Increased branching independent of strigolactone in cytokinin oxidase 2-overexpressing tomato is mediated by reduced auxin transport

Tomato production is influenced by shoot branching,which is controlled by different *** we produced tomato plants overexpressing the cytokinin-deactivating gene CYTOKININ OXYDASE 2(CKX2).CKX2-overexpressing(CKX2-OE)plants showed an excessive growth of axillary shoots,the opposite phenotype expected for plants with reduced cytokinin content,as evidenced by LC-MS analysis and ARR5-GUS *** TCP transcription factor SlBRC1b was downregulated in the axillary buds of CKX2-OE and its excessive branching was dependent on a functional version of the GRAS-family gene LATERAL SUPPRESSOR(LS).Grafting experiments indicated that increased branching in CKX2-OE plants is unlikely to be mediated by root-derived *** CKX2-OE plants with transgenic antisense plants for the strigolactone biosynthesis gene CAROTENOID CLEAVAGE DIOXYGENASE(CCD7-AS)produced an additive phenotype,indicating independent effects of cytokinin and strigolactones on increased *** the other hand,CKX2-OE plants showed reduced polar auxin transport and their bud outgrowth was reduced when combined with auxin ***,CKX2-OE basal buds did not respond to auxin applied in the decapitated *** results suggest that tomato shoot branching depends on a fine-tuning of different hormonal balances and that perturbations in the auxin status could compensate for the reduced cytokinin levels in CKX2-OE plants.

Pediatric autoimmune neuropsychiatric disorders associated with or without streptococcal and other bacterial infections (PANDAS/CANS) are emerging as a featured pediatric disorder. Although there is some controversy regarding treatment approaches, especially related to the behavioral sequelae, we have hypothesized in other published work that it is characterized by the rapid onset of Reward Deficiency Syndrome (RDS) in children. We propose utilizing a multi-systems biological approach involving the coupling of genetic addiction risk testing and pro-dopamine regulation (KB220/POLYGEN®) to help induce “dopamine homeostasis” in patients with PANDAS, especially those with known DNA-induced hypodopaminergia. This case study examines a 12-year-old Caucasian male with no prior psychiatric issues who presented with a sudden onset of severe anxiety, depression, emotional liability, and suicidal ideation. The patient underwent genotyping and the genetic addiction risk score (GARS) testing, which revealed risk polymorphisms in the dopamine D2 (-DRD2/ANKK (Taq1A), OPRM1 (A/G), DRD3 (C/T), and MAOA (4R) genes. These polymorphisms have been linked to hypodopaminergia. The patient was subsequently placed on research ID-KB220ZPBMPOLY (POLYGEN®), and albeit the possibility of bias, based upon self and parental assessment, a marked rapid improvement in psychiatric symptoms was observed. In the second phase of treatment (102 days utilizing KB220), the patient received standard antibody testing, which was positive for Lyme. Antibacterial therapy started immediately, and KB220z was discontinued to provide a wash-out period. A monotonic trend analysis was performed on each outcome measure, and a consistently decreasing trend was observed utilizing antibacterial therapy. Our recommendation, albeit only one case, is to utilize and further research a combined therapeutic approach, involving precision-guided DNA testing and pro-dopamine regulation along with antibacterial therapy, as well as

It has been demonstrated that it is safe to give Gm-Csf, together with Doxorubicin, by continuous intravenous infusion, thereby substantially increasing the amount of Doxorubicin administered to the average patient, and assuring that each patient receives an individually-determined safe and maximal amount of drug. It is known that Gm-Csf is a potent inducer of components that are major factors in an immunologic attack upon neoplasms. For that reason, we thought it would be worth evaluating in 4 patients’ surface markers of dendritic precursor cells, dendritic cells [DC], and natural killer [NK] cells during the infusion. While there was substantial variation in individual responses, all 4 patients receiving Gm-Csf developed persistent marked increases in cells with each of these markers. The significance of these findings will be discussed.

A Systems Biology Approach for Studying Heterotopic Ossification： Proteomic Analysis of Clinical Serum and Tissue Samples

Heterotopic ossification （HO） refers to the abnormal formation of bone in soft tissue. Although some of the underlying processes of HO have been described, there are currently no clinical tests using validated biomarkers for predicting HO formation. As such, the diagnosis is made radiographically after HO has formed. To identify potential and novel biomarkers for HO, we used isobaric tags for relative and absolute quantitation （iTRAQ） and high-throughput antibody arrays to produce a semi-quantitative proteomics survey of serum and tissue from subjects with （HO ＋） and without （HO-） heterotopic ossification. The resulting data were then analyzed using a systems biology approach. We found that serum samples from subjects experiencing traumatic injuries with resulting HO have a different proteomic expression controls. Subsequent quantitative ELISA identified profile compared to those from the matched five blood serum proteins that were differentially regulated between the HO-- and HO- groups. Compared to HO- samples, the amount of insulin-like growth factor I （IGF1） was up-regulated in HO＋ samples, whereas a lower amount of osteopontin （OPN）, myeloperoxidase （MPO）, runt-related transcription factor 2 （RUNX2）,and growth differentiation factor 2 or bone morphogenetic protein 9 （BMP-9） was found in HO ＋ samples （Welch two sample t-test; P 〈 0.05）. These proteins, in combination with potential serum biomarkers previously reported, are key candidates for a serum diagnostic panel that may enable early detection of HO prior to radiographic and clinical manifestations.

Bulk sweeteners are generally carbohydrates, and they provide energy (calories) and bulk to food. On the other hand, high-intensity sweeteners possess a sweet taste, but are non-caloric and provide essentially no bulk to food. There are several different high-intensity, low-calorie sweeteners. Some of the sweeteners are naturally-occurring, while others are artificial or synthetic. The chemical formulas of the different sweeteners vary considerably, but generally they are relatively low molecular weight substances. They may range from simple dipeptides (e.g., aspartame) to complex organic molecules (e.g., stevioside). Most of the more commonly available high-intensity sweeteners and/or their metabolites are rapidly absorbed in the gastrointestinal tract. For example, acesulfame-K and saccharin are not metabolized and are excreted unchanged by the kidney. Sucralose, stevioside, and cyclamate undergo degrees of metabolism, and their metabolites are readily excreted. Gastrointestinal flora may, in part, assist in the metabolic breakdown depending on the parent compound. Unlike the other high-intensity sweeteners, aspartame is not absorbed intact but is metabolized in the gastrointestinal tract to naturally occurring dietary *** potential toxicity of a particular high-intensity sweetener varies. None of the more highly used high-intensity sweeteners are mutagenic, but large doses of cyclamate or saccharin in rodents have been associated with the production of bladder tumors. There is no evidence for the available high-intensity sweeteners being teratogenic or embryotoxic. High-intensity sweeteners are often non-cariogenic, i.e., they do not support growth of oral cavity microorganisms, and hence may be useful in preventive dentistry. Thus, the utilization of safe, high-intensity sweeteners in food products provides the Chinese people the opportunity to enjoy sweetness of sugar without the extra calories or cariogenic potential.
