The systemic processes involved in the manifestation of life・threatening COVID-19 and in disease recovery are still incompletely understood,despite investigations focusing on the dysregulation of immune responses after SARS-CoV-2 *** define hallmarks of severe COVID-19 in acute disease(n=58)and in disease recovery in con valesce nt patie nts(n=28)from Han nover Medical School,we used flow cytometry and proteomics data with unsupervised clustering *** our observational study,we combined analyses of immune cells and cytokine/chemokine networks with endothelial activation and *** patients displayed an altered immune signature with prolonged lymphopenia but the expansion of granulocytes and plasmablasts along with activated and terminally differentiated T and NK cells and high levels of SARS-CoV-2-specific *** core signature of seven plasma proteins revealed a highly inflammatory microenvironment in addition to endothelial injury in severe *** within this sign ature were associated with either disease progression or *** summary,our data suggest that besides a strong inflammatory response,severe COVID-19 is driven by endothelial activation and barrier disruption,whereby recovery depends on the regeneration of the endothelial integrity.

Neutralizing antibodies targeting the receptor-binding domain(RBD)of the SARS-CoV-2 spike(S)block severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)entry into cells via surface-expressed angiotensin-converting enzyme 2(ACE2).We used a surrogate virus neutralization test(sVNT)and SARS-CoV-2 S protein-pseudotyped vesicular stomatitis virus(VSV)vector-based neutralization assay(pVNT)to assess the degree to which serum antibodies from coronavirus disease 2019(COVID-19)convalescent patients interfere with the binding of SARS-CoV-2 S to *** tests revealed neutralizing anti-SARS-CoV-2 S antibodies in the sera of ~90% of mildly and 100% of severely affected COVID-19 convalescent ***,sVNT and pVNT results correlated strongly with each other and to the levels of anti-SARS-CoV-2 S1 IgG and IgA ***,levels of neutralizing antibodies correlated with the duration and severity of clinical symptoms but not with patient *** to pVNT,sVNT is less sophisticated and does not require any biosafety *** this assay is also much faster and cheaper,sVNT will not only be important for evaluating the prevalence of neutralizing antibodies in a population but also for identifying promising plasma donors for successful passive antibody therapy.

Spatiotemporal characteristics of cultivated land use eco-efficiency and its influencing factors in China from 2000 to 2020

Improving cultivated land use eco-efficiency(CLUE)can effectively promote agricultural sustainability,particularly in developing countries where CLUE is generally *** study used provincial-level data from China to evaluate the spatiotemporal evolution of CLUE from 2000 to 2020 and identified the influencing factors of CLUE by using a panel Tobit *** addition,given the undesirable outputs of agricultural production,we incorporated carbon emissions and nonpoint source pollution into the global benchmark-undesirable output-super efficiency-slacks-based measure(GB-US-SBM)model,which combines global benchmark technology,undesirable output,super efficiency,and slacks-based *** results indicated that there was an upward trend in CLUE in China from 2000 to 2020,with an increase rate of 2.62%.The temporal evolution of CLUE in China could be classified into three distinct stages:a period of fluctuating decrease(2000-2007),a phase of gradual increase(2008-2014),and a period of rapid growth(2015-2020).The major grain-producing areas(MPAs)had a lower CLUE than their counterparts,namely,non-major grain-production areas(non-MPAs).The spatial agglomeration effect followed a northeast-southwest strip distribution;and the movement path of barycentre revealed a"P"shape,with Luoyang City,Henan Province,as the *** terms of influencing factors of CLUE,investment in science and technology played the most vital role in improving CLUE,while irrigation index had the most negative *** should be noted that these two influencing factors had different impacts on MPAs and ***,relevant departments should formulate policies to enhance the level of science and technology,improve irrigation condition,and promote sustainable utilization of cultivated land.

The emergence of severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)in 2019 prompted scientific,medical,and biotech communities to investigate infection-and vaccine-induced immune responses in the context of this pathogen.B-cell and antibody responses are at the center of these investigations,as neutralizing antibodies(nAbs)are an important correlate of protection(COP)from infection and the primary target of SARS-CoV-2 vaccine *** addition to absolute levels,nAb longevity,neutralization breadth,immunoglobulin isotype and subtype composition,and presence at mucosal sites have become important topics for scientists and health policy *** recent pandemic was and still is a unique setting in which to study de novo and memory B-cell(MBC)and antibody responses in the dynamic interplay of infection-and vaccine-induced *** also provided an opportunity to explore new vaccine platforms,such as mRNA or adenoviral vector vaccines,in unprecedented cohort *** with the technological advances of recent years,this situation has provided detailed mechanistic insights into the development of B-cell and antibody responses but also revealed some unexpected *** this review,we summarize the key findings of the last 2.5 years regarding infection-and vaccine-induced B-cell immunity,which we believe are of significant value not only in the context of SARS-CoV-2 but also for future vaccination approaches in endemic and pandemic settings.

The impact of demographic dynamics on food consumption and its environmental outcomes:Evidence from China

With increasing population and changing demographics,food consumption has experienced a significant transition in quantity and ***,a dearth of knowledge remains regarding its environmental impacts and how it responds to demographic dynamics,particularly in emerging economies like *** the two-stage Quadratic Almost Demand System(QUAIDS)model,this study empirically examines the impact of demographic dynamics on food consumption and its environmental outcomes based on the provincial data from 2000 to 2020 in *** various scenarios,according to changes in demographics,we extend our analysis to project the long-term trend of food consumption and its environmental impacts,including greenhouse gas(GHG)emissions,water footprint(Wf),and land appropriation(LA).The results reveal that an increase in the proportion of senior people significantly decreases the consumption of grain and livestock meat and increases the consumption of poultry,egg,and aquatic products,particularly for urban ***,an increase in the proportion of males in the population leads to higher consumption of poultry and aquatic ***,in the current scenario of an increased aging population and sex ratio,it is anticipated that GHG emissions,Wf,and LA are likely to decrease by 1.37,2.52,and 3.56%,*** importantly,in the scenario adhering to the standards of nutritional intake according to the Dietary Guidelines for Chinese Residents in 2022,GHG emissions,Wf,and LA in urban areas would increase by 12.78,20.94,and 18.32%,*** findings suggest that changing demographics should be considered when designing policies to mitigate the diet-environment-health trilemma and achieve sustainable food consumption.

Python Server Page Performance Analysis and Modeling

Today, in the field of computer networks, new services have been developed on the Internet or intranets, including the mail server, database management, sounds, videos and the web server itself Apache. The number of solutions for this server is therefore growing continuously, these services are becoming more and more complex and expensive, without being able to fulfill the needs of the users. The absence of benchmarks for websites with dynamic content is the major obstacle to research in this area. These users place high demands on the speed of access to information on the Internet. This is why the performance of the web server is critically important. Several factors influence performance, such as server execution speed, network saturation on the internet or intranet, increased response time, and throughputs. By measuring these factors, we propose a performance evaluation strategy for servers that allows us to determine the actual performance of different servers in terms of user satisfaction. furthermore, we identified performance characteristics such as throughput, resource utilization, and response time of a system through measurement and modeling by simulation. finally, we present a simple queue model of an Apache web server, which reasonably represents the behavior of a saturated web server using the Simulink model in Matlab (Matrix Laboratory) and also incorporates sporadic incoming traffic. We obtain server performance metrics such as average response time and throughput through simulations. Compared to other models, our model is conceptually straightforward. The model has been validated through measurements and simulations during the tests that we conducted.

Modelling of the elastoplastic behaviour of the bio-cemented soils using an extended Modified Cam Clay model

An elastoplastic constitutive model based on the Modified Cam Clay(MCC)model is developed to describe the mechanical behaviour of soils cemented via microbially induced calcite precipitation(MICP).It considers the increase of the elastic stiffness,the change of the yield surface due to MICP cementation and the degradation of calcium carbonate bonds during ***,to capture the typical contraction-dilation transition in MICP soils,the original volumetric hardening rule in the MCC model is modified to a combined deviatoric and volumetric hardening *** model could reproduce a series of drained triaxial tests on MICP-treated soils with different calcium carbonate ***,we carry out a parametric study and observe numerical instability in some *** combination with an analytical analysis,our numerical modelling has identified the benefits and limitations of using MCCbased models in the simulation of MICP-cemented soils,leading to suggestions for further model development.

A simulation study on spatial and time resolution for a cost-effective positron emission particle tracking system

This work is the second part of a simulation study investigating the processing of densely packed and moving granular assemblies by positron emission particle tracking (PEPT).Since medical positron emission tomography (PET) scanners commonly used for PEPT are very expensive,a PET-like detector system based on cost-effective organic plastic scintillator bars is being developed and tested for its *** this context,the spatial resolution of a resting positron source,a source moving on a freely designed model path,and a particle motion given by a discrete element method (DEM) simulation is studied using Monte Carlo simulations and the software toolkit *** not only extended the simulation and reconstruction to a moving source but also significantly improved the spatial resolution compared to previous work by adding oversampling and iteration to the reconstruction ***,in the case of a source following a trajectory developed from DEM simulations,a very good resolution of about 1 mm in all three directions and an average 3D deviation between simulated and reconstructed events of 2.3 mm could be ***,the resolution for realistic particle motion within the generic grate system (which is the test rig for further experimental studies) is well below the smallest particle *** simulation of the dependence of the reconstruction accuracy on tracer particle location revealed a nearly constant efficiency within the entire detector system,which demonstrates that boundary effects can be neglected.
