in lowering inflammation, the efficacy must be weighed against the potential for adverse events. The treatment-induced immunosuppression is suspected to increase the risk of infections, including the risk of reactivation of latent tuberculosis, as the TNF-α cytokine plays an important role in the immune function. In this topic highlight a short overview of the infection risk associated with TNF-α inhibiter therapy is outlined with a focus on the overall risk of serious infections, mycobacterial infection and latent viral infections.

The emergence of immunotherapy as a cancer therapy has dramatically changed the treatment paradigm of systemic cancer *** have been several trials evaluating immune checkpoint blockade(ICI)in soft tissue *** there is generally a limited response in sarcoma,a subset of patients has durable responses to *** is attributable to a variety of factors including histologic subtype,tumor-infiltrating lymphocytes,and the tumor microenvironment among *** is ongoing translational and clinical research evaluating ICI resistance in sarcoma and identifying therapeutic strategies to overcome this ***,we provide a review of the current data,proposed mechanisms of resistance,and potential approaches to overcome this resistance.

Many cancers lack functional expression of the enzyme argininosuccinate synthetase 1(ASS1)that is necessary for synthesis of *** cancers must import arginine for survival and growth,and this reliance can be targeted by arginine-degrading extracellular enzymatic drugs,most commonly PEGylated arginine *** enzymes can become targets of the immune system,reducing their effectiveness,but PEGylation improves the in vivo *** deprivation causes cell death in some cancers,but others gain resistance by expressing ASS1 after a starvation response is *** resistance mechanisms are possible and explored,but these have not been observed specifically in response to arginine *** studies,especially focusing on the mechanisms of ASS1 upregulation and metabolic adaptations,may yield insights into preventing or taking advantage of resistance adaptations to make arginine deprivation therapy more effective.

Morphological and functional evaluation of chronic pancreatitis with magnetic resonance imaging

Magnetic resonance imaging(MRI)techniques for assessment of morphology and function of the pancreas have been improved dramatically the recent years and MRI is very often used in diagnosing and follow-up of chronic pancreatitis(CP)*** MRI including fat-suppressed T1-weighted and T2-weighted imaging techniques reveal decreased signal and glandular atrophy of the pancreas in *** contrast-enhanced MRI of the pancreas in CP the pancreatic signal is usually reduced and delayed due to decreased perfusion as a result of chronic inflammation and ***,morphological changes of the ductal system can be assessed by magnetic resonance cholangiopancreatography(MRCP).Furthermore,secretin-stimulated MRCP is a valuable technique to evaluate side branch pathology and the exocrine function of the pancreas and diffusion weighted imaging can be used to quantify both parenchymal fibrotic changes and the exocrine function of the *** standard and advanced MRI techniques are supplementary techniques to reveal morphological and functional changes of the pancreas in ***,spectroscopy has been used for assessment of metabolite concentrations in-vivo in different tissues and may have the potential to offer better tissue characterization of the ***,the purpose of the present review is to provide an update on standard and advanced MRI techniques of the pancreas in CP.

At the forefront of cancer research is the rapidly evolving understanding of metabolic reprogramming within cancer *** expeditious adaptation to metabolic inhibition allows cells to evolve and acquire resistance to targeted treatments,which makes therapeutic exploitation complex but achievable.3-phosphoglycerate dehydrogenase(PHGDH)is the rate-limiting enzyme of de novo serine biosynthesis and is highly expressed in a variety of cancers,including breast cancer,melanoma,and Ewing’s *** review will investigate the role of PHGDH in normal biological processes,leading to the role of PHGDH in the progression of *** an understanding of the molecular mechanisms by which PHGDH expression advances cancer growth,we will highlight the known mechanisms of resistance to cancer therapeutics facilitated by PHGDH biology and identify avenues for combatting PHGDH-driven resistance with inhibitors of PHGDH to allow for the development of effective metabolic therapies.
