Background:DNA methylation and gene expression are known to play important roles in the etiology of human diseases such as prostate cancer(PCa).However,it has not yet been possible to incorporate information of DNA methylation and gene expression into polygenic risk scores(PRSs).Here,we aimed to develop and validate an improved PRS for PCa risk by incorporating genetically predicted gene expression and DNA methylation,and other genomic information using an integrative ***:Using data from the PRACTICAL consortium,we derived multiple sets of genetic scores,including those based on available single-nucleotide polymorphisms through widely used methods of pruning and thresholding,LDpred,LDpred-funt,AnnoPred,and eBPRS,as well as PRS constructed using the genetically predicted gene expression and DNA methylation through a revised pruning and thresholding *** the tuning step,using the Uk Biobank data(1458 prevalent cases and 1467 controls),we selected PRSs with the best *** an independent set of data from the Uk Biobank,we developed an integrative PRS combining information from individual ***,in the testing step,we tested the performance of the integrative PRS in another independent set of Uk Biobank data of incident cases and ***:Our constructed PRS had improved performance(C statistics:76.1%)over PRSs constructed by individual benchmark methods(from 69.6%to 74.7%).Furthermore,our new PRS had much higher risk assessment power than family *** overall net reclassification improvement was 69.0%by adding PRS to the baseline model compared with 12.5%by adding family ***:We developed and validated a new PRS which may improve the utility in predicting the risk of developing *** innovative method can also be applied to other human diseases to improve risk prediction across multiple outcomes.

为了解汶川地震后滑坡体的植被恢复状况及驱动因子,于2012年8-9月在都江堰市四川龙溪–虹口国家级自然保护区对地震滑坡体上的植物物种组成、生长状况及环境因子进行了调查,共调查了127个10 m×10 m样方;并利用广义线性模型分析了影响滑坡体自然植被恢复的关键因子.结果显示:(1)滑坡体生长的维管植物有38科67属75种,以菊科(13种)和蔷薇科(7种)物种最为丰富,其余科的种类都较少;(2)物种生活型方面,草本植物种类最多,有47种,占总数的62.67%;其次为木本植物,有25种(包括灌木19种,小乔木6种);藤本植物仅3种;(3)滑坡体植被类型主要为灌丛及灌草丛,进一步可划分为10个群系,主要以灌丛为主,川莓(Rubus setchuenensis)和光滑悬钩子(***)为主要的建群种,在滑坡体上常构成单优势片层或混合生长形成共优种;(4)植被总盖度大于30%的样方占62.99%,最高达85%;植被平均高度超过1 m的样方占60.63%,最高达2.5 m;植物物种数超过5种的样方占84.25%,最多的有17种;(5)广义线性模型结果表明,坡度、样方位置、距林缘最近距离以及地表砾石盖度为影响滑坡体自然植被恢复的关键因子:坡度越大,灌木盖度越低,植物物种数也越少(B1);砾石盖度越高,植被总盖度、灌木盖度和植被平均高度都越低,植物物种数也越少(B0,P0,P1).研究表明,汶川地震后滑坡体上的植被大多都能够进行自然恢复,并逐渐向灌木和乔木林方向正向演替,但不同区域的恢复速度和建群种会存在差异;次生演替初期,小尺度范围内地形条件及地表基质状况的差异性对植被的恢复状况以及演替进程都有着显著的影响.

A new subclass of analytic functions that generalize some known subclasses of analytic functions was defined and investigated. This study was designed to investigate the molecular epidemiology of mobile colistin resistance(mcr)using a"One-Health"approach in Laos and to predict whether any dominant plasmid backbone and/or strain type influences the dissemination of *** collected 673 samples from humans(rectal normal flora),poultry,and the environment(water,flies,birds,etc.)in Vientiane,Lao People’s Democratic Republic(Laos),from May to September 2018.A total of 238 escherichia coli(***)isolated from nonduplicative samples,consisting of 98 MCR-positive ***(MCRPeC)("mcr"denotes the gene encoding mobile colistin resistance,and"MCR"denotes the subsequent protein encoded by mcr)and 140 MCRnegative ***(MCRNeC),were characterized by phenotype and Illumina sequencing.A subset of MCRPeC was selected for Min ION sequencing,conjugation assay,plasmid stability,and growth kinetics in *** prevalence of MCRPeC was found to be 14.6%(98/673),with the highest prevalence in human rectal swabs(45.9%(45/98),pence interval(CI):0.077-0.202).The percentages of MCRPeC from other samples were 14.3%(2/14)in dog feces,12.0%(24/200)in flies,11.0%(11/100)in chicken meat,8.9%(8/90)in chicken cloacal,8.0%(4/50)in chicken caeca,and 7.5%(4/53)in *** was significantly more resistant to co-amoxiclav,sulfamethoxazoletrimethoprim,levofloxacin,ciprofloxacin,and gentamicin than MCRNeC(penomic analysis revealed the distribution of MCRPeC among diverse clonal *** putative plasmid Inc types associated with mcr-1 were Inc X4,Inc HI2,Inc P1,Inc I2,and Inc FIA,and those associated with mcr-3 were Inc FII,Inc FIA,Inc FIB,Inc P1,and Inc *** of highly similar plasmids from both flies and other sampling sectors implied the role of flies in the dissemination of ***-positive plasmids were shown to be conjugative,and a significantly high transfer rate into a hypervirulent clone ST1193 was *** containing mcr irrespective of Inc type w

Immiscible metallic melts in the deep earth: clues from moissanite(SiC)in volcanic rocks

The occurrence of moissanite(SiC), as xenocrysts in mantle-derived basaltic and kimberlitic rocks sheds light on the interplay between carbon, hydrogen and oxygen in the lithospheric and sublithospheric mantle. SiC is stable only at fO2e the lithospheric mantle and related melts commonly are considered to be much more oxidized. SiC grains from both basaltic volcanoclastic rocks and kimberlites contain metallic inclusions whose shapes suggest they were entrapped as melts. The inclusions consist of Si^0+ Fe3Si7± FeSi2 Ti ± CaSi2Al2± FeSi2Al3± CaSi2, and some of the phases show euhedral shapes toward Si^0. Crystallographically-oriented cavities are common in SiC, suggesting the former presence of volatile phase(s), and the volatiles extracted from crushed SiC grains contain H2+ CH4± CO2± *** observations suggest that SiC crystalized from metallic melts(Si-Fe-Ti-C ± Al ± Ca), with dissolved H2+ CH4± CO2± CO derived from the sublithospheric mantle and concentrated around interfaces such as the lithosphere-asthenosphere and crust-mantle boundaries. When mafic/ultramafic magmas are continuously fluxed with H2+ CH4 they can be progressively reduced, to a point where silicide melts become immiscible, and crystallize phases such as SiC. The occurrence of SiC in explosive volcanic rocks from different tectonic settings indicates that the delivery of H2+ CH4 from depth may commonly accompany explosive volcanism and modify the redox condition of some lithospheric mantle volumes. The heterogeneity of redox states further influences geochemical reactions such as melting and geophysical properties such as seismic velocity and the viscosity of mantle rocks.

While stable isotopes of water have been used to study water movement through the environment, they generally have not been used to examine shorter, more transient events, e.g., rainfall of e development of robust methods that use isotope ratio infrared spectrometry, evaluating samples has become faster and simpler, allowing more soil and plant samples to be collected and analyzed. Using larger sampling rates can therefore increase the resolution of changes in stable isotopes within an ecosystem, and allows for a better understanding of how quickly rainwater that enters the soil by infiltration is transpired by a plant via root-water uptake. Quantifying rainwater uptake by plants is essential to increase crop production in rainfed agriculture. Thus the objective of this study was to measure the time required by a plant to transpire water from a source of water with a different isotopic signature than the water that the plant was irrigated. To this end, cotton (Gossypium hirsutum (L.)) plants were grown in a greenhouse and the time required for the enriched water added the soil to show up in the meristematic petioles of cotton leaves was measured. The initial divergence from the irrigation water signature occurred as quickly as 4 hours. The water from the sampled petioles then reached equilibrium with the new source water within 12 hours.
