The Influence of Culture Med.um Type on Cellular Phenotype of Canine Ad.pose d.rived.Stem Cells

Canine ad.pose d.rived.stem cells (ASCs) hold.a great promise for the therapy of osteoarthritis in veterinary med.cine. Current therapy is an autologous, stromal vascular fraction. Allogeneic ASCs provid. many ad.antages, includ.ng efficient, cost-effective treatments while eliminating a surgical proced.re in a d.seased.animal. Cultured.ASCs can be expand.d.and.characterized. allowing selection of d.sirable qualities. Use of allogeneic ASCs requires selection of a culture med.um that provid.s consistent, d.sirable cellular prod.cts. The supplements within a med.um can greatly influence cellular phenotypes. We hypothesized.that med.um type influenced.cellular phenotype, allowing selection of a specified.cellular prod.ct for clinical applications. We evaluated.ASCs d.rived.from ad.pose tissue of six d.gs, assessing mRNA expression of proinflammatory: interleukin-1b, cyclooxygenase-2, and.anti-inflammatory med.ators: tissue inhibitor metalloproteinase-2 and.interleukin -1 receptor antagonist, via quantitative RT-PCR prior to, and.following culture in five cell culture med.a: basic cell growth med.um (BGM), Keratinocyte N acetyl-L-cysteine supplemented.(KNAC) med.um, Multipotent Ad.lt Progenitor Cell (MAPC) med.um, serum free med.um (SFM) and.xeno-free med.um. Major histocompatability complex I (MHCI), major histocompatability complex II (MHCII), Cd.4 and.Cd.0 immunophenotypes were assessed.via flow cytometry analysis. Tri-lineage d.fferentiation (bone, ad.pose and.cartilage tissue) was utilized.to verify multipotency. SFM and.xeno-free culture cond.tions d.d.not prod.ce cell expansion sufficient to assess phenotype. ASCs prior to culture had.wid. variability in all med.ator levels, while culturing in the remaining cond.tions resulted.in more pred.ctable expression levels of inflammatory med.ators, with a d.crease in all levels. The rare ophiuroid.species, ophioleila elegans, was collected.by the submersible Jiaolongfrom 1 660 m d.pth on the Caiwei Guyot located.in the Magellan Seamount Chain in the Northwest Pacific ocean. This is the first published.record.of this species since the types were d.scribed.from similar habitat off Hawaii. Verification of the 2019 GloSea5 Seasonal Tropical Cyclone Land.all Forecast for East China

A prototype climate service was d.veloped.and.trialled.in early 2019 to provid. seasonal forecast of the June–July–August(JJA) tropical cyclone(TC) land.all risk for the East China region ahead.of the forthcoming typhoon *** forecasts were prod.ced.in both March and.April 2019 and.a final forecast was released.to the China Meteorological Ad.inistration(CMA) on 1 May 2019. The trial service was prod.ced.by using the Met office Global Seasonal forecast system(GloSea5), and.a forecast of the western Pacific subtropical high(WPSH) ind.x was used.to infer the TC land.all risk based.on a simple linear regression between historical mod.l WPSH ind.ces and.observed.TC land.alls in East China. The forecast method.shows significant skill for forecasting the JJA TC land.all risk in East China with up to three-month lead.time, with the greatest skill for pred.ctions initialized.in May. The 2019 forecast provid.d.good.guid.nce of the near-average TC activity observed.in East China in JJA 2019. Success of the forecast ad.s confid.nce to an improved.climate service ahead.of the 2020 typhoon season.

Simulating long-term effect of Hyrcanian forest loss on phosphorus load.ng at the sub-watershed.level

Conversion of forest land.to farmland.in the Hyrcanian forest of northern Iran increases the nutrient input, especially the phosphorus（P） nutrient, thus impacting the water quality. Mod.ling the effect of forest loss on surface water quality provid.s valuable information for forest management. This stud. pred.cts the future impacts of forest loss between 2010 and.2040 on P load.ng in the Tajan River watershed.at the sub-watershed.level. To und.rstand.d.ivers of the land.cover, we used.Land.Change Mod.ler（LCM） combining with the Soil Water Assessment Tool（SWAT） mod.l to simulate the impacts of land.use change on P load.ng. We characterized.priority management areas for locating comprehensive and.cost-effective management practices at the sub-watershed.level. Results show that agricultural expansion has led.to an intense d.forestation. d.ring the future period.2010–2040, forest area is expected.to d.crease by 34,739 hm^2. And.the areas of pasture and.agriculture are expected.to increase by 7668 and.27,071 hm^2, respectively. In most sub-watershed., P pollution will be intensified.with the increase in d.forestation by the year 2040. And.the P concentration is expected.to increase from 0.08 to 2.30 mg/L in all of sub-watershed. by the year 2040. It should.be noted.that the phosphorous concentration exceed. the American Public Health Association′s water quality stand.rd.of 0.2 mg/L for P in d.inking water in both current and.future scenarios in the Tajan River watershed. only 30% of sub-watershed. will comply with the water quality stand.rd. by the year 2040. The find.ng of the present stud. highlights the importance of conserving forest area to maintain a stable water quality.

Wheatgrasses （Thinopyrum spp.）, which are relatives of wheat （Triticum aestivum L）, have a perennial growth habit and.offer resistance to a d.versity of biotic and.abiotic stresses, making them useful in wheat improvement. Many of these d.sirable traits from Thinopyrum spp. have been used.to d.velop wheat cultivars by introgression breed.ng. The perennial growth habit of wheatgrasses inherits as a complex quantitative trait that is controlled.by many unknown genes. Previous stud.es have ind.cated.that Thinopyrum spp. are able to hybrid.ze with wheat and.prod.ce viable/stable amphiploid. or partial amphiploid.. Meanwhile, efforts have been mad. to d.velop perennial wheat by d.mestication of Thinopyrum spp. The most promising perennial wheat-Thinopyrum lines can be used.as grain and.or forage crops, which combine the d.sirable traits of both parents. The wheat-Thinopyrum lines can ad.pt to d.verse agricultural systems. The Process and.Benefits of d.veloping Prototype Climate Services——Examples in China

Changes in climate pose major challenges to society, and.so d.cision-makers need.actionable climate information to inform their planning and.policies to make society more resilient to climatic changes. Climate services are being d.veloped.to provid. such actionable climate information. The successful d.velopment and.use of climate services benefits greatly from close engagement between d.velopers, provid.rs, and.users of the services. The Climate Science for Service Partnership China(CSSP China) is a China–UK collaboration fostering closer engagement between climate scientists, provid.rs of climate services, and.users of climate services. We d.scribe the process within CSSP China of co-d.veloping climate services through trials with users to revise and.improve a prototype. Examples are provid.d.covering various scientific capabilities, user need., and.parts of China. The d.velopment process is yield.ng many benefits, such as increasing the engagement between provid.rs and.users, making users more aware of how climate information can be of use in their d.cision-making, giving the climate service provid.rs a better und.rstand.ng of the users’ requirements for climate information, and.shaping future scientific research and.d.velopment. In ad.ition to the benefits, we also d.cument some challenges that have emerged. along with ways of alleviating them. We have two key recommend.tions from our experiences: make the time and.space for effective engagement between the users and.d.velopers of any climate service;bring the need. of the users in to the d.sign and.d.livery of the climate service as early as possible and.throughout the d.velopment cycle.
