Objective:To observe the effects of different hyperbaric oxygen treatment time windows on the prognosis and neurological function of acute cerebral infarction.Method:160 patients with acute cerebral infarction admitted to Xiangyang Central Hospital in Hubei Province were randomly divided into four groups,each with 40 cases,using a random number table method.According to the 2017 guidelines for the treatment of cerebral infarction,the control group received routine treatment for acute cerebral infarction;On the basis of the control group,patients in Group A received hyperbaric oxygen therapy within 48 hours of onset;Group B patients receive hyperbaric oxygen therapy within 3-6 days of onset;Group C patients receive hyperbaric oxygen therapy within 7-12 days of onset.Observe the efficacy,recurrence,and neurological function recovery of four groups of patients after treatment.Result:There was no statistically significant difference in the National Institutes of Health Stroke Scale(NIHSS)and Barthel Index(BI)scores among the four groups before treatment(P>0.05).There were statistically significant differences in NIHSS and BI scores between 14 and 30 days after treatment and before treatment(F=16.352,27.261,11.899,28.326,P0.05).After 30 days of treatment,the total effective rate of Group A was higher than that of the control group and Group C,and the difference was statistically significant(X2=6.135,P<0.05).The one-year recurrence rate of Group A and Group B is lower than that of Group C and the control group,and the difference is statistically significant(X2=8.331,P<0.05).There was no statistically significant difference in adverse reactions among th

在电力系统和天然气系统紧密耦合的背景下,任意子系统中任意脆弱线路的扰动或故障都可能会传播到另一个子系统,进而引起整个电-气互联系统的故障。为此,提出一种电-气互联系统脆弱线路辨识方法。首先,提出考虑风光出力不确定性的电-气互联系统协调调度随机优化模型,求解得到电-气互联系统故障前后的运行状态;而后,基于局部集中度和潮流变化熵分别从拓扑结构脆弱性和运行特性脆弱性两方面分析电-气互联系统中线路的脆弱性,构建综合考虑线路拓扑结构脆弱性和运行特性脆弱性的电-气互联系统线路脆弱度指标,对互联系统的脆弱线路进行辨识。通过计算脆弱线路断开后互联系统的测地线脆弱度(geodesic vulnerability)及供需不平衡量,对所辨识的脆弱线路进行评估。6节点电网-7节点气网和24节点电网-20节点气网2个电-气互联系统的算例分析结果验证了所提脆弱线路辨识方法的有效性。

Background Chinese indigenous pigs are popular with consumers for their juiciness,flavour and meat quality,but they have lower meat production.Insulin-like growth factor 2(IGF2) is a maternally imprinted growth factor that promotes skeletal muscle growth by regulating cell proliferation and differentiation.A single nucleotide polymorphism(SNP) within intron 3 of porcine IGF2 disrupts a binding site for the repressor,zinc finger BED-type containing 6(ZBED6),leading to up-regulation of IGF2 and causing major effects on muscle growth,heart size,and backfat thickness.This favorable mutation is common in Western commercial pig populations,but absent in most Chinese indigenous pig breeds.To improve meat production of Chinese indigenous pigs,we used cytosine base editor 3(CBE3)to introduce IGF2 intron3-C3071T mutation into porcine embryonic fibroblasts(PEFs) isolated from a male Liang Guang Small Spotted pig(LGSS),and single-cell clones harboring the desired mutation were selected for somatic cell nuclear transfer(SCNT) to generate the founder line of IGF2^(T/T) pigs.Results We found the heterozygous progeny IGF2^(C/T) pigs exhibited enhanced expression of IGF2,increased lean meat by 18%-36%,enlarged loin muscle area by 3%-17%,improved intramuscular fat(IMF) content by 18%-39%,marbling score by 0.75-1,meat color score by 0.53-1.25,and reduced backfat thickness by 5%-16%.The enhanced accumulation of intramuscular fat in IGF2^(C/T) pigs was identified to be regulated by the PI3K-AKT/AMPK pathway,which activated SREBP1 to promote adipogenesis.Conclusions We demonstrated the introduction of IGF2-intron3-C3071T in Chinese LGSS can improve both meat production and quality,and first identified the regulation of IMF deposition by IGF2 through SREBP1 via the PI3KAKT/AMPK signaling pathways.Our study provides a further understanding of the biological functions of IGF2and an example for improving porcine economic traits through precise base editing.

In this paper,a novel power grid evolving model,which can well describe the evolving property of power grids,is presented. Based on the BA model,motivated by the fact that in real power grids,connectivity of node not only depends on its degree,but also is influenced by many uncertain factors,so we introduce the subconnection factor K for each node. Using the mean-field theory,we get the analytical expression of power-law degree distribution with the exponent γ∈ (3,∞ ). Finally,simulation results show that the new model can provide a satisfactory description for empirical characteristics of power network,and power network falls somewhere in between scale-free network and uncertain network.

A lasting imbalance between fatty acid synthesis and consumption leads to non-alcoholic fatty liver disease(NAFLD),coupled with hepatitis and insulin resistance.Yet the details of the underlying mechanisms are not fully understood.Here,we unraveled that the expression of the transcription factor Zbtb18 is markedly decreased in the livers of both patients and murine models of NAFLD.Hepatic Zbtb18 knockout promoted NAFLD features like impaired energy expenditure and fatty acid oxidation(FAO),and induced insulin resistance.Conversely,hepatic Zbtb18 overexpression alleviated hepato-steatosis,insulin resistance,and hyperglycemia in mice fed on a high-fat diet(HFD)or in diabetic mice.Notably,in vitro and in vivo mechanistic studies revealed that Zbtb18 transcriptional activation of Farnesoid X receptor(FXR)mediated FAO and Clathrin Heavy Chain(CLTC)protein hinders NLRP3 inflammasome activity.This key mechanism by which hepatocyte’s Zbtb18 expression alleviates NAFLD and consequent liver fibrosis was further verified by FXR’s deletion and forced expression in mice and cultured mouse primary hepatocytes(MPHs).Moreover,CLTC deletion significantly abrogated the hepatic Zbtb18 overexpression-driven inhibition of NLRP3 inflammasome activity in macrophages.Altogether,Zbtb18 transcriptionally activates the FXR-mediated FAO and CLTC expression,which inhibits NLRP3 inflammasome’s activity alleviating inflammatory stress and insulin resistance,representing an attractive remedy for hepatic steatosis and fibrosis.

采用烟毒性测试箱研究了铁氟龙AFT250航空电线燃烧后生成的毒害气体HF、CO、CO_(2)浓度的变化特性。结果表明:在点火温度为1150℃、点火持续时间10 s条件下,铁氟龙AFT250电线燃烧生成HF有毒气体质量浓度为8 mg/m^(3),生成CO和CO_(2)气体体积浓度分别为0.0015%和0.12%,说明在该条件下已生成一定量的毒害气体,但毒害气体浓度并不大。随截取电线长度在10~100 cm内增加,生成有毒有害气体HF、CO、CO_(2)的浓度均不断增大。当电线长度为100 cm时,HF质量浓度增至18 mg/m^(3),CO体积浓度增至0.004%,CO_(2)体积浓度增至0.425%,这主要由于增加电线长度会增加绝缘层材料聚四氟乙烯中F、C元素含量,导致更多的F、C原子和环境中的H、O原子结合。随点火持续时间在10~50 s内延长,燃烧生成的毒害气体HF、CO、CO_(2)浓度均显著增大,HF气体质量浓度增幅为7 mg/m^(3),CO和CO_(2)体积浓度增幅分别为0.0015%和0.07%,这主要是由于点火持续时间越长,会导致更多的C-C和C-F化学键断裂,从而增大生成的HF、CO和CO_(2)浓度,极大地增加人体中毒与窒息的风险。

目的探讨子宫内膜癌不同技术方法分子分型的差异以探索符合国情的分子分型方案。方法收集复旦大学附属妇产科医院病理科2021年4月至2022年3月诊断的子宫内膜癌254例,采用二代测序和免疫组织化学检测综合方案进行分子分型,并以POLE和TP53突变频率≥20%设定为Sanger测序检测极限值模拟其结果,比较不同组合方案下分子分型结果。结果254例子宫内膜癌患者,年龄24~89岁,平均年龄51岁。包含有POLE的第9~14号外显子、TP53全外显子和微卫星不稳定(MSI)检测的二代测序panel为较好的独立解决方案,结合MSI片段分析检测和常规免疫组织化学初筛为最佳的综合解决方案,和癌症基因组图谱(TCGA)数据及近期文献比对有最佳符合率,POLE超突变型、错配修复缺陷型、TP53突变型及非特异分子特征型占比分别为11.4%(29/254)、31.5%(80/254)、22.4%(57/254)和34.6%(88/254);如改用Sanger测序进行POLE和TP53突变检测,则分别为9.1%(23/254)、31.5%(80/254)、12.9%(33/254)和46.6%(118/254),大幅度增加了非特异分子特征型的数量;如采用Sanger测序检测POLE,其余分型均用免疫组织化学检测替代,则分别为9.1%(23/254)、42.2%(92/218)、13.8%(35/254)和40.9%(105/254),将增加错配修复缺陷组的假阳性判断。结论包含MSI检测的中小型二代测序panel为分子分型的较好解决方案,Sanger测序对POLE突变检测目前可满足基本要求,但对TP53突变检测灵敏度不够,和p53免疫组织化学判断TP53突变效率相当。进一步优化含MSI检测和POLE、TP53全外显子的中小型二代测序方案是未来符合国情的最优选择。

自然灾害是一种发生概率低但破坏力强的极端事件,发生时会给气电联合配网系统中造成严重影响。为提高气电联合配网系统在自然灾害下的韧性,考虑线路/管道加固、应急资源配置与气电联合需求响应,该文提出基于鲁棒优化的韧性提升策略。首先,在气电联合配网系统中考虑到线路/管道加固后在面对严重自然灾害时可能失效的情形,提出一种表示不同灾害程度与加固程度的多等级加固模型,并建立相应的线路/管道状态约束。然后,在气电联合配网系统运行约束基础上,建立气电应急资源的优化配置模型,引入气电联合需求响应机制,提出以气电联合配网系统中加权失负荷最小为目标的三层防御-攻击-防御韧性提升模型,并采用列与约束生成算法求解。最后,在算例中证明了模型的可行性和求解方法的有效性。

近年来,在未来多能互补、综合能源系统的背景下,气电联合配网得到迅速的发展。然而,当前全球各地极端灾害频发,给能源系统的安全性带来了严峻的挑战。针对极端灾害对系统造成的失负荷影响,为兼顾系统的经济性与安全性,提出了一种考虑极端灾害下系统韧性约束的气电联合配网分布鲁棒扩展规划模型。首先,以规划成本与运行成本最小化为目标,建立考虑电动汽车充电站、分布式燃气机组和储能设备的气电联合配网扩展规划模型;然后,考虑极端灾害的作用,建立考虑韧性约束的气电联合配网分布鲁棒扩展规划模型,该模型包括基础场景与极端灾害最坏场景,其中基础场景考虑系统的经济性,以规划成本与基础场景运行成本最小为目标,极端灾害最坏场景则通过韧性约束保证系统的安全性,所提模型通过主问题–子问题迭代求解。最后,算例仿真发现:通过增加电动汽车充电站、分布式燃气机组和储能设备的规划投建,系统的经济性得到明显提升;而在面对极端灾害的影响下,通过分布式燃气机组的发电和电动汽车、储能设备的放电,系统失负荷情况得到减少,但随着系统韧性的增大所需单位规划成本会大幅增加。本文为在规划层面提升系统韧性提供了一种有效与实用的方法,为未来减小极端灾害对系统造成的影响的方案提供了一定参考。
