Identification of CD13,CD107a,and CD164 as novel basophil-activation markers and dissection of two response patterns in time kinetics of IgE-de-pendent upregulation

Using two-colour flow cytometry >200 antibodies submitted to the 8th International Workshop of Human Leukocyte Differentiation Antigens (HLDA8) have been analyzed for their reactivity with resting and activated CD203c+ basophils. Four antibodies either non-reactive or weakly reactive with resting basophils exhibited an increased reactivity with basophils activated by anti-IgE-mediated cross-linking of the high affinity IgE receptor (FcεRI). These include antibod- ies against CD164 (WS-80160, clone N6B6 and WS-80162, clone 67D2), as well as two reagents with previously unknown specificities that were identified as CD13 (WS-80274, clone A8) and CD107a (WS-80280, clone E63-880). The activation patterns followed either the “CD203c-like” or “CD63-like” activation profile. The CD203c profile is characterized by a rapid and significant upregulation (of CD13, CD164, and CD203c), reaching maximum levels after 5- 15 min of stimulation. The phosphoinositide-3-kinase (PI3K)-specific inhibitor wortmannin inhibited the upregulation of these markers whereas 12-O-tetradecanoyl-phorbol-13-acetate (TPA) induced a rapid and FcεRI-independent upregulation within 1-2 min. In the CD63 profile, maximum upregulation (of CD63 and CD107a) was detected only after 20-40 min, and upregulation by TPA reached maximum levels after 60 min. In summary, our data identify CD13, CD107a, and CD164 as novel basophil-activation antigens. Based on time kinetics of upregulation, we hypothesize that molecules of the “CD203c group” and the “CD63 group” are linked to two different mechanisms of basophil activation.

DNA studies of endangered or extinct species often rely on ancient or degraded *** majority of ancient DNA(aDNA)extraction protocols focus on skeletal elements,with skin and hair samples rarely *** to that found in bones and teeth,DNA extracted from historical or ancient skin and fur samples is also extremely fragmented with low endogenous content due to natural degradation ***,the development of effective DNA extraction methods is required for these ***,we compared the performance of two DNA extraction protocols(commercial and custom laboratory aDNA methods)on hair and skin samples from decades-old museum specimens to Iron Age archaeological *** found that apart from the impact sample-specific taphonomic and handling history has on the quantity and quality of DNA preservation,skin yielded more endogenous DNA than hair of the samples and protocols *** both methods recovered DNA from ancient soft tissue,the laboratory method performed better overall in terms of DNA yield and quality,which was primarily due to the poorer performance of the commercial binding buffer in recovering aDNA.

Trachypithecus,which currently contains 20 species divided into four groups,is the most speciose and geographically dispersed genus among Asian *** several morphological and molecular studies,however,its evolutionary history and phylogeography remain poorly ***’s langur(Trachypithecus phayrei) is one of the most widespread members of the genus,but details on its actual distribution and intraspecific taxonomy are limited and ***,to elucidate the evolutionary history of Trachypithecus and to clarify the intraspecific taxonomy and distribution of ***,we sequenced 41 mitochondrial genomes from georeferenced fecal samples and museum specimens,including two *** analyses revealed a robustly supported phylogeny of Trachypithecus,suggesting that the *** group branched first,followed by the *** group,and the *** and *** groups most *** four species groups diverged from each other 4.5-3.1 million years ago(Ma),while speciation events within these groups occurred much more recently(1.6-0.3 Ma).Within ***,we found three clades that diverged 1.0-0.9 Ma,indicating the existence of three rather than two *** the phylogenetic species concept and based on genetic,morphological,and ecological differences,we elevate the *** subspecies to species level,describe a new species from central Myanmar,and refine the distribution of the three ***,our study highlights the importance of museum specimens and provides new insights not only into the evolutionary history of *** but the entire Trachypithecus genus as well.

Over the last six years many experiments have been done at the National Ignition Facility to measure the Hugoniot of materials,such asCHplastic at extreme pressures,up to 800 ***“Gbar”design employs a strong spherically converging shock launched through a solid ball of material using a hohlraum radiation *** shock front conditions are characterized using x-ray *** this paper we examine the role of radiation in heating the unshocked material in front of the shock to understand the impact it has on equation of state measurements and how it drives the measured data off the theoretical Hugoniot *** particular,the two main sources of radiation heating are the preheating of the unshocked material by the high-energy kilo-electron-volt x-rays in the hohlraum and the heating of the material in front of the shock,as the shocked material becomes hot enough to radiate *** our model,we estimate that preheating can reach 4 eV in unshocked material,and that radiation heating can begin to drive data off the Hugoniot significantly,as pressures reach above 400 Mb.
