Performance and Application of Double-layered Microcapsule Corrosion Inhibitors

Double-layered microcapsule corrosion inhibitors were developed by sodium monofluorophosphate as the core material,polymethyl methacrylate as the inner wall material,and polyvinyl alcohol as the outer wall material combining the solvent evaporation method and spray drying method.The protection by the outer capsule wall was used to prolong the service life of the corrosion inhibitor.The dispersion,encapsulation,thermal stability of microcapsules,and the degradation rate of capsule wall in concrete pore solution were analyzed by ultra-deep field microscopy,scanning electron microscopy,thermal analyzer,and sodium ion release rate analysis.The microcapsules were incorporated into mortar samples containing steel reinforcement,and the effects of double-layered microcapsule corrosion inhibitors on the performance of the cement matrix and the actual corrosion-inhibiting effect were analyzed.The experimental results show that the double-layered microcapsules have a moderate particle size and uniform distribution,and the capsules were completely wrapped.The microcapsules as a whole have good thermal stability below 230 ℃.The monolayer membrane structure microcapsules completely broke within 1 day in the simulated concrete pore solution,and the double-layer membrane structure prolonged the service life of the microcapsules to 80 days in the simulated concrete pore solution before the core material was completely released.The mortar samples containing steel reinforcement incorporated with the double-layered microcapsule corrosion inhibitors still maintained a higher corrosion potential than the monolayer microcapsule corrosion inhibitors control group at 60 days.The incorporation of double-layered microcapsules into the cement matrix has no significant adverse effect on the setting time and early strength.

Uncertainty-aware complementary label queries for active learning

Many active learning methods assume that a learner can simply ask for the full annotations of some training data from annotators.These methods mainly try to cut the annotation costs by minimizing the number of annotation actions.Unfortunately,annotating instances exactly in many realworld classification tasks is still expensive.To reduce the cost of a single annotation action,we try to tackle a novel active learning setting,named active learning with complementary labels(ALCL).ALCL learners ask only yes/no questions in some classes.After receiving answers from annotators,ALCL learners obtain a few supervised instances and more training instances with complementary labels,which specify only one of the classes to which the pattern does not belong.There are two challenging issues in ALCL:one is how to sample instances to be queried,and the other is how to learn from these complementary labels and ordinary accurate labels.For the first issue,we propose an uncertainty-based sampling strategy under this novel setup.

Recent advances in“universal”influenza virus antibodies:the rise of a hidden trimeric interface in hemagglutinin globular head

Influenza causes seasonal outbreaks yearly and unpredictable pandemics with high morbidity and mortality rates.Despite significant efforts to address influenza,it remains a major threat to human public health.This issue is partially due to the lack of antiviral drugs with potent antiviral activity and broad reactivity against all influenza virus strains and the rapid emergence of drug-resistant variants.Moreover,designing a universal influenza vaccine that is sufficiently immunogenic to induce universal antibodies is difficult.Some novel epitopes hidden in the hemagglutinin(HA)trimeric interface have been discovered recently,and a number of antibodies targeting these epitopes have been found to be capable of neutralizing a broad range of influenza isolates.These findings may have important implications for the development of universal influenza vaccines and antiviral drugs.In this review,we focused on the antibodies targeting these newly discovered epitopes in the HA domain of the influenza virus to promote the development of universal anti-influenza antibodies or vaccines and extend the discovery to other viruses with similar conformational changes in envelope proteins.

食管鳞癌患者同步放化疗期间发生严重淋巴细胞减少的影响因素分析及预测模型建立

目的分析食管鳞癌(ESCC)患者同步放化疗(CCRT)期间发生严重绝对淋巴计数(ALC)减少的影响因素,建立ESCC患者CCRT期间发生严重ALC减少的预测模型并进行效能验证。方法收集接受CCRT的286例ESCC患者的临床基线资料、放化疗方案,放疗剂量学参数等资料。根据治疗前ALC及治疗期间每周ALC,分析CCRT期间ALC动力学。根据患者CCRT期间是否发生严重ALC减少分为严重ALC减少组与非严重ALC减少组,比较两组收集的上述资料,并对CCRT期间发生严重ALC减少患者进行多因素Logistic回归分析筛选ESCC患者CCRT期间发生严重ALC减少的独立影响因素,基于影响因素构建严重ALC减少的风险预测模型(列线图)。采用受试者工作特征(ROC)、一致性指数(C-index)、校准曲线、决策分析(DCA)曲线评估列线图模型的区分度、一致性、校准度及临床适用性。结果 ESCC患者在CCRT期间ALC呈现逐渐下降趋势(P<0.05)。两组BMI、治疗前ALC、平均椎体剂量(MVD)、循环免疫细胞估计剂量(EDIC)比较差异有统计学意义(P均<0.05)。BMI(OR=0.829,95%CI:0.760~0.900)、治疗前ALC(OR=0.412,95%CI:0.253~0.648)、MVD(OR=1.079,95%CI:1.011~1.155)和EDIC(OR=1.711,95%CI:1.422~2.292)是ESCC患者CCRT期间发生严重ALC减少的独立影响因素(P<0.05)。基于上述影响因素构建列线图模型。列线图模型预测CCRT期间发生严重ALC减少的ROC下面积为0.799(95%CI:0.748~0.850);C-index为0.789;预测ALC减少风险与实际发生的风险较一致;预测发生严重ALC减少的可能在0.25~0.90概率范围内具有较好的临床适用性。结论 ESCC患者在CCRT期间发生严重ALC减少的独立影响因素为BMI、治疗前ALC、MVD、EDIC,基于上述影响因素建立的预测ESCC患者在CCRT期间发生严重ALC减少的列线图模型具有良好的区分度、一致性、校准度及临床实用性。

human monoclonal antibodies as candidate therapeutics against emerging viruses

The emergence of new pathogens, such as severe acute respiratory syndrome coronavirus （SARS-CoV）, Middle East respiratory syndrome coronavirus （MERS-CoV）, and Ebola virus, poses serious challenges to global public health and highlights the urgent need for novel antiviral approaches. Monoclonal antibodies （mAbs） have been successfully used to treat various diseases, particularly cancer and immunological disorders. Antigen-specific mAbs have been isolated using several different approaches, including hybridoma, transgenic mice, phage display, yeast display, and single B-celi isolation. Consequently, an increasing number of mAbs, which exhibit high potency against emerging viruses in vitro and in animal models of infection, have been developed. In this paper, we summarize historical trends and recent developments in mAb discovery, compare the advantages and disadvantages of various approaches to mAb production, and discuss the potential use of such strategies for the development of antivirals against emerging diseases. We also review the application of recently developed human mAbs against SARS-CoV, MERS-CoV, and Ebola virus and discuss prospects for the development of mAbs as therapeutic agents against emerging viral diseases.

Discovery and druggability evaluation of pyrrolamide-type GyrB/ParE inhibitor against drug-resistant bacterial infection

The bacterial ATP-competitive GyrB/ParE subunits of type II topoisomerase are important anti-bacterial targets to treat super drug-resistant bacterial infections.Herein we discovered novel pyrrolamide-type GyrB/ParE inhibitors based on the structural modifications of the candidate AzD5099 that was withdrawn from the clinical trials due to safety liabilities such as mitochondrial toxicity.The hydroxyisopropyl pyridazine compound 28 had a significant inhibitory effect on Gyrase(GyrB,IC_(50)=49 nmol/L)and a modest inhibitory effect on TopoⅣ(ParE,IC_(50)=1.513μmol/L)of Staphylococcus aureus.It also had significant antibacterial activities on susceptible and resistant Gram-positive bacteria with a minimum inhibitory concentration(MIC)of less than 0.03μg/mL,which showed a time-dependent bactericidal effect and low frequencies of spontaneous resistance against S.aureus.Compound 28 had better protective effects than the positive control drugs such as DS-2969(5)and AZD5099(6)in mouse models of sepsis induced by methicillin-resistant Staphylococcus aureus(MRSA)infection.It also showed better bactericidal activities than clinically used vancomycin in the mouse thigh MRSA infection models.Moreover,compound 28 has much lower mitochondrial toxicity than AZD5099(6)as well as excellent therapeutic indexes and pharmacokinetic properties.At present,compound 28 has been evaluated as a pre-clinical drug candidate for the treatment of drug-resistant Gram-positive bacterial infection.On the other hand,compound 28 also has good inhibitory activities against stubborn Gram-negative bacteria such as Escherichia coli(MIC=1μg/mL),which is comparable with the most potent pyrrolamide-type GyrB/ParE inhibitors reported recently.In addition,the structure-activity relationships of the compounds were also studied.

断面规划思想是基于生态理论提出的一种可持续和连贯的城乡发展模式。本文以云南省大理州洱源县右所镇为例,将该思想应用于小城镇风貌规划中,将小城镇的村庄和城镇视作连续统一的整体,根据不同风貌特征确定出不同的断面特征,并针对不同断面形式提出相应的控制要求。小城镇集自然、农田、村庄、镇区等要素为一体,更接近于生态学中的生态聚落,将断面规划引入到风貌规划控制中,为小城镇风貌规划控制提供一种新的视角,同时可为小城镇风貌规划控制实践提供借鉴。

动力电池是新能源汽车的核心零部件,对新能源汽车的动力性、安全性、续驶里程和使用寿命起着决定性作用。当前,国内外标准化组织机构相继制定并发布了多项动力电池标准,但现有标准内容仍然不足以覆盖动力电池全生命周期、实际复杂应用场景以及新技术等多方面使用需求。对此,本文系统归纳了国内外动力电池标准化组织机构及其发布、在研标准进展和主要内容,同时综合标准分类和实际应用场景等方面内容对比分析了国内外动力电池标准在制定原则、编制思路以及技术要求和试验方法设定等方面的差异性及主要考量因素。进一步,结合现有国内外动力电池标准现状以及技术发展趋势,重点从动力电池全生命周期的安全性、寿命及残余价值评估、全气候综合性能、绿色低碳循环要求、兼容互换要求以及现有标准体系对于新技术的适用性等六方面内容论述了现有标准体系的主要缺失项目和标准化新需求,同时在此基础之上提出了标准制修订建议,为优化完善我国动力电池标准体系提供借鉴和参考。

采用溶胶-凝胶法制备了SmMnO_(3)钙钛矿型氧化物,将其作为催化剂,在固定床反应器中研究了催化剂的焙烧温度和混合气体积空速对甲苯催化氧化效果的影响,并通过XRD、SEM、XPS、H_(2)-TPR等技术表征了催化剂的微结构及化学性质。结果表明:催化剂对甲苯的催化氧化活性随焙烧温度升高而提高;焙烧温度为800℃时制备的催化剂(SmMnO_(3)-800)对甲苯的催化氧化性能最佳,T_(50)=230℃,T_(90)=263℃,这可能归因于其较高的氧含量、较大的表面n(Mn^(4+))∶n(Mn^(3+))以及较好的低温催化氧化能力;随着混合气体积空速的增加,SmMnO_(3)-800催化剂活性有所降低;在350℃下连续反应30 h,甲苯转化率始终保持在95%以上,表明催化剂具有较好的活性稳定性。

The current COVID-19 pandemic has heavily burdened the global public health system and may keep simmering for years.The frequent emergence of immune escape variants have spurred the search for prophylactic vaccines and therapeutic antibodies that confer broad protection against SARS-CoV-2 variants.Here we show that the bivalency of an affinity maturated fully human singledomain antibody(n3113.1-Fc)exhibits exquisite neutralizing potency against SARS-CoV-2 pseudovirus,and confers effective prophylactic and therapeutic protection against authentic SARS-CoV-2 in the host cell receptor angiotensin-converting enzyme 2(ACE2)humanized mice.The crystal structure of n3113 in complex with the receptor-binding domain(RBD)of SARS-CoV-2,combined with the cryo-EM structures of n3113 and spike ecto-domain,reveals that n3113 binds to the side surface of up-state RBD with no competition with ACE2.The binding of n3113 to this novel epitope stabilizes spike in up-state conformations but inhibits SARS-CoV-2 S mediated membrane fusion,expanding our recognition of neutralization by antibodies against SARS-CoV-2.Binding assay and pseudovirus neutralization assay show no evasion of recently prevalent SARS-CoV-2 lineages,including Alpha(B.1.1.7),Beta(B.1.351),Gamma(P.1),and Delta(B.1.617.2)for n3113.1-Fc with Y58L mutation,demonstrating the potential of n3113.1-Fc(Y58L)as a promising candidate for clinical development to treat COVID-19.
