In vivo radiometric analysis of glucose uptake and distribution in mouse bone

Bone formation and remodeling occurs throughout life and requires the sustained activity of osteoblasts and osteoclasts, particularly during periods of rapid bone growth. Despite increasing evidence linking bone cell activity to global energy homeostasis, little is known about the relative energy requirements or substrate utilization of bone cells. In these studies, we measured the uptake and distribution of glucose in the skeleton in vivo using positron-emitting 18F-fluorodeoxyglucose （[lSF]-FDG） and non-invasive, high-resolution positron emission tomography/computed tomography （Pe~/CT） imaging and ex vivo autoradiography. Assessment of [~SF]-FDG uptake demonstrated that relative to other tissues bone accumulated a significant fraction of the total dose of the glucose analog..Skeletal accumulation was greatest in young mice undergoing the rapid bone formation that characterizes early development. PeT/CT imaging revealed that [lSF]-FDG uptake was greatest in the epiphyseal and metaphyseal regions of long bones, which accords with the increased osteoblast numbers and activity at this skeletal site. Insulin administration significantly increased skeletal accumulation of [lSF]-FDG, while uptake was reduced in mice lacking the insulin receptor specifically in osteoblasts or fed a high-fat diet. Our results indicated that the skeleton is a site of significant glucose uptake and that its consumption by bone cells is subject to regulation by insulin and disturbances in whole-body metabolism.

Background: The Slender-billed Parakeet(enicognathus leptorhynchus) is a psittacine endemic to southern Chile and an obligate secondary cavity-nester. In the central valley of southern Chile, most(94%) of the known Slender-billed Parakeet nests have occurred in large, mature southern beech(lophozonia obliqua) trees(locally known as "pellines"). As relicts of the original old-growth forests of southern Chile, most pellines have been lost due to extensive landclearing throughout the region, potentially threatening long-term persistence of the Slender-billed ***: We conducted our study in the central valley of southern Chile, near the city of Osorno during three consecutive nesting seasons(November–January, 2008–2011). Nest trees used by Slender-billed Parakeets were located by direct observation of parakeet activities and through interviews with local residents, some of whom were former parrot nest poachers. Nest cavities were accessed, inspected and measured using single-rope climbing techniques. We report means, standard errors, 95% confidence intervals and ranges for 11 cavity-related variables. We also report clutch sizes encountered in active nests, and age estimates of nest trees based on known growth rates of lophozonia trees in southern Chile. linear regressions were used to evaluate potential relationships between cavity-related variables and clutch ***: We located and measured 38 lophozonia tree cavities used for nesting by Slender-billed Parakeets. Compared to those used by other psittacines, nest trees were relatively large, averaging 30.4 breast height of 134.5 ter ± 1.1 m in height with a mean diameter at ± 4.7 cm. Based on estimated annual diameincrement, ages of nest trees ranged from approximately 209–485 years. Nest cavities entrances averaged 12.5 ± 0.9 m in height above ground level. Cavity entrance widths averaged 51.0 ± 13.3 cm(vertical) by 11.5 ± 0.7 cm(horizontal). Cavity entrance orientations were apparently random, with

We define a generalization of Mackey first countability and prove that it is equivalent to being docile. A consequence of the main result is to give a partial affirmative answer to an old question of Mackey regarding arbitrary quotients of Mackey first countable spaces. Some applications of the main result to spaces such as inductive limits are also given.

effects of Zr on Microstructure and Short-TermStrength in GH586

0.020, 0.045 and 0.075 wt pct Zr were added to a Ni-base superalloy GH586 to investigate their effects on the microstructure and the short-term strengths at 850 degrees C. The results showed that proper Zr addition could improve the alloy tensile strength at 850 degrees C and short-term rupture life at 850 degrees C, 580 MPa. Zr exists in both gamma' phase and gamma matrix, which caused Zr to distribute in grain instead of segregating in grain boundaries, therefore increased both of their lattice parameters but decreased the mismatch between them. Meanwhile, it also increased the amount of grain-boundary carbides and decreased their size markedly. These contributed to the improvement of short-term rupture life. However, further addition of Zr resulted in forming Zr4C2S2, a plate-like stable sulphurocarbides in grain boundaries, which decreased short-term strength at high temperature, and led to a worse hot workability. According to the experimental results, the optimum content of Zr should be 0.020 wt pct.

Quantum dynamical maps are defined and studied for quantum statistical physics based on Orlicz *** complements earlier work[26]where we made a strong case for the assertion that statistical physics of regular systems should properly be based on the pair of Orlicz spaces(lcosh-1,l log(l+1)),since this framework gives a better description of regular observables,and also allows for a well-defined entropy *** the present paper we"complete"the picture by addressing the issue of the dynamics of such a system,as described by a Markov semigroup corresponding to some Dirichlet form(see[4,13,14]).Specifically,we show that even in the most general non-commutative contexts,completely positive Markov maps satisfying a natural Det ailed Balance condition canonically admit an action on a large class of quantum Orlicz *** is achieved by the development of a new interpolation strategy for extending the action of such maps to the appropriate intermediate spaces of the pair(l∞,l1).As a consequence,we obtain that completely positive quantum Markov dynamics naturally extends to the context proposed in[26].

A Systems Biology Approach for Studying Heterotopic Ossification： Proteomic Analysis of Clinical Serum and Tissue Samples

Heterotopic ossification （HO） refers to the abnormal formation of bone in soft tissue. Although some of the underlying processes of HO have been described, there are currently no clinical tests using validated biomarkers for predicting HO formation. As such, the diagnosis is made radiographically after HO has formed. To identify potential and novel biomarkers for HO, we used isobaric tags for relative and absolute quantitation （iTRAQ） and high-throughput antibody arrays to produce a semi-quantitative proteomics survey of serum and tissue from subjects with （HO ＋） and without （HO-） heterotopic ossification. The resulting data were then analyzed using a systems biology approach. We found that serum samples from subjects experiencing traumatic injuries with resulting HO have a different proteomic expression controls. Subsequent quantitative elISA identified profile compared to those from the matched five blood serum proteins that were differentially regulated between the HO-- and HO- groups. Compared to HO- samples, the amount of insulin-like growth factor I （IGF1） was up-regulated in HO＋ samples, whereas a lower amount of osteopontin （OPN）, myeloperoxidase （MPO）, runt-related transcription factor 2 （RUNX2）,and growth differentiation factor 2 or bone morphogenetic protein 9 （BMP-9） was found in HO ＋ samples （Welch two sample t-test; P 〈 0.05）. These proteins, in combination with potential serum biomarkers previously reported, are key candidates for a serum diagnostic panel that may enable early detection of HO prior to radiographic and clinical manifestations.

Decreased bioavailability of aminomethylphosphonic acid(AMPA)in genetically modified corn with activated carbon or calcium montmorillonite clay inclusion in soil

The widespread use of pesticides has resulted in detectable residues throughout the environment,sometimes at concentrations well above regulatory ***,the development of safe,effective,field-practical,and economically feasible strategies to mitigate the effects of pesticides is *** is an organophosphorus herbicide that is degraded to aminomethylphosphonic acid(AMPA),a toxic and persistent metabolite that can accumulate in soil and sediment and translocate to *** this study,we investigated the binding efficacy of activated carbon(AC)and calcium montmorillonite(CM)clay to decrease AMPA bioavailability from soil and AMPA translocation to *** isotherms and thermodynamic studies on AC and CM were conducted and showed tight binding(enthalpy values>-20 k J/mol)for AMPA with high capacities(0.25 mol/kg and 0.38 mol/kg,respectively),based on derivations from the langmuir model.A hydra assay was utilized to indicate toxicity of AMPA and the inclusion of 1%AC and CM both resulted in 90%protection of the hydra(**p≤0.01).Further studies in glyphosate-contaminated soil showed that AC and CM significantly reduced AMPA bioavailability by 53%and 44%,*** in genetically modified(GM)corn showed a conversion of glyphosate to AMPA in roots and sprouts over a 10-day exposure *** of AC and CM reduced AMPA residues in roots and sprouts by 47%-61%.These studies collectively indicate that AC and CM are effective sorbents for AMPA and could be used to reduce AMPA bioavailability from soil and AMPA residues in GM corn plants.

Beyond the RPA and GW methods with adiabatic xc-kernels for accurate ground state and quasiparticle energies

We review the theory and application of adiabatic exchange–correlation(xc)-kernels for ab initio calculations of ground state energies and quasiparticle excitations within the frameworks of the adiabatic connection fluctuation dissipation theorem and Hedin’s equations,*** different xc-kernels,which are all rooted in the homogeneous electron gas,are introduced but hereafter we focus on the specific class of renormalized adiabatic kernels,in particular the rAlDA and *** kernels drastically improve the description of short-range correlations as compared to the random phase approximation(RPA),resulting in significantly better correlation *** effect greatly reduces the reliance on error cancellations,which is essential in RPA,and systematically improves covalent bond energies while preserving the good performance of the RPA for dispersive *** quasiparticle energies,the xc-kernels account for vertex corrections that are missing in the GW *** this context,we show that the short-range correlations mainly correct the absolute band positions while the band gap is less affected in agreement with the known good performance of GW for the *** renormalized xc-kernels offer a rigorous extension of the RPA and GW methods with clear improvements in terms of accuracy at little extra computational cost.
