Comparison of cardioprotective effects of mibefradil and ramipril in stroke-prone spontaneously hypertensive rats

AIM: To elucidate the cardioprotective effects of T-type calcium channel blocker mibefradil and compare with that of the angiotensin-converting enzyme inhibitor ramipril in a stroke-prone spontaneously hypertensive rats (SHR- SP) model of congestive heart failure (CHF) after myocardial infarction (MI). METHODS: SHR-SP rats were subjected to permanent ligation of the left anterior decending coronary artery. Treatment with mibefradil (10 mg·kg-1·d-1), ramipril (10 mg·kg-1·d-1), or placebo was initiated 4 weeks before surgery and continued up to 6 weeks after induction of MI. Sham-operated rats served as controls. RESULTS: In placebo-treated MI rats, six weeks after MI, left ventricular circumference, inner diameter, and left ventricular end-diastolic pressure (LVEDP) were increased, whereas mean arterial blood pressure (MAP) and maximum rate of rise of left ventricular pressure (dp/ dtmax) were decreased compared with sham-operated controls (P05), and dp/dtmax was improved (P<0.01) compared with placebo-treated MI rats. In contrast, in mibefradil-treated MI rats, heart weight, heart weight to body weight ratio were slightly but not signifi- cantly reduced, LVEDP was slightly elevated compared with placebo-treated MI rats, and was elevated (P<0.05) compared with ramipril-treated MI rats, although interstitial collagen content were reduced (P<0.01) compared with placebo-treated MI ***: Chronic treatment with ramipril diminishes cardiac remodeling of heart failure after MI to a greater extent than mibefradil. Moreover, 6 weeks after MI, mibefradil treatment results in a slight rise in LVEDP compared with placebo-treated rats. Background Stroke aetiology remains cryptogenic in a relevant proportion of patients with acute ischaemic stroke(AIS).We assessed whether enhanced diagnostic workup after AIS yields a higher rate of prespecified pathological findings compared with routine diagnostic care *** Hospitalised patients with AIS were prospectively enrolled in the investigator-initiated observational HEart and BRain Interfaces in Acute Ischaemic Stroke(HEBRAS)study at the Charité,Berlin,*** with AIS without known atrial fibrillation(AF)underwent cardiovascular MR imaging(CMR),MR-angiography of the aortic arch and prolonged Holter-ECG monitoring on top of routine diagnostic *** Among 356 patients with AIS(mean age 66 years,37.6%female),enhanced workup yielded a higher rate of prespecified pathological findings compared with routine care(17.7%vs 5.3%;p<0.001).Consequently,fewer patients were classified as cryptogenic after enhanced diagnostic workup(38.5%vs 45.5%,p<0.001).Routine care included echocardiography in 228(64.0%)*** was successfully performed in 292(82.0%)patients and revealed more often a prespecified pathological finding compared with routine echocardiography(16.1%vs 5.3%).Furthermore,study-related ECG monitoring(median duration 162 hours(IQR 98–210))detected AF in 16(4.5%)patients,while routine monitoring(median duration 51 hours(IQR 34–74))detected AF in seven(2.0%)*** Enhanced diagnostic workup revealed a higher rate of prespecified pathological findings in patients with AIS compared with routine diagnostic care and significantly reduced the proportion of patients with cryptogenic *** registration number NCT02142413.

Industry 4.0 and Cyber Physical Production Systems (CPPS) are often discussed and partially already sold. One important feature of CPPS is fault tolerance and as a consequence self-configuration and restart to increase Overall Equipment Effectiveness. To understand this challenge at first the state of the art of fault handling in industrial automated production systems (aPS) is discussed as a result of a case study analysis in eight companies developing aPS. In the next step, metrics to evaluate the concept of self-configuration and restart for aPS focusing on real-time capabilities, fault coverage and effort to increase fault coverage are proposed. Finally, two different lab size case studies prove the applicability of the concepts of self-configuration, restart and the proposed metrics.

Forest fires are key ecosystem modifiers affecting the biological,chemical,and physical attributes of forest *** extent of soil disturbance by fire is largely dependent on fire intensity,duration and recurrence,fuel load,and soil *** impact on soil properties is intricate,yielding different results based on these *** paper reviews research investigating the effects of wildfire and prescribed fire on the biological and physico-chemical attributes of forest soils and provides a summary of current knowledge associated with the benefits and disadvantages of such ***-intensity fires with ash deposition on soil surfaces cause changes in soil chemistry,including increase in available nutrients and *** intensity fires are noted for the complete combustion of organic matter and result in severe negative impacts on forest *** intensity fires result in nutrient volatilization,the break down in soil aggregate stability,an increase soil bulk density,an increase in the hydrophobicity of soil particles leading to decreased water infiltration with increased erosion and destroy soil *** soil heating(> 120℃) from high-intensity forest fires is detrimental to the soil ecosystem,especially its physical and biological *** this regard,the use of prescribed burning as a management tool to reduce the fuel load is highly recommended due to its low intensity and limited soil ***,the use of prescribed fires to manage fuel loads is critically needed in the light of current global warming as it will help prevent increased wildfire *** review provides information on the impact of forest fires on soil properties,a key feature in the maintenance of healthy *** addition,the review should prompt comprehensive soil and forest management regimes to limit soil disturbance and restore fire-disturbed soil ecosystems.

Effects of endothelin ETA receptor blocker LU 135252 on cardiac remod-eling and survival in a hypertensive rat model of chronic heart failure

Aim:To investigate whether the endothelin ETAreceptor blocker provides similarbenefit on cardiac remodeling and survival in a hypertensive rat model of chronicheart failure（CHF）.Methods:Male stroke-prone spontaneously hypertensive（SHR-SP）rats were subjected to permanent ligation of the left coronary artery andwere treated for 6 weeks with the endothelin ETAreceptor blocker LU 135252(***-1.d-1)starting 24 h after ligation or ***-operated rats servedas normal *** mean arterial blood pressure（MAP）,heart rate（HR）,leftventricular end-diastolic pressure（LVEDP）,left ventricular contractility(LVdp/dtmax),left ventricular inner diameter（LVD）and circumference（LVC）,septalthickness,left ventricular interstitial collagen content（ICC）and heart weight（HW）were measured at the end of the ***:Compared with the untreatedgroup,LU 135252 tended to increase HW（1.43±0.03 vs 1.38±0.04 g;P>0.05）,increased LVD（7.65±0.24 mm vs 6.58±0.14 mm;P0.05）,reduced LVEDP（144mmHg vs 27±4 mmHg;P<0.05）andimproved LV dp/dtmax（4230±450 mmHg/s vs 1950±400 mmHg/s;P<0.05）;survivalwas not prolonged significantly（13% vs 11%;P=NS）.Conclusion:In this hyper-tensive rat model of CHF,chronic endothelin ETA receptor blockade with LU 135252improves cardiac hemodynamics,however,it does not affect long-term survivaland worsens cardiac ***,endothelin ETAreceptor antagonists areunlikely to have an important role in the management of patients with CHF.
