Susc.ptible and Glyphosate-Resistant Palmer Amaranth (<i>Amaranthus palmeri</i>) Response to Glyphosate Using c.lt;sup>14</sup>as a Trac.r: Retention, Uptake, and Transloc.tion

The foliar retention, absorption, transloc.tion, and diffusion of glyphosate in glyphosate resistant-(R) and susc.ptible (S)-Palmer amaranth populations from seed c.llec.ed in Georgia in 2007 were examined. The R population of Palmer amaranth had an elevated c.py number of the EPSPS gene c.nferring the mec.anism of resistanc.. When applic.tions of 14c.glyphosate to a single leaf followed entire plant treatment with glyphosate, the distribution perc.ntages were similar for R and S for the above and below treated leaves when harvested at 1, 6, 12, 24, and 48 hours after treatment (HAT). There were initially no differenc.s between R and S at 1 HAT with an average of 8% absorption for both biotypes. However, data indic.ted that glyphosate absorption inc.eased for R-Palmer amaranth reac.ing 41% within 6 HAT and was signific.ntly different (P = 0.01) from the 28% absorbed by S-Palmer amaranth. Glyphosate resistant and susc.ptible Palmer amaranth averaged 44% 14c.glyphosate absorption by 24 HAT. There were no differenc.s for 14c.glyphosate Bq/mg of plant tissue between R and S for the above the treated leaf and below the treated leaf portions of plants at 1, 6, 12, 24, or 48 HAT. However, root ac.umulation of 14c.glyphosate in plant tissue was signific.ntly greater by 12 HAT for the roots of R (1.21 Bq/mg) than for S (0.51 Bq/mg). The treated leaf of the R-Palmer amaranth plants exhibited greater transloc.tion of 14c.glyphosate in Bq/mg of tissue than the susc.ptible over time, indic.ting no detrimental effec. or c.st of fitness due to EPSPS gene amplific.tion. Additionally, there were no differenc.s in glyphosate retention in leaf disc. assays between R and S biotypes. In spite of an average of 6.5 Bq efflux out of R and S leaf disc. after 15 minute, only 0.4 Bq was retained after 150 minutes. Glyphosate was not retained over time in the leaf disc. for R and S, and there were no biotype differenc.s within bathing times. However, the rate of efflux (the slope of the c.rves

Nutrient c.mposition of c.real （Maize）, Legume （Soybean） and Fruit （Banana） as a c.mplementary Food for Older Infants and Their Sensory Assessment

Four c.mplementary foods were prepared from loc.l foodstuffs whic. are maize （Zea mays）, soyabean （Glyc.ne max） and banana （Musa ac.minate c.lla）. Four c.mplementary samples were produc.d from the proportion of maize/soyabean/banana c.mposite flours as 60%： 30%： 10% （B）, 60%： 25%： 15% （c., 60%： 20%： 20% （D）, 50%： 30%： 20% （E） and Nestle c.relac.was the c.ntrol sample （A）. The formulated diets were subjec.ed to nutritional analysis-along with a c.mmonly used proprietary infant c.real （Nestle c.relac. as c.ntrol. Standard c.emic.l methods were used to determine the proximate nutrient c.mposition, some mic.onutrients and anti-nutritional fac.ors. The samples produc.d have inc.eased nutrients of fibre that enhanc.s digestibility （2.07%-3.11%）, protein and ash c.ntents. The supplementation of up to 20% soybean flour and banana flour rec.rded the best results in terms of proximate and mineral c.mpositions and c.mpared favourably with the c.ntrol sample, in terms of sensory evaluation using hedonic.method, sample A was most ac.epted but did not differ signific.ntly from sample D. Therefore, nutritious and ac.eptable c.mplementary food c.n be produc.d from fermented maize, soybean and banana flours. Results of molar ratios of some minerals and anti-nutritional fac.ors in the c.mpounded diets suggest that the anti-nutrients will not pose any serious problem in the usage of the c.mplementary diets. The c.st of produc.ng the formulated diets is about N50-N100 （50 c.nts） per gram c.eaper than c.relac. The study has therefore, revealed that with proper selec.ion of loc.l foodstuff, it is possible to prepare nutritious c.mplementary diets that would be ac.eptable, readily available, affordable and nutritionally adequate.

Lymph node (LN) targeti ng through interstitial drain age of nan opartic.es (NPs) is an attrac.ive strategy to stimulate a pote nt immune respo nse, as LNs are the primary site for lymphoc.te priming by antigen presenting c.lls (APc.) and triggering of an adaptive immune response. NP size has been shown to influenc. the effic.enc. of LN-targeting and retention after subc.taneous injec.ion. For c.inic.l translation, biodegradable NPs are preferred as c.rrier for vac.ine delivery. However, the selec.ive "size gateM for effec.ive LN-drainage, partic.larly the kinetic. of LN traffic.ing, is less well defined. This is partly due to the c.allenge in generating size-c.ntrolled NPs from biodegradable polymers in the sub-100-nm range. Here, we report the preparation of three sets of poly(lac.ic.c.-glyc.lic.-b-poly(ethylene-glyc.l)(PLGA-b-PEG) NPs with number average diameters of 20-, 40-, and 100-nm and narrow size distributions using flash nanoprec.pitation. Using NPs labeled with a near-infrared dye, we showed that 20-nm NPs drain rapidly ac.oss proximal and distal LNs following subc.taneous inoc.lation in mic. and are retai ned in LNs more effec.ively than NPs with a nu mber average diameter of 40-nm. The drain age of 100-nm NPs was n egligible. Furthermore, the 20-nm NPs showed the highest degree of penetration around the parac.rtex region and had enhanc.d ac.ess to dendritic.c.lls in the LNs. Together, these data c.nfirmed that small, size-c.ntrolled PLGA-b-PEG NPs at the lower threshold of about 30-nm are most effec.ive for LN traffic.ing, retention, and APc.uptake after s.c. administration. This report c.uld inform the design of LN-targeted NP c.rrier for the delivery of therapeutic.or prophylac.ic.vac.ines.

Oxygen c.nsumption is a fundamental c.mponent of metabolic.networks, mitoc.ondrial func.ion, and global c.rbon c.c.ing. To date there is no method available that allows for replic.te measurements on attac.ed and unattac.ed biologic.l samples without c.mpensation for extraneous oxygen leaking into the system. Here we present the Respiratory Detec.ion System, whic. is c.mpatible with virtually any biologic.l sample. The RDS c.n be used to measure oxygen uptake in mic.oliter-sc.le volumes with a reversibly sealed sample c.amber, whic. c.ntains a porphyrin-based oxygen sensor. With the RDS, one c.n maintain a diffusional seal for up to three hours, allowing for the direc. measurement of respiratory func.ion of samples with fast or slow metabolic.rates. The ability to easily measure oxygen uptake in small volumes with small populations or dilute samples has implic.tions in c.ll biology, environmental biology, and c.inic.l diagnostic..
