Patient-derived cancer cells(PDCs)and patient-derived xenografts(PDXs)are often used as tumor models,but have many *** not only lack diversity in terms of cell type,spatial organization,and microenvironment but also have adverse effects in stem cell cultures,whereas PDX are expensive with a low transplantation success rate and require a long culture *** recent years,advances in three-dimensional(3D)organoid culture technology have led to the development of novel physiological systems that model the tissues of origin more precisely than traditional culture ***-derived cancer organoids bridge the conventional gaps in PDC and PDX models and closely reflect the pathophysiological features of natural tumorigenesis and metastasis,and have led to new patient-specific drug screening techniques,development of individualized treatment regimens,and discovery of prognostic biomarkers and mechanisms of *** combinations of cancer organoids with other technologies,for example,organ-on-a-chip,3D bio-printing,and CRISPR-Cas9-mediated homology-independent organoid transgenesis,and with treatments, such as immunotherapy, have been useful in overcoming their limitations and led to the development of more suitable model systems that recapitulate the complex stroma of cancer, inter-organ and intra-organ communications,and potentially multiorgan metastasis. In this review, we discuss various methods for the creation of organ-specific cancer organoids and summarize organspecific advances and applications, synergistic technologies, and treatments aswell as current limitations and future prospects for cancer organoids. Furtheradvances will bring this novel 3D organoid culture technique closer to clinicalpractice in the future.

目的探讨miR-155在肺腺癌侵袭和转移中的作用。方法采用实时荧光定量PCR和原位杂交检测miR-155在肺腺癌和癌旁组织以及转移淋巴结中的表达，采用划痕实验和Transwell迁移实验检测miR-155对A549细胞的迁移和侵袭能力的影响；利用生物信息学软件预测miR-155的靶基因，并采用双荧光素酶检测靶基因；采用Westernblot和实时荧光定量PCR检测miR-155调控靶基因鼠抗人第10号染色体同源缺失性磷酸酶张力蛋白（PTEN）的表达水平。结果实时荧光定量PCR检测显示，肺腺癌组织、癌旁正常组织和转移淋巴结中的miR-155表达水平分别为9．6±3．1、4．1±0．5和7．8±2．2。原位杂交检测显示，肺腺癌组织、癌旁正常组织和转移淋巴结中miR-155阳性表达率分别为（75．4±20．2）％、（23．2±15．3）％和（60．4±25．1）％。划痕实验检测显示，miR-155模拟物组、miR-155模拟物对照组、miR-155抑制剂组和miR-155抑制剂对照组24h伤口愈合率分别为（43．2±2．2）％、（21．3±4．2）％、（24．3±5．3）％和（35．2±5．1）％，而48h伤口愈合率分别为（75．2±4．5）％、（52．6±5．2）％、（39．4±4．2）％和（51．5±4．3）％。双荧光素酶报告基因检测显示，miR-155模拟物组与含有PTEN3’UTR野生型和突变型质粒共转染的荧光素酶值分别为4．7±0．5和7．3±0．7，而miR-155模拟物对照组与含有PTEN3’UTR野生型和突变型质粒共转染的荧光素酶值分别为7．8±0．9和7．5±0．8。实时荧光定量PCR检测显示，miR-155模拟物组、miR-155模拟物对照组、miR-155抑制剂组和miR-155抑制剂对照组的PTENmRNA相对表达水平分别为0．5±0．3、1．0±0．1、2．2±0．2和1．2±0．1。Westernblot检测显示，miR-155模拟物组、miR-155模拟物对照组、miR-155抑制剂组和miR-155抑制剂对照组的PTEN蛋白相对表达水平分别为0．4±0．1、1．0±0．3、2．8±0．2和1．4±0．1。miR-155模拟物组与miR-155模拟物对照组、miR-155抑制剂组与miR-155抑制剂对照组的PTENmRNA和蛋白表达差异均有统计学意义（均P〈0．05）。结论miR-155可能是通过下调靶基因PTEN的表达而促进肺腺癌的侵袭和转移。

Hydroclimatic variations over the eastern Central Asia are highly sensitive to changes in hemispheric-scale atmospheric circulation systems. To fully understand the long-term variability and relationship between hydroclimate and atmospheric circulation system, we present a high-resolution lascustrine record of late Holocene hydroclimate from Lake Sayram, Central Tianshan Mountains, China, based on the total organic carbon, total nitrogen, and carbonate contents, carbon/nitrogen ratios, and grain size. Our results reveal four periods of substantially increased precipitation at the interval of 4000–3780, 3590–3210, 2800–2160, and 890–280 cal yr BP, and one period of slightly increased precipitation from 1700–1370 cal yr BP. These wetter periods broadly coincide with those identified in other records from the mid-latitude Westerlies-dominated eastern Central Asia, including the northern Tibetan Plateau. As such, a similar hydroclimatic pattern existed over this entire region during the late Holocene. Based on a close similarity of our record with reconstruction of North Atlantic Oscillation indices and solar irradiance, we propose that decreased solar irradiance and southern migration of the entire circum-North Atlantic circulation system, particularly the main pathway of the mid-latitude Westerlies, significantly influenced hydroclimate in eastern Central Asia during the late Holocene. Finally, the inferred precipitation at Lake Sayram has increased markedly over the past 100 years, although this potential future changes in hydroclimate in Central Asia need for further investigation.

为解决施肥不科学影响‘凤丹’牡丹品质产量提升的问题,以5年生‘凤丹’牡丹为试验材料,采用分阶段控量施肥的方法,研究其对‘凤丹’牡丹光合特性、产量的影响。结果表明,‘凤丹’叶片的净光合速率、气孔导度、蒸腾速率、叶绿素含量均随着复合肥施入量的增加、喷施硼肥浓度的增加呈先上升后下降的趋势,在D3B4(复合肥225 kg/hm^(2),硼肥0.3%)水平时达到最大值。叶片胞间CO_(2)浓度趋势相反,呈先降后升的变化趋势,在D3B4水平达到最低值。同时,在D3B4水平时,‘凤丹’的单株果荚数、果荚直径、千粒重、单株籽粒产量均达到最大值。本研究以盛花期复合肥底肥225 kg/hm^(2)+结果初期叶喷0.3%硼肥为‘凤丹’牡丹高产的最佳施肥模式,可为‘凤丹’牡丹科学施肥提供一定参考依据。

We report the investigation of the thermoelectric properties of large-scale solution-synthesized Bi2Te3 nanocomposites prepared from nanowires hot- pressed into bulk pellets. A third element, Se, is introduced to tune the carrier concentration of the nanocomposites. Due to the Se doping, the thermoelectric figure of merit （ZT） of the nanocomposites is significantly enhanced due to the increased power factor and reduced thermal conductivity. We also find that thermal transport in our hot-pressed pellets is anisotropic, which results in different thermal conductivities along the in-plane and cross-plane directions. Theoretical calculations for both electronic and thermal transport are carried out to establish fundamental understanding of the material system and provide directions for further ZT optimization with adjustments to carrier concentration and mobility.
