Push forward LC-MS-based therapeutic drug monitoring and pharmacometabolomics for anti-tuberculosis precision dosing and comprehensive clinical management

The spread of tuberculosis(TB),especially multidrug-resistant TB and extensively drug-resistant TB,has strongly motivated the research and development of new anti-TB drugs.New strategies to facilitate drug combinations,including pharmacokinetics-guided dose optimization and toxicology studies of first-and second-line anti-TB drugs have also been introduced and recommended.Liquid chromatography-mass spectrometry(LC-MS)has arguably become the gold standard in the analysis of both endo-and exo-genous compounds.This technique has been applied successfully not only for therapeutic drug monitoring(TDM)but also for pharmacometabolomics analysis.TDM improves the effectiveness of treatment,reduces adverse drug reactions,and the likelihood of drug resistance development in TB patients by determining dosage regimens that produce concentrations within the therapeutic target window.Based on TDM,the dose would be optimized individually to achieve favorable outcomes.Pharmacometabolomics is essential in generating and validating hypotheses regarding the metabolism of anti-TB drugs,aiding in the discovery of potential biomarkers for TB diagnostics,treatment monitoring,and outcome evaluation.This article highlighted the current progresses in TDM of anti-TB drugs based on LC-MS bioassay in the last two decades.Besides,we discussed the advantages and disadvantages of this technique in practical use.The pressing need for non-invasive sampling approaches and stability studies of anti-TB drugs was highlighted.Lastly,we provided perspectives on the prospects of combining LC-MS-based TDM and pharmacometabolomics with other advanced strategies(pharmacometrics,drug and vaccine developments,machine learning/artificial intelligence,among others)to encapsulate in an all-inclusive approach to improve treatment outcomes of TB patients.

Wedelolactone from Eclipta prostrata(L)L.suppresses inflammation and improves insulin resistance

Objective:To evaluate the effects of wedelolactone,a major flavonoid from Vietnamese Eclipta prostrata(L)L.,on inflammation and insulin resistance.Methods:Wedelolactone was extracted from the leaves of Vietnamese Eclipta prostrata(L.)L.with methanol by Soxhlet.The effects of wedelolactone on lipopolysaccharide(LPS)-induced cytokine production,reactive oxygen species(ROS)generation,and nicotinamide adenine dinucleotide phosphate(NADPH)oxidase activities in Raw 264.7 cells were measured by enzyme-linked immunosorbent assay(ELISA),specific immunofluorescent dyes and luminometric analysis,respectively.In addition,its effects on glucose uptake and the protein expression of insulin receptor substrate 1(IRS1)and glucose transporter 4(GLUT4)were examined in 3T3-L1 cells by immunofluorescent dyes and Western blot.Results:Wedelolactone at 30μg/mL significantly inhibited LPS-induced production of tumor necrosis factor-α,interleukin(IL)-6,and IL-8(P<0.01)with no noticeable effects on IL-10 level.It also reduced ROS generation and NADPH oxidase activities in LPS-stimulated Raw 264.7 cells(P<0.01).Furthermore,wedelolactone showed anti-insulin resistance activity,as evidenced by improved glucose uptake and the upregulated expression of IRS1 and GLUT4 in 3T3-L1 cells(P<0.01).Conclusions:Wedelolactone exhibits anti-inflammation and anti-insulin resistance effects,which may be used for the treatment of diabetes and inflammation-associated diseases.

Advancing Wound Filling Extraction on 3D Faces:An Auto-Segmentation and Wound Face Regeneration Approach

Facial wound segmentation plays a crucial role in preoperative planning and optimizing patient outcomes in various medical applications.In this paper,we propose an efficient approach for automating 3D facial wound segmentation using a two-stream graph convolutional network.Our method leverages the Cir3D-FaIR dataset and addresses the challenge of data imbalance through extensive experimentation with different loss functions.To achieve accurate segmentation,we conducted thorough experiments and selected a high-performing model from the trainedmodels.The selectedmodel demonstrates exceptional segmentation performance for complex 3D facial wounds.Furthermore,based on the segmentation model,we propose an improved approach for extracting 3D facial wound fillers and compare it to the results of the previous study.Our method achieved a remarkable accuracy of 0.9999993% on the test suite,surpassing the performance of the previous method.From this result,we use 3D printing technology to illustrate the shape of the wound filling.The outcomes of this study have significant implications for physicians involved in preoperative planning and intervention design.By automating facial wound segmentation and improving the accuracy ofwound-filling extraction,our approach can assist in carefully assessing and optimizing interventions,leading to enhanced patient outcomes.Additionally,it contributes to advancing facial reconstruction techniques by utilizing machine learning and 3D bioprinting for printing skin tissue implants.Our source code is available at https://***/SIMOGroup/WoundFilling3D.

Comparative study on water-soluble inorganic ions in PM2.5 from two distinct climate regions and air quality

Recently, air quality has significantly improved in developed country, but that issue is of concern in emerging megacity in developing country.In this study, aerosols and their precursor gas were collected by NILU filter pack at two distinct urban sites during the winter and summer in Osaka, Japan and dry and rainy seasons in Ho Chi Minh City(HCMC),Vietnam.The aims are to investigate the contribution of water-soluble inorganic ions(WSIIs) to PM2.5, thermodynamic characterization and possible formation pathway of secondary inorganic aerosol(SIA).The PM2.5 concentration in Osaka(15.8 μg/m^3) is lower than that in HCMC(23.0 μg/m^3), but the concentration of WSIIs in Osaka(9.0 μg/m^3) is two times higher than that in HCMC(4.1 μg/m^3).Moreover, SIA including NH4^+, NO3^-and SO4^2-are major components in WSIIs accounting for 90% and 76%(in molar) in Osaka and HCMC,respectively.Thermodynamic models were used to understand the thermodynamic characterization of urban aerosols.Overall, statistical analysis results indicate that very good agreement(R2> 0.8) was found for all species, except for nitrate aerosol in HCMC.We found that when the crustal species present at high amount, those compositions should be included in model calculation(i.***.in the HCMC situation).Finally, we analyzed the characteristics of NH4^+– NO3^-– SO4^2-system.A possible pathway to produce fine nitrate aerosol in Osaka is via the homogeneous reaction between NH3 and HNO3, while nonvolatile nitrate aerosols can be formed by the heterogeneous reactions in HCMC.

Landslide susceptibility maps (LSMs) are very crucial for planningpolicies in hazardous areas. However, the accuracy and reliability ofLSMs depend on available data and the selection of suitable methods.This study is conducted to produce LSMs by combinations of machinelearning methods and weighting techniques for Ha Giang province,Vietnam, where has limited data. In study area, we gather 11 landslideconditioning factors and establish a landslide inventory map.Computing the weights of classes (or factors) is very important toprepare data for machine learning methods to generate LSMs. Wefrst use frequency ratio (FR) and analytic hierarchy process (AHP)techniques to generate the weights. Then, random forest (RF), supportvector machine (SVM), logistic regression (LR), and AHP methods arecombined with FR and AHP weights to yield accurate and reliableLSMs. Finally, the performance of these methods is evaluated by fivestatistical metrics, ROC and R-index. The empirical results have shownthat RF is the best method in terms of R-index and the five metrics, i.e.TP rate (0.9661), FP rate (0.0), ACC (0.9835), MAE (0.0046), and RMSE(0.0350) for this study area. This study opens the perspective of weightbasedmachine learning methods for landslide susceptibility mapping.

Modified Inertial Projection Method for Solving Pseudomonotone Variational Inequalities with Non-Lipschitz in Hilbert Spaces

This paper deals with a class of inertial gradient projection methods for solving a vari-ational inequality problem involving pseudomonotone and non-Lipschitz mappings in Hilbert spaces.The proposed algorithm incorporates inertial techniques and the projection and contraction method.The weak convergence is proved without the condition of the Lipschitz continuity of the mappings.Meanwhile,the linear convergence of the algorithm is established under strong pseudomonotonicity and Lipschitz continuity assumptions.The main results obtained in this paper extend and improve some related works in the literature.

Although immunotherapy has revolutionized cancer treatment and achieved remarkable success across many different cancer types,only a subset of patients shows meaningful clinical responses.In particular,advanced prostate cancer exhibits overwhelming de novo resistance to immune checkpoint blockade therapy.This is primarily due to the immunosuppressive tumor microenvironment of prostate cancer.Therefore,it is paramount to understand how prostate cancer cell-intrinsic mechanisms promote immune evasion and foster an immunosuppressive microenvironment.Here,we review recent findings that reveal the roles of the genetic alterations,androgen receptor signaling,cancer cell plasticity,and oncogenic pathways in shaping the immunosuppressive microenvironment and thereby driving immunotherapy resistance.Based on preclinical and clinical observations,a variety of therapeutic strategies are being developed that may illuminate new paths to enhance immunotherapy efficacy in prostate cancer.

CLS-Miner: efficient and effective closed high-utility itemset mining

High-utility itemset mining (HUIM) is a popular data mining task with applications in numerous domains. However, traditional HUIM algorithms often produce a very large set of high-utility itemsets (HUIs). As a result, analyzing HUIs can be very time consuming for users. Moreover, a large set of HUIs also makes HUIM algorithms less efficient in terms of execution time and memory consumption. To address this problem, closed high-utility itemsets (CHUIs), concise and lossless representations of all HUIs, were proposed recently. Although mining CHUIs is useful and desirable, it remains a computationally expensive task. This is because current algorithms often generate a huge number of candidate itemsets and are unable to prune the search space effectively. In this paper, we address these issues by proposing a novel algorithm called CLS-Miner. The proposed algorithm utilizes the utility-list structure to directly compute the utilities of itemsets without producing candidates. It also introduces three novel strategies to reduce the search space, namely chain-estimated utility co-occurrence pruning, lower branch pruning, and pruning by coverage. Moreover, an effective method for checking whether an itemset is a subset of another itemset is introduced to further reduce the time required for discovering CHUIs. To evaluate the performance of the proposed algorithm and its novel strategies, extensive experiments have been conducted on six benchmark datasets having various characteristics. Results show that the proposed strategies are highly efficient and effective, that the proposed CLS-Miner algorithm outperforms the current state-ofthe- art CHUD and CHUI-Miner algorithms, and that CLSMiner scales linearly.

In this paper,we investigate pseudomonotone and Lipschitz continuous variational inequalities in real Hilbert spaces.For solving this problem,we propose a new method that combines the advantages of the subgradient extragradient method and the projection contraction method.Some very recent papers have considered different inertial algorithms which allowed the inertial factor is chosen in[0;1].The purpose of this work is to continue working in this direction,we propose another inertial subgradient extragradient method that the inertial factor can be chosen in a special case to be 1.Under suitable mild conditions,we establish the weak convergence of the proposed algorithm.Moreover,linear convergence is obtained under strong pseudomonotonicity and Lipschitz continuity assumptions.Finally,some numerical illustrations are given to confirm the theoretical analysis.
