Differential signaling regulatory networks governing hormone refractory prostate cancers

To understand the organization of the biological networks that might potentially govern the pathogenesis of hormone refractory prostate cancer （HRPC）, we investigated the transcriptional circuitry and signaling in androgen-dependent 22Rvl and MDA PCa 2b cells, androgen- and estrogen-dependent LNCaP cells, and androgen-independent DU 145 and PC-3 prostate cancer （PCa） cell lines. We used microarray analyses, quantitative real-time PCR, pathway prediction analyses, and determination of Transcription Factor Binding Site （TFBS） signatures to dissect HRPC regulatory networks. We generated graphical representations of global topology and local network motifs that might be important in prostate carcinogenesis. Many important putative biomarker ＇target hubs＇ were identified in the current study including AP-1, NF-KB, EGFR, ERK1/2, JNK, p38 MAPK, TGF beta, VEGF, PDGF, CD44, Akt, PI3K, NOTCH1, CASP1, MMP2 and AR. Our results suggest that complex cellular events including autoregulation, feedback loops and cross-talk might govern progression from early lesion to clinically diagnosed PCa, as well as metastatic potential of pre-existent high-grade prostate intraepithelial neoplasia （HG-PIN） and/or advancement to HRPC. The identification of TFBS signatures for TCF/LEF, SOX9 and ELK1 in the regulatory elements suggests additional biomarkers for the potential development of chemopreventive/therapeutic strategies against PCa. Taken together, in this study, we have identified putative biomarker ＇target hubs＇ in the architecture of PCa signaling networks, and investigated TFBS signatures that might enhance our understanding of key regulatory nodes in the progression and pathogenesis of HRPC.

The incidence of prostate cancer is rising in the Asia-Pacific region as well as other countries. Androgen-ablation therapy is clinically useful in the androgen-dependent phenotype however, many patients progress to hormone refractory prostate cancer that is difficult to treat and needs newer interventions that are more effective. The objective of this study was to determine functionally-relevant biological networks, to appreciate the potential crosstalk between signaling members, and to identify biomarker signatures in prostate cancer. We used microarray analyses to identify key genes that were upregulated or down regulated at least five-fold in human prostate cancer and constructed canonical interaction networks that are important in prostate cancer through metabolomics analyses. Our prostate cancer network architecture revealed several key biomarkers including ERK1/2, JNK, p38, MEK, PI3 K, NFκB, AP-1, 14-3-3, VEGF, PDGF, Rb, WNT8 A, WNT10 A, CD44, ESR2, FSH and LH. Furthermore, the top ten transcription factors identified by TFBS-association signature analysis in the regulatory elements of co-regulated biomarkers were delineated, which may crosstalk with upstream or downstream genes elicited in our network architecture. Taken together, our results demonstrate that the regulatory interaction networks in prostate cancer provide a universal view of crosstalk between important biomarkers, i.e., key players in the pathogenesis of this disease. This will facilitate more rapid screening of functional biomarkers in early/intermediate drug discovery.

Characterization of Myanmar Paw San Hmwe Accessions Using Functional Genetic Markers

Paw San Hmwe （PSM） rice has been cultivated in many areas of Myanmar for a long time. Strong aroma, good taste and its elongation during cooking are the key characteristics of PSM rice. Thirty-one PSM accessions were genotypically characterized, and their physical grain and cooking quality traits were studied. We used specific gene markers associated with aroma, apparent amylose content （AAC） and alkali spreading value to determine the alleles carried by different PSM accessions. The results revealed that six PSM accessions （PSM10, PSM12, PSM13, PSM21, PSM22 and PSM30） had a 3-bp insertion in Os2AP gene. Gel consistency （GC） allele was predominant among the PSM accessions for gelatinization temperature （GT）, however, the phenotype observed was between low and intermediate GT because of the combination of the GC allele with the presence of low GT allele at heterozygous state from the other loci of the SSIla gene. Intermediate to high AAC was observed among the PSM accessions corresponding to the haplotype identified for the single nucleotide polymorphism G/T and the （Cm）n repeat in the Wx gene. The characterization and grouping data of PSM accessions posted benefits to Myanmar seed banks, and our results will help in maintaining the integrity of PSM rice variety.

In disaster response,collaboration facilitates interactions among actors,such as the government,the military,nongovernmental organizations,and civil society *** study examined the longitudinal changes in collaborative governance in Myanmar’s disaster responses based on cases of flooding in 2015,2016,and *** examine the mechanisms underlying this dynamic network formation,the collaborative ties of the actors involved in search and rescue activities were converted into longitudinal relational data sets,and the evolution of collaborative governance was analyzed by relying on the assumptions of social capital,transaction cost,homophily,and resource dependency theories and using a longitudinal social network analysis *** findings show that the collaborative networks of search and rescue processes in disaster response evolved and changed over time according to the hypothesized patterns of strong,weak,and preferential tie *** study also revealed that the collaborative governance system assumes the form of a hierarchy rather than a generalized exchange,and the actors’reliance on military organizations is not obvious due to the emerging alternative non-military actors and diverse local actors observed in the cases.

Seasonal Prevalence of <i>Aedes aegypti</i>in Semi-Urban Area of Yangon Region, Myanmar

Prevalence rate of Aedes aegypti was conducted in 20 houses from semi-urban areas of Yangon Region. Larval surveys were done at indoors and outdoors water containers of five types. Prevalence rate of larval density larvae was investigated monthly by standard indices. The highest infestation rate of the container index (CI) was in June 2018 (56.52%), the second highest was in July 2017 (48.36%) and the lowest rate was in April 2017 (5.07%);those of the Jar index (JI) was highest (36.49%) in June and second highest rate (23.8%) was in October 2017. Reasoning the Metal drum (MI) was highest (13.95%) in June 2018 and second highest (6.25%) was in July 2017. The larval infestation rate of Earthen pot (EI) was highest (42.1%) in July 2017. The larval incident rate in almost all indices showed that the highest rate was at the beginning of monsoon season, in June and July, while in the remaining months, the larval incident rate was found to decrease due to the application of insecticides in the study area by the Township Public Health Department. However, the application of insecticides did not cover all the breeding sites of the mosquitoes, the water puddles under their houses were left to apply the insecticides. The positive larval incident rate was assessed by Household (HI), Container index (CI), Breteau index (BI). The highest and second highest positive larval incident rates were all in June 2018 and July 2017 in all indices, HI (27.3% and 23.4%), CI (56.52% and 48.36%), BI (17.56% and 16.79%) and SI (28.49% and 24.38%) respectively. The lowest rate in all indices was 2.56% (HI), 5.07% (IC), 2.67% (BI) and 1.91% (SI) in April. In this study, the fluctuation of indices of infestation rates and positive larval index value was positively correlated in similar trends in the study months. Diversity of Insect Pest and Predator Species in Monsoon and Summer Rice Fields of Taungoo Environs, Myanmar

Paddy fields are natural and artificial wetland ecosystems that supply rice for the people and provide the wildlife especially insect diversity of different functional aspects. A total of 71 insect species belonging to 40 families under eight orders were observed during the study period. Among the 71 insect species, 18 species of beetles, nine species of bugs, eight species of dragonfly, five species of butterflies, four species of leafhoppers, plant hoppers and moths, three borer and spiders, two crickets, one species of skippers, grass hopper, hispa, ant, weevil, hairy caterpillar, leaf roller, katydid, thrips, maggot and water boatmen were recorded in the study sites. Total of 41 species of pests, 18 species of predators and 12 species of beneficial species (they function as pollinating the flowering plants in the paddy field wetland ecosystem) were recorded in the study sites. In the monsoon season, the 41 species of pest species, 18 species of predator species and 12 species of beneficial species were recorded from monsoon rice field. According to Shannon Evenness value (H'/S = -0.012564), the data showed that the insect species recorded from the one habitat was not the similar to another. In summer paddy fields, 36 species of pest species, 16 species of predator species and 9 species of beneficial species were recorded. Total arthropod insect species were recorded 61 species from the dry rice fields. According to Shannon Evenness value (H'/S = -0.000120), the data showed that the insect species recorded from the one habitat was not the similar to another. Population growth and duration of life cycle of insects is mainly dominated by the temperature, the duration of life cycle is shorter in the high temperature than in the low temperature.

Evaluation of Stope Stability in Underground Mine;Hermyingyi (Sn-W Deposit) Mine in Myanmar

The stability of underground excavations has become an important issue in order to extend underground mining operations and extract deeper deposits. The increasing demand for tin-Tungsten (Sn-W) for industry and its market price has created a motivation for mining companies to extract deep-seated Sn-W ore deposits in Myanmar. Thus, this paper aims to investigate the stability of underground openings, especially, the stope with considering the mining methods. To meet the objective, FLAC3D 5.0 simulation was used for the assessment of stope under different stress ratios, 0.5, 1.0, and 1.5 for two types of underground mines;Open stoping and Cut and Fill stoping. The results show that the risk of instability of stope is high under the stress ratio of K = 0.5 than that of K = 1.0 and K = 1.5 in both mining methods. However, the stability of the stope in open stope method is lower than that of cut-and-fill method obviously. This result shows that the appropriate mining method has to be selected for extraction of Sn-W deposit carefully in terms of the balance of safety and cost.
