Gene transfer to human trabecular meshwork cells in vitro and ex vivo using HIV-based lentivirus

AIM: To investigate whether the enhanced green fluorescent protein(EGFP) reporter gene could be transferred into human trabecular meshwork(HTM) cells by a HIV-based lentivirus both in vitro and ex ***: The HIV-based lentivirus that contains an EF1-α promoter driving EGFP expression cassette was constructed following the standard molecular cloning methods. The cultured HTM cells were transduced at a range of multiplicity of infection(MOI) with HIV-based lentivirus. EGFP positive cell populations were detected by flow cytometry. Human anterior eye segments were cultured with perfusion culture system and transfected by HIV-based lentivirus with a 1 ×108transducing unit(TU) virus in perfusion liquid. The intraocular pressure was recorded every 8h for 21 d. The expression of EGFP in the anterior segment of the human eye was detected by fluorescence microscopy. Furthermore, the distribution of EGFP expression was confirmed by anti-EGFP immunohistochemical ***: The HIV-based lentivirus which contains an EF1-α promoter driving EGFP expression cassette was constructed successfully. After HTM cells were transduced with HIV-based lentivirus containing EGFP in vitro, the ratio of EGFP positive cells to the total cell number reached 92.3%, with the MOI of 15. After the lentivirus containing EGFP were used to transduce human anterior eye segments, the EGFP could be directly detected by fluorescence microscopy in *** staining revealed that 88.19%EGFP-positive trabecular meshwork(TM) cells were observed in the human anterior segment. Nevertheless,the intraocular pressure in the lentivirus-transduced group kept constant when compared with control group(P >0.05).CONCLUSION: EGFP gene could be efficiently transferred into HTM cells both in vitro and ex vivo by using HIV-based lentivirus.

Structural and magnetic properties of CeZnAl3 single crystals

We have synthesized single crystals of CeZnAl3, which is a new member of family of Ce-based intermetallics CeTX3 （T=transition metal, X=Si, Ge, A1）, crystallizing in the non-centrosymmetric tetragonal BaNiSns-type structure. Magnetization, specific heat and resistivity measurements all show that CeZnAls orders magnetically below around 4.4 K. Furthermore, magnetization measurements exhibit a hysteresis loop at low temperatures and fields, indicating the presence of a ferromagnetic component in the magnetic state. This points to a different nature of the magnetism in CeZnAl3 compared to the other isostructural CeTAl3 compounds. Resistivity measurements under pressures up to 1.8 GPa show a moderate suppression of the ordering temperature with pressure, suggesting that measurements to higher pressures are required to look for quantum critical behavior.

To improve the mechanical properties of AZ91D magnesium alloy,the submicrocrystal Al-Ti-B master alloy was prepared with copper mold inject casting method,and the influence of submicrocrystal Al-Ti-B master alloy on the microstructure and mechanical properties of AZ91D was *** show that,the distribution of Ti B_2 phase in submicrocrystal Al-Ti-B alloy is even and disperse,and the average size of Ti Al_3 phase is reduced from 10-30μm to～1μ*** properties of AZ91D refined with submicrocrystal Al-Ti-B master alloy are better than that with coarse-grained Al-Ti-B master alloy without copper mold inject *** tensile strength,elongation and Brinell hardness of AZ91D are increased by 10.6%,25%and 18.1%,***,refinement of AZ91D with submicrocrystal Al-Ti-B that is obtained by copper mold inject casting is an effective method to improve its mechanical properties.

To date,there is little information about the demography of vasectomy reversal(VR)patients or the factors currently influencing VR effectiveness in China,especially after the universal two-child policy was released in *** this research,demographic data and perioperative medical records of VR patients were extracted from seven major hospitals in different provinces or municipalities of ***,a telephone survey of the patients was conducted to collect follow-up ***,448 VR cases from the past 13 years were *** results were analyzed by stratified comparison to investigate factors that can influence postoperative vas deferens patency and pregnancy *** statistical methods were used,and all of the protocols were approved by the Ethics Committees of the institutes in this *** results showed that the annual operation volume of VR quadrupled after the two-child policy was *** and a long duration of vasectomy were significantly associated with a lower patency rate.A follow-up survey showed that the general postoperative pregnancy rate was 27.2%.For female partners over the age of 35 years,the postoperative pregnancy rate showed a more severe decline,but only 35.5%of them had been given a fertility examination before their husbands’VR *** work revealed that more patients in China have been demanding VR in recent ***-quality microsurgery and a short duration of vasectomy are crucial for restoring patency by *** andrologists should perform a preoperative fertility evaluation of the patients’female partners.

Pollution characteristics of particulate matters emitted from outdoor barbecue cooking in urban Jinan in eastern China

To understand the pollution characteristics of particulate matter emitted from outdoor barbecue conking in eastern China, measurements of the PM2.5 mass concentration, the number concentration of particles with a diameter of 0.01 to 1.0 pm, and the particle size distribution from 0.3 to 25 μm were carried out at seven barbecue restaurants in urban Jinan. The average PM2.5 mass concentration and sub-micron particle number concentrations at a distance of 1 m from the grills were 250 to 1083μtg/m-3 and 0.90 × 105 to 2.23 × 105 cm 3 respectively, which were much higher than those in the ambient air of the urban area. Compared to the ambient atmosphere, barbecue cooking emitted very high levels of particles with a larger increase in the concentrations of super-micron particles than that of sub-micron particles. The super-micron particle number concentrations at the barbecue restaurants were 10 to I O0 times higher than those observed in the ambient urban atmosphere. The barbecue smoke had a significant effect on the particle concentrations in the surrctindng region.. Both mass and number concentrations of particles exhibited maximum values immediately-near the barbecue grills and often reached a peak at a distance of 10 to 15 m. The removal efficiency of a range hood for the cooking particles was tested in an indoor kitchen. The range hood effectively cleaned the particulate matter pollution caused by cooking with a removal efficiency larger than 80%, Therefore, the use of a range hood is recommended for outdoor barbecue restaurants coupled with a smoke purifier to clean the emitted high concentrations of particles.

Caledonian reworking of Paleoproterozoic basement in the Cathaysia Block: Constraints from zircon U-Pb dating, Hf isotopes and trace elements

A combined study of zircon U-Pb dating, Hf isotopes and trace elements has been carried out for granodioritic neosomes of migmatites from the tianjingping area in northwestern Fujian Province. Zircons are characterized by zoning, higher Th/U ratios (mostly ≥0.1), HREE enrichment, and positive Ce and negative Eu anomalies, and show features similar to magmatic or anatectic zircons. Apparent 206Pb/238U ages for the zircons are 447±2 Ma (95% conf., MSWD = 0.88), corresponding to a Caledonian event. εHf(t) values are ?13.3 to ?9.7, indicating a crustal source. Two-stage Hf model ages are 1.7 to 1.9 Ga, suggesting that protolith of the migmates was probably formed in the Paleoproterozoic. The granodioritic neosomes have the characteristics of peraluminous calc-alkaline granite, and their REE patterns and trace elements spidergrams show features of middle to upper crustal rocks. Together with previous studies, we conclude that the protolith of the Cathaysia basement in the tianjingping area was likely formed in the middle-late Paleoproterozoic and experienced partial melting during the Caledonian period. The recognition of Caledonian reworking of the Paleoproterozoic basement in the Cathaysia Block provides a new insight into the tectonic evolution of the Cathaysia Block in the Caledonian pe- riod and the interaction between the Cathaysia Block and the Yangtze Block.

NUMERICAL SIMULATION OF ICE CONTROL EFFECT OF SOLAR ENERGY ON TIBETAN CHANNELS

With the Tanghe Diversion Channel in Tibet as an example, the theoretical study on the ice control effect of the solar sacks was conducted based on the previous study. The numerical simulation method was adopted and a one-dimensional numerical model for ice crystal in diversion channels in high-altitude cold regions was developed in this article. The heat transfer through the air-water interface and the mass transfer between ice and water were considered in the model. The model was validated by the field observation data on the diversion channel of the Tanghe Hydropower Station. The results show that the ice control effect of the solar sacks is obvious in the channel with large mass flow rate in the high-altitude cold regions.

Microstructures and Mechanical Properties of Nb-Ti Bearing Hot-rolled TRIP Steels

Nb-Tihot-rolled TRIP-assisted steel with high plasticity and appropriate volume percentage of retained austenite based on fine ferrite grain have been developed in the experiment. The test results showed that niobium tend to exist in solution state in matrix with less precipitation, and niobium-titanium could be precipitated in form of （Nb, Ti）C or （Nb, Ti）（C, N）, which play an important role in increasing yield strength （from 495 MPa to 610 MPa）. Besides, the retained austenite had a positive effect on improving the plasticity by transformation into martensite during tensile deformation.

Due to chemical inertness of nickel and boron,the preparation of nickel borides and corresponding layered ternary transition metal borides Ni_(3)ZnB_(2)(MAB phase)has always required high-temperature and/or high-pressure ***,an innovative and efficient approach to preparing Ni_(3)ZnB_(2)at only 600℃and without applied pressure is presented in this *** is discovered that by simply adjusting the temperature,a phase transition from Ni_(3)ZnB_(2)to Ni4B3 with a layered structure could be *** transition between the binary-component and the ternary-component brings about significant variation in electromagnetic wave(EMW)shielding/absorption performance of prepared *** instance,Ni2B has good EMW shielding performance(42.54 dB in X band)and Ni_(3)ZnB_(2)is of weak EMW shielding(13.43 dB in X band);Ni_(3)ZnB_(2)has poor EMW absorption performance(−5 dB)while Ni4B3 has excellent EMW absorption performance(−45.19 dB)at a thickness of 2.7 mm with effective absorption bandwidth(10.4 GHz).

A novel method through solid phase extraction combined with gradient elution for concentration and separation of 66(ultra)trace persistent toxic pollutants in Antarctic waters

This study developed a method to perform the simultaneous concentration and selective separation of 66 （ultra） trace persistent toxic substances in Antarctic waters. The substances included 30 polychlorinated biphenyls, 17 organochlorine pesticides, 16 polycyclic aromatic hydrocarbons, 3 hexabromocyclododecanes. Solid phase extraction was performed using a C18 membrane and silica gel column. Gradient elution was conducted using organic solvents with different polarities; as a result, the efficiency of the C18 film is improved and the interferences from impurities and target compounds are eliminated. Extracts were subsequently analyzed through gas chromatography or liquid and gas chromatography coupled to tandem mass spectrometry. Method validation yielded the following values： recoveries of all target analytes in the Antarctic water ranged from 87.3% to 117.6% and reproducibility as percent relative standard deviation was lower than 5%. Quantification limits ranged from 0.004 μg L^-1 to 0.030 μg L^-1. The established method improved the recoveries and reduced the limits of detection. Results indicated the method exhibited good performance in the simultaneous concentration and selective separation of 66 （ultra） trace organic pollutants; Therefore, the proposed sample pretreatment can potentially eliminate the effects of various classes of impurities to some extent.
