In vitro biocompatibility evaluation of silk-fibroin/ polyurethane membrane with cultivation of HUVECs

In order to investigate the in vitro biocompatibility of a novel polyurethane （PU） membrane modified by incorporation of superfine silk-fibroin powder （SFP）, which was prepared for small-diameter vascular grafts, with the cultivation of human umbilical vein endothelial cells （HUVECs）, PU and SFP were mixed with the ratios of 9：1, 7：3, 5：5, 3：7 （PU：SFP） to make four composite materials. Unmodified PU and polytetrafluoroethylene （PTFE） were added as control groups. CCK-8 assay was used to evaluate the cytotoxicity of these biomaterials. Data were processed using SPSS, and P〈 0.05 was considered to be statistically significant. Adherence and spreading of HUVECs on the surface of specimens was observed using direct contact cultivation. The toxicity ratings of the novel composites were grade 0-1, which is in the acceptable range. In all the experimental groups except control, SFP/PU with ratio of 1：9 had the least cytotoxicity property, and more content of SFP in the composite showed no improvement of the biocompatibility. HUVECs strongly attached to and grew on the surface of the biomaterials, and proliferated rapidly. The proliferation ability increased with increased proportion of SFP; however the cell quantity on the surface of the materials decreased when the proportion of SFP was equal to or larger than that of PU in the composite. It is concluded that this novel material has excellent cellular affinity with no cytotoxicity to HUVECs. Adding SFP gives PU better biocompatibility, while further research on optimum blend ratios is still needed.

Be able to form large particles with protein via electrostatic force to enhance the intensity of resonance light scattering（RLS）,（NH4）2SO4 was used as a RLS probe to determine bovine serum albumin（BSA）,human serum albumin（HSA） and ovum albumin（OVA）.The experimental conditions were investigated,including the acidity and saturation degree of （NH4）*** the optimum conditions,the enhanced RLS intensity is proportional to the concentration of BSA,HSA and OVA in the ranges of 5×10-8―1×10-6,2.5×10-8―8×10-7 and 5×10-8―1.5×10-6 mol/L,*** detection limits for BSA,HAS and OVA are 6.6×10-9,3.8×10-9 and 7.4×10-9 mol/L,*** effects of foreign substances were also *** practical and synthetic samples were analyzed with satisfactory results.

Genetic diversity of Ustilago hordei in Tibetan areas as revealed by RAPD and SSR

Covered smut, which is caused by Ustilago hordei（Pers.） Lagerh., is one of the most damaging diseases of highland barley（Hordeum vulgare Linn. var. nudum Hook. f） in Tibetan areas of China. To understand the molecular diversity of U. hordei, a total of 27 isolates, which were collected from highland barley plants from Tibet, Sichuan, qinghai, and Gansu provinces/autonomous region, were analyzed using random amplified polymorphic DNA（RAPD） and simple sequence repeat（SSR） markers. Among the 100 RAPD primers used, 24 primers exhibited polymorphism. A total of 111 fragments were amplified, of which 103 were polymorphic with a polymorphic rate of 92.79%. The average observed number of alleles（Na）, effective number of alleles（Ne）, Nei＇s genetic diversity（H）, Shannon＇s information index（I） and polymorphism information content（PIC） value in the RAPD markers were 1.9279, 1.5016, 0.2974, 0.4503 and 0.6428, respectively. For the SSR markers, 40 of the 111 primer pairs exhibited polymorphism and provided a total of 119 bands, of which 109 were polymorphic and accounted for 91.60% of the total bands. The Na, Ne, H, I and PIC values of the SSR markers were 1.9160, 1.4639, 0.2757, 0.4211 and 0.4340, respectively. The similarity coefficients ranged from 0.4957 to 0.9261 with an average of 0.7028 among all the 27 isolates used. The dendrogram, which was developed based on the RAPD and SSR combined marker dataset showed that the 27 U. hordei isolates were divided into 3 clusters at similarity coefficient of 0.7314. We determined that RAPD and SSR markers can be successfully used to assess the genetic variation among U. hordei isolates. The RAPD markers revealed higher levels of genetic polymorphism than did the SSR markers in this study. There existed a moderate genetic difference among isolates. The molecular variation and differentiation was somewhat associated with geographical origin but not for all of the isolates.

3UVH2 and SUVH9 Couple Two Essential Steps For Transcriptional Gene Silencing in Arabidopsis

In Arabidopsis, an RNA-directed DNA methylation pathway （RdDM） is responsible for de novo establish- ment of DNA methylation and contributes to transcriptional gene silencing. Recently, the microrchidia （MORC）-type ATPases were shown to play essential roles in enforcing transcriptional gene silencing of a subset of genes and transposons by regulating the formation of higher-order chromatin architecture. How- ever, how MORC proteins cooperate with the RdDM pathway components to regulate gene expression re- mains largely unclear. In this study, SUVH9 and MORC6 were identified from a screening of suppressors of idml, which is a mutant defective in active DNA demethylation. SUVH9 and MORC6 are required for silencing of two reporter genes and some endogenous genes without enhancing DNA methylation levels. SUVH9, one of SU（VAR）3-9 homologs involved in RdDM, directly interacts with MORC6 and its two close homologs, MORC1 and MORC2. Similar to MORC6, SUVH9 and its homolog SUVH2 are required for hetero- chromatin condensation and formation of 3D chromatin architecture at SDC, and Solo-LTR loci. We propose that SUVH2 and SUVH9 bind to the methylated DNA and facilitate the recruitment of a chromatin- remodeling complex to the target loci in association with MORC proteins.

Mass Spectrometric Studies on Metal-hexafluorobenzene Anionic Complexes(M=Ag, Au, Pd, Pt, Pb and Bi)

The anionic products from the reactions between metal（M=Ag, Au, Pd, Pt, Pb and Bi） vapour produced by laser ablation and hexafluorobenzene seeded in carrier gas（Ar） were studied by means of a homemade reflectron time-of-flight mass spectrometry（RTOF-MS）. Experimental results show that the dominant products were [MmC6F6]^- complexes for the reactions ofAg, Au, Pd and Pt with C6F6, while the dominant products were [MmC6F5]^- complexes for the reactions of Pb and Bi with C6F6. The formation mechanisms of the products, including the adsorption of metal cluster anions on hexafluorobenzene and the C--F cleavage induced by metal cluster anions, were discussed.

Although lithium iodide(LiI)as a redox mediator(RM)can decrease the overpotential in Li-O_(2)batteries,the stability of the Li anode is still one critical issue due to the redox ***,we firstly present a novel approach for generating Ag and LiTFSI enriched Li anode(designated as ALE@Li anode)via a spontaneous substitution between pure Li and bis(trifluoromethanesulfonyl)imide silver,in a LiI-participated Li-O_(2)*** can induce the generation of a lithiophilic solid electrolyte interphase(SEI)enriched with Ag,F,and N species(e.g.,Ag_(2)O,Li-Ag alloy,LiF,and Li_(3)N)during cell operation,which contributes to promoting the electrochemical performance through the shuttling *** to a cell with a bare Li anode,the one with as-prepared ALE@Li anode shows an enhanced cyclability,a considerable rate capability,and a good *** addition,a synchrotron X-ray computed tomography technique is employed to investigate the inhibition mechanism for shuttling effect by monitoring the morphological evolution on the cell ***,this work highlights the deliberate design in the modified Li anode in an easy-to-operate and cost-effective way as well as providing guidance for the construction of artificial SEI layers to suppress the redox shuttling of RMs in Li-O_(2)batteries.

Whether Fanconi anemia(FA)heterozygotes are predisposed to bone marrow failure and hematologic neoplasm is a crucial but unsettled issue in cancer prevention and family *** retrospectively analyzed rare possibly significant variations(PSVs)in the five most obligated FA genes,BRCA2,FANCA,FANCC,FANCD2,and FANCG,in 788 patients with aplastic anemia(AA)and hematologic ***-eight variants were identified in 66 patients(8.38%).FANCA was the most frequently mutated gene(n=29),followed by BRCA2(n=20).Compared with that of the ExAC East Asian dataset,the overall frequency of rare PSVs was higher in our cohort(P=0.016).BRCA2 PSVs showed higher frequency in acute lymphocytic leukemia(P=0.038),and FANCA PSVs were significantly enriched in AA and AML subgroups(P=0.020;P=0.008).FA-PSV-positive MDS/AML patients had a higher tumor mutation burden,higher rate of cytogenetic abnormalities,less epigenetic regulation,and fewer spliceosome gene mutations than those of FA-PSV-negative MDS/AML patients(P=0.024,P=0.029,P=0.024,and P=0.013).The overall PSV enrichment in our cohort suggests that heterozygous mutations of FA genes contribute to hematopoietic failure and leukemogenesis.

Objective:To evaluate the efficacy and safety of Chinese herbal medicines(CHMs)combined with chemical drugs for the treatment of Alzheimer's disease(AD).Materials and Methods:Databases were searched to retrieve randomized controlled trials(RCTs)of CHMs combined with tacrine,galantamine,rivastigmine,donepezil,or memantine,following strict inclusion and exclusion *** research papers published in English,Chinese,and Japanese were *** primary outcome was the mini-mental state examination(MMSE)score and the secondary outcomes were AD assessment scale cognitive subscale(ADAS-Cog)score and safety ***-analysis was carried out using Rev Man 5.3 and subgroup analysis was conducted to identify the sources of ***:A total of 15 RCTs with 1386 participants were included in this *** donepezil was used in the retrieved ***-analyses showed that the combination of CHMs with donepezil led to significant improvement in the MMSE score(Z=9.45;P<0.00001;weighted mean difference[WMD]=2.68,95%confidence interval[CI]:2.12-3.24)and a significant decrease in the ADAS-Cog score(Z=5.53;P<0.00001;WMD=-3.79;95%CI:-5.13--2.44).Safety evaluation demonstrated that these combination treatments relieved adverse events such as insomnia(risk ratio[RR]=0.20,95%CI:0.06-0.68;P=0.01)and hive(RR=0.10;95%CI:0.01-0.73;P=0.02).Conclusions:The combination of CHMs with a chemical drug like donepezil led to significant improvements in patient cognition as well as a better safety profile when compared to the application of a chemical drug alone.

A non-opioid pathway for dynorphin-caused spinal cord injury in rats

Intrathecal injection of dynorphin into rats via subarachnoid catheter induces damage to spinal cord tissue and motor function. Injection of the kappa opioid receptor antagonist nor-binaltorphine, or the excitatory amino acid N-methyl-D-aspartate receptor antagonist MK-801 into rats alleviated the pathological changes of dynorphin-caused spinal cord tissue injury and reduced the acid phosphatase activity in the spinal cord. The experimental findings indicate that there are opioid and non-opioid pathways for dynorphin-induced spinal cord injury, and that the non-opioid receptor pathway may be mediated by the excitatory amino acid N-methyl-D-aspartate receptor.
