Development and characterization of monoclonal antibodies against human aryl hydrocarbon receptor

Aryl hydrocarbon receptor（Ah R）, a ligand-dependent nuclear receptor, is involved in a diverse spectrum of biological and toxicological effects. Due to the lack of three dimensional（3D）crystal or nuclear magnetic resonance structure, the mechanisms of these complex effects of AhR remain to be unclear. Also, commercial monoclonal antibodies（mA bs） against human AhR protein（h Ah R）, as alternative immunological tools, are very limited. Thus, in order to provide more tools for further studies on h Ah R, we prepared two m Abs（1D6 and 4A6） against h Ah R. The two newly generated m Abs specifically bound to amino acids 484–508（located in transcription activation domain） and amino acids 201–215（located in Per-ARNT-Sim domain）of h Ah R, respectively. These epitopes were new as compared with those of commercial m *** m Abs were also characterized by enzyme-linked immunosorbent assay, western blot,immunoprecipitation and indirect immunofluorescence assay in different cell lines. The results showed that the two m Abs could recognize the linearized AhR s in six different human cell lines and a rat hepatoma cell line, as well as the h Ah R with native conformations. We concluded that the newly generated m Abs could be employed in AhR-based bioassays for analysis of environmental contaminants, and held great potential for further revealing the spatial structure of AhR and its biological functions in future studies.

The thermodynamic systems and dynamic model suitable for determining the nonlinear chemical fingerprints of samples were *** results indicated that the damp nonlinear chemical reactions in close systems away from the equilibrium and open systems without the complementarity of the dissipation substances have important significance for the throng characterization and whole content analysis of chemical components in *** factors influencing on nonlinear chemical fingerprint,such as reactant species and their concentrations,electrode types,temperature,stir rate,the sort,dosage and granularity of the sample,*** amply researched by a nonlinear chemistry reaction,namely,damp B-Z oscillation which used acetone and glucose as the main dissipative *** addition,the quantitative information on the whole of chemical components in samples and the traits and applications of the fingerprint were *** method and its important conditions for determining nonlinear chemistry fingerprint used in distinguishing and evaluating complex samples were successfully put forward.

Analysis on Topic Selection of Urban Planning Social Investigation Report and Its Pedagogical Exploration

Social investigation is an indispensable skill of urban planners, while "topic selection" is the fi rst step of it. With the award-winning social investigation reports of Chinese colleges and universities with undergraduate program in urban and rural planning from 2000 to 2009 as samples, this paper conducts statistical and cross analyses on the characteristics of topic selection to grasp its development tendency, and explores teaching ideas in terms of topic selection, thoughts, and awareness.

目的探讨加巴喷丁联合甲钴胺对脑梗死继发不宁腿综合征的治疗效果。方法纳入2022年1月—2023年12月在南阳市第一人民医院接受治疗的脑梗死继发不宁腿综合征患者62例,根据治疗方式分为对照组(n=30,甲钴胺)和观察组(n=32,加巴喷丁联合甲钴胺)。治疗前和治疗后评估两者患者不宁腿症状、焦虑情绪、抑郁情绪、睡眠质量,分析血红蛋白水平与不宁腿症状相关性。结果治疗前,两组患者不宁腿症状评分、汉密尔顿焦虑量表(HAMA)评分、汉密尔顿抑郁量表(HAMD)评分和匹兹堡睡眠质量指数(PSQI)评分均相近,差异无统计学意义(均P>0.05);治疗后,观察组和对照组不宁腿症状评分[分别为(8.35±2.91)分vs(18.83±3.74)分]、HAMA评分[分别为(8.91±2.10)分vs(16.23±2.78)分]、HAMD评分[分别为(7.47±2.92)分vs(17.76±2.34)分]和PSQI评分[分别为(6.35±1.36)分vs(13.83±1.64)分]比较,差异均有统计学意义(均P<0.05)。53例(85%)患者血红蛋白水平正常,9例(15%)患者血红蛋白水平偏低。结论加巴喷丁联合甲钴胺对脑梗死继发不宁腿综合征可能具有更好的治疗效果,值得在临床中进一步大样本研究。

球磨与蒸汽联合处理玉米秸秆提高水解效率的研究

为了开发绿色且高效的木质纤维素处理新技术,以玉米秸秆为模型原料,将球磨预处理与蒸汽预处理相结合用以提高玉米秸秆水解效率。研究结果表明:球磨预处理可显著提高秸秆水解效率,在10%固载量条件下,4 mm秸秆经过球磨预处理后,水解后葡萄糖和还原糖的回收率分别由38%和36%提高至65%和58%;在球磨预处理的基础上,将秸秆浸入6 g·L-1NaOH并在121℃条件下蒸汽预处理1 h,水解后葡萄糖和还原糖的回收率均达到100%,且在此预处理条件下,当固载量由10%增加至30%时,葡萄糖和还原糖的回收率仍可达到77.14%和75.82%;另外,TOC和TN分析表明,在预处理条件优化和水解效率显著提高的同时,秸秆中的非糖副产物的水解率也显著提高,圆红酵母最高可在秸秆固载量为20%时获得的水解液中进行生长和油脂发酵,生物量、油脂产量和油脂含量分别为16.00 g·L-1,6.00 g·L-1和35.61%。

恶性肿瘤的发生发展是一个多因素、多步骤参与的复杂过程,临床上肿瘤治疗存在复发率高且易耐药等现象,寻找肿瘤易转移、易复发及耐药的干预靶点对恶性肿瘤的诊断和治疗意义重大。多功能蛋白p53促凋亡刺激蛋白2(ASPP2)是一种单倍体不足肿瘤抑制因子,自被发现以来其在肿瘤中的作用备受关注。ASPP2在多种恶性肿瘤中的表达均明显下调,且其下调水平与肿瘤晚期及不良预后相关,表明其在肿瘤发生发展中扮演重要角色。本文主要综述ASPP2在肿瘤转移、耐药和代谢等方面的研究进展,为以ASPP2为治疗靶点的研究提供理论依据。

The portable adaptive optics(PAO)device is a low-cost and compact system,designed for 4-meter class telescopes that have no adaptive optics(AO)system,because of the physical space limitation at the Nasmyth or Cassegrain focus and the historically high cost of conventional *** initial scientific observations of the PAO are focused on the direct imaging of exoplanets and sub-stellar *** paper discusses the concept of PAO and the associated high-contrast imaging performance in our recent observational *** deliver a Strehl ratio better than 60%in H band under median seeing conditions of 1".Combined with our dedicated image rotation and subtraction(IRS)technique and the optimized IRS(O-IRS)algorithm,the averaged contrast ratio for a 5≤V_(mag)≤9 primary star is 1.3×10^(-5)and3.3×10^(-6)at angular distance of 0.36"with exposure time of 7 minutes and 2 hours,*** has successfully revealed the known exoplanet ofκAnd b in our recent observation with the 3.5-meter ARC telescope at Apache Point *** have performed the associated astrometry and photometry analysis of the recoveredκAnd b planet,which gives a projected separation of 0.98"±0.05",a position angle of 51.1°±0.5°and a mass of 10.15_(-1.255)^(+2.19) *** results demonstrate that PAO can be used for direct imaging of exoplanets with medium-sized telescopes.

Real-data assimilation experiment with a joint data assimilation system: assimilating carbon dioxide mole fraction measurements from the Greenhouse gases Observing Satellite

The performance of a joint data assimilation system（Tan-Tracker）,which is based on the PODEn4 Dvar assimilation method,in assimilating Greenhouse gases Observing SATellite（GOSAT） carbon dioxide（CO2） data,was *** 3D CO2 concentrations and CO2 surface fluxes（CFs） from2010 were simulated using a global chemistry transport model（GEOS-Chem）.TheTan-Tracker system used the simulated CO2 concentrations and fluxes as a background field and assimilated the GOSAT column average dry-air mole fraction of CO2（X（CO2）） data to optimize CO2 concentrations and CFs in the same assimilation *** simulated X（CO2）（X（CO2）Sim）） and assimilated X（CO2）（X（CO2）,TT） data retrieved at different satellite scan positions were compared with GOSAT-observed X（CO2）（X（CO2）,obs）*** average RMSE between the monthly X（CO2）,TT and X（CO2）,Obs data was significantly（30%） lower than the average RMSE between X（CO2）,Sim and X（CO2）,Obs）.Specifically,reductions in error were found for the positions of northern Africa（the Sahara）,the Indian peninsula,southern Africa,southern North America,and western *** difference between the correlation coefficients of the X（CO2）,Sim）and X（CO2）,Obs and those of the X（CO2）Π）,TT and X（CO2）,Obs was only *** general,the Tan-Tracker system performed very well after assimilating the GOSAT data.
