禽源沙门氏菌荧光定量PCR方法的构建及应用

试验旨在建立一种快速检测禽源沙门氏菌SYBR Green Ⅰ荧光定量PCR(qPCR)的方法,即根据沙门氏菌invA基因的保守序列设计引物,利用普通PCR方法扩增沙门氏菌invA基因保守基因片段,将其克隆到pMD18-T载体上,将获得的重组质粒pMD18-T-invA作为标准阳性模板。经qPCR条件优化后,进行特异性、灵敏性和重复性试验。结果显示,所建立的SYBR Green Ⅰ qPCR方法的Ct值与标准品在1.4~1.4×10^(10)拷贝/μL范围内呈良好的线性关系,R2为0.9963,扩增效率为95%,检测下限为1.4拷贝/μL;与大肠埃希菌、金黄色葡萄球菌、链球菌、痢疾志贺菌、多杀性巴氏杆菌无交叉反应;该方法组内变异系数和组间变异系数均小于2.5%;对44份粪便样本和132份蛋液样本进行qPCR方法和常规PCR方法检测,结果显示该qPCR方法的阳性检出率分别为22.7%(10/44)、0.8%(1/132),常规PCR的阳性检出率分别为9.1%(4/44),0%(0/132)。结果表明:试验成功建立禽源沙门氏菌qPCR检测方法,可为禽源沙门氏菌的快速检测提供技术支撑。

Background Heterotopic cesarean scar pregnancy （HCSP） is a very rare but life-threatening entity and there is no optimal management strategy. Here we report a successfully managed case of HCSP with expectant treatment in a tertiary referral hospital. Methods A woman with HCSP after in vitro fertilization-embryo transfer opted for expectant treatment after five days of mild bleeding and ultrasound demonstrated cardiac activity disappearance of the scar pregnancy at 8＋4 weeks of gestation. Results The patient had mild to moderate bleeding during close monitoring. Three days later, speculum examination revealed the gestational mass was partly protruding at the os of the cervix and it was removed with forceps without massive hemorrhage. A healthy male baby was delivered by cesarean section at gestational age of 36＋4 weeks. Conclusions The expectant method might be an alternative option for a HCSP with loss of cardiac activity of the scar pregnancy, when applied under supportive management and with available emergency surgery facilities.

The increasing amount of videos on the Internet and digital libraries highlights the necessity and importance of interactive video services such as automatically associating additional materials(e.g.,advertising logos and relevant selling information) with the video content so as to enrich the viewing *** this end,this paper presents a novel approach for user-targeted video content association(VCA) .In this approach,the salient objects are extracted automatically from the video stream using complementary saliency *** to these salient objects,the VCA system can push the related logo images to the *** the salient objects often correspond to important video content,the associated images can be considered as *** VCA system also allows users to associate images to the preferred video content through simple interactions by the mouse and an infrared ***,by learning the preference of each user through collecting feedbacks on the pulled or pushed images,the VCA system can provide user-targeted *** results show that our approach can effectively and efficiently extract the salient ***,subjective evaluations show that our system can provide content-related and user-targeted VCA services in a less intrusive way.

为了筛选出高酚酸含量且耐盐的金银花品种,在河北、山东、河南不同地点采集金银花品种17个,统一移植到河北省农林科学院滨海农业研究所实验基地的盐渍土(NaCl含量0.1%)上栽培繁殖,盐胁迫14 d后取枝条进行酚酸物质含量检测。采用高效液相色谱法(HPLC)与指纹图谱综合质量评价相结合的方法,以酚酸含量高为目标,对金银花品种进行筛选。结果表明:不同的金银花品种具有共同的特征色谱峰12个,特征峰中以绿原酸、异绿原酸A、异绿原酸C、木犀草苷这4种酚酸物质含量表现较高;不同品种的酚酸物质含量差异较大,其中变异株的绿原酸(15.7 mg/g)以及总酚酸物质含量(20.35 mg/g)均为最高,其次是古郡红(绿叶)(绿原酸含量为11.87 mg/g,总酚酸物质含量为17.05 mg/g),二者指标值均显著>其他品种。变异株和古郡红(绿叶)为本次试验筛选出的适合在盐碱地推广种植的品种。

Bioactive compounds could form aggregates that influence the bio-interactive processes. In this letter, based on π-π stacking models, quantitative aggregation-activity relationship （QAAR） studies were carried out on a series of sulfonylurea herbicides with good solubility. Four QAAR/QSAR models were constructed, which indicated that the bioactivity may strongly depend on both the characters of the dimeric aggregates and the monomer, The QAAR approach based on dimer-aggregates was also applicable for the highly water-soluble sulfonylurea herbicides that can form π-π stacking interactions. It was expected that the QAAR studies based on molecular aggregation state would be applied to other pesticide systems.

Background Small for gestational age （SGA） infants are associated with a high rate of oligohydramnios, stillbirth and cesarean delivery. Among SGA patients there is a higher risk of neonatal complications, such as polycythemia, hyperbilirubinemia, and hypothermia. Additionally, the SGA infant is prone to suffer from major neurologic sequelae, as well as cardiovascular system disease, in later life. Proper monitoring and therapy during pregnancy are, therefore, of utmost importance. The present study aimed to investigate the influential and prognostic factors of SGA infants.Methods From January 2001 to June 2007, a total of 55 SGA neonatal infants were included in a study group. All were born at Peking Union Medical College Hospital, with regular formal antenatal examinations. In addition, a total of 122 cases of appropriate for gestational age （AGA） infants were born at the same time and were registered into a control group. All cases were singleton pregnancies with detailed information of the maternal age, gravidity, parity, maternal height and weight, complications, uterine height and abdominal circumference, results from transabdominal ultrasonography between 32-38 gestational weeks, pregnancy duration, delivery manner, placenta, umbilical cord, and neonatal *** Significant differences were observed in placenta weight and neonatal malformations between the study and control groups. Multivariate analysis revealed increased parity, maternal hyperthyroidism and hyperthyroidism history as risk factors. Fetal abdominal circumferences less than 30 and 32 cm at 32-38 gestational weeks respectively, as determined by ultrasonography, resulted in a Youden index of 0.62. Conclusions SGA infants were associated with a greater risk of smaller placentas and infant malformations. Autophagy and apoptosis are both highly regulated biological processes that play essential roles in tissue homeostasis,development and *** is also described as a mechanism of death pathways,however,the precise mechanism of how autophagy links to cell death remains to be fully *** 1 is a dual regulator for both autophagy and *** this study we found that Beclin 1 was a substrate of caspase-3 with two cleavage sites at positions 124 and 149,***,the autophagosome formation occurred,followed by the appearance of morphological hallmarks of apoptosis after staurosporine *** cleavage products of Beclin 1 reduced autophagy and promoted apoptosis in HeLa cells and the cells in which Beclin 1 was stably knocked down by specific *** addition,the cleavage of Beclin 1 resulted in abrogating the interaction between Bcl-2 with Beclin 1,which could be blocked by ***,our results suggest that the cleavage of Beclin 1 by caspase-3 may contribute to inactivate autophagy leading towards augmented apoptosis.
