化合物A34对糖尿病小鼠的降糖作用及机制探讨

为研究一种新型可溶性环氧化物水解酶(soluble ep.xide hydr.lase,sEH)抑制剂(A34)对糖尿病小鼠的降糖作用,探讨其作用机制,连续5天给小鼠腹腔注射链脲佐菌素(str.p.ozotocin,STZ,50 mg/(kg·d)),诱导1型糖尿病(typ. 1 diabetes mellitus,T1DM)小鼠模型。通过饮水给药,考察A34对糖尿病小鼠饮水摄食量、非禁食血糖、空腹血糖、葡萄糖耐量和血浆胰岛素水平的影响,研究A34的降糖作用;通过研究胰岛中sEH表达水平、胰岛形态变化及血浆中细胞因子干扰素-γ(INF-γ)和白介素-4(IL-4)的含量,分析A34降糖的作用机制。结果表明:与模型组相比,A34有效改善了糖尿病小鼠多饮、多食症状及葡萄糖耐量情况(p.0.01),降低了空腹血糖和非禁食血糖(p.0.01),提高了血浆胰岛素水平(p.0.05);小鼠胰岛中sEH表达量降低(p.0.01),胰岛受损减轻,血浆中IFN-γ与IL-4含量比值降低(p.0.05)。A34可能通过调控促炎和抗炎细胞因子的动态平衡,保护胰岛β细胞免受损伤,从而改善小鼠胰岛β细胞功能,发挥降糖作用。

目的了解2009—2023年我国16个省份急性呼吸道感染(Ar.)病例中人呼吸道合胞病毒(Hr.V)的流行特征。方法研究资料来源于2009—2023年我国16个省份Ar.病例监测数据,共有28 278例Ar.病例纳入研究。监测病例的临床标本进行了Hr.V核酸筛查,并分析不同年龄组、不同地区和不同月份病例的Hr.V检出率差异。结果 2009年1月至2023年9月,共纳入28 278例Ar.病例。病例年龄范围为2岁,年龄M(Q1,Q3)为3岁(1岁,9岁)。28 278例Ar.病例中有3 062例Hr.V核酸阳性,总检出率为10.83%。2009—2019年Hr.V检出率为9.33%,病毒主要以冬春季流行为主。在新型冠状病毒感染(COVID-19)流行期间,Hr.V检出率在6.32%~18.67%之间波动;2022年底至2023年初未出现Hr.V传统的冬季流行高峰,在2023年4—5月出现了Hr.V反季节流行。87.95%(2 693/3 062)Hr.V阳性病例为 5岁以下的儿童,不同年龄组中Hr.V检出率的差异具有统计学意义(p.0.001),呈现出随着年龄增加Hr.V检出率降低的趋势(p.0.001),其中以r.V检出率最高(25.69%)。和2009—2019年相比,2020—2023年期间不同年龄组的Hr.V检出率从高到低的排序发生变化,Hr.V阳性病例的年龄M(Q1,Q3)从1岁(6月龄,3岁)增加至2岁(11月龄,3岁)。结论通过连续15年的Hr.V监测分析发现,5岁以下的儿童,尤其是r.V感染的主要高发人群。COVID-19流行期间,我国Hr.V流行水平和流行模式发生了变化。
