Pediatric autoimmune neuropsychiatric disorders associated with or without streptococcal and other bacterial infections (PANdAS/CANS) are emerging as a featured pediatric disorder. Although there is some controversy regarding treatment approaches, especially related to the behavioral sequelae, we have hypothesized in other published work that it is characterized by the rapid onset of Reward deficiency Syndrome (RdS) in children. We propose utilizing a multi-systems biological approach involving the coupling of genetic addiction risk testing and pro-dopamine regulation (KB220/POLYGEN®) to help induce “dopamine homeostasis” in patients with PANdAS, especially those with known dNA-induced hypodopaminergia. This case study examines a 12-year-old Caucasian male with no prior psychiatric issues who presented with a sudden onset of severe anxiety, depression, emotional liability, and suicidal ideation. The patient underwent genotyping and the genetic addiction risk score (GARS) testing, which revealed risk polymorphisms in the dopamine d2 (-dRd2/ANKK (Taq1A), OPRM1 (A/G), dRd3 (C/T), and MAOA (4R) genes. These polymorphisms have been linked to hypodopaminergia. The patient was subsequently placed on research Id-KB220ZPBMPOLY (POLYGEN®), and albeit the possibility of bias, based upon self and parental assessment, a marked rapid improvement in psychiatric symptoms was observed. In the second phase of treatment (102 days utilizing KB220), the patient received standard antibody testing, which was positive for Lyme. Antibacterial therapy started immediately, and KB220z was discontinued to provide a wash-out period. A monotonic trend analysis was performed on each outcome measure, and a consistently decreasing trend was observed utilizing antibacterial therapy. Our recommendation, albeit only one case, is to utilize and further research a combined therapeutic approach, involving precision-guided dNA testing and pro-dopamine regulation along with antibacterial therapy, as well as

Effects of a regenerating matrix on the survival of birds in tropical forest fragments

Background: Vast areas of lowland neotropical forest have regenerated after initially being cleared for agricultural purposes. The ecological value of regenerating second growth to forest-dwelling birds may largely depend on the age of the forest, associated vegetative structure, and when it is capable of sustaining avian demographics similar to those found in pristine ***: To determine the influence of second growth age on bird demography, we estimated the annual survival of six central Amazonian bird species residing in pristine forest, a single 100 and a single 10 ha forest fragment, taking into consideration age of the surrounding matrix(i.e. regenerating forest adjacent to each fragment) as an explanatory ***: Study species exhibited three responses: arboreal, flocking and ant-following insectivores(Willisornis poecilinotus, Thamnomanes ardesiacus and Pithys albifrons) showed declines in survival associated with fragmentation followed by an increase in survival after 5 years of matrix regeneration. Conversely, Percnostola rufifrons, a gap-specialist, showed elevated survival in response to fragmentation followed by a decline after 5 years of regeneration. Lastly, facultative flocking and frugivore species(Glyphorynchus spirurus and dixiphia pipra, respectively) showed no response to adjacent clearing and subsequent ***: Our results in association with previous studies confirm that the value of regenerating forest surrounding habitat patches is dependent on two factors: ecological guild of the species in question and second growth age. Given the rapid increase in survival following succession, we suggest that the ecological value of young tropical forest should not be based solely on a contemporary snapshot, but rather, on the future value of mature second growth as well.

We present some ideas based on structural and functional properties of plants and animals that led to the development of two types ofbiomimetic cable entry systems. Those systems have been realized on the level of functional demonstrators.

Ensemble and single particle studies of the excitation power density (P)-dependent upconversion luminescence (UCL) of core and core-shell β-NaYF_(4):Yb,Er upconversion nanoparticles (UCNPs) doped with 20% Yb^(3+) and 1% or 3% Er^(^(3+)) performed over a P regime of 6 orders of magnitude reveal an increasing contribution of the emission from high energy Er^(3+) levels at P > 1 kW/cm^(2). This changes the overall emission color from initially green over yellow to white. While initially the green and with increasing P the red emission dominate in ensemble measurements at P dies results in a multitude of emission bands in the ultraviolet/visible/near infrared (UV/vis/NIR) accompanied by a decreased contribution of the red luminescence. Based upon a thorough analysis of the P-dependence of UCL, the emission bands activated at high P were grouped and assigned to 2–3, 3–4, and 4 photonic processes involving energy transfer (ET), excited-state absorption (ESA), cross-relaxation (CR), back energy transfer (BET), and non-radiative relaxation processes (nRP). This underlines the P-tunability of UCNP brightness and color and highlights the potential of P-dependent measurements for mechanistic studies required to manifest the population pathways of the different Er^(3+) levels.

AIM： To define the （co-）expression pattern of target receptor-tyrosine-kinases （RTK） in human gastric adenocarcinoma. METHOdS： The （co-）expression pattern of VEGFR1-3, PdGFR（α/β and EGFR1 was analyzed by RT-PCR in 51 human gastric adenocarcinomas. In addition, IHC staining was applied for confirmation of expression and analysis of RTK localisation. RESULTS： The majority of samples revealed a VEGFR1 （98%）, VEGFR2 （80%）, VEGFR3 （67%）, PdGFRα （82%） and PdGFRβ （82%） expression, whereas only 62% exhibited an EGFR1 expression. 78% of cancers expressed at least four out of six RTKs. While VEGFR1-3 and PdGFRα revealed a predominantly cytoplasmatic staining in tumor cells, accompanied by an additional nuclear staining for VEGFR3, EGFR1 was almost exclusively detected on the membrane of tumor cells. PdGFRβ was restricted to stromal pericytes, which also depicted a PdGFRα expression. CONCLUSION： Our results reveal a high rate ofreceptor-tyrosine-kinases coexpression in gastric adenocarcinoma and might therefore encourage an application of multiple-target RTK-inhibitors within a combination therapy.

ZINC FINGER- ANd TAL EFFECTOR- BASEd dNA BINdING PROTEINSProgrammable dNA binding proteins （PdPs） that selectively bind user-defined nucleotide sequences can navigate protein domains to any desired target within the genome. different functional domains can be fused to PdPs to achieve （1） acti- vation or repression of transcription, （2） nicking or cleavage of dNA, or （3） modification or removal of epigenetic marks at the targeted genome spot. Given their broad applicabil- ity, the development of highly specific, easy-to-use PdPs has been a holy grail of biotechnology since the determination of the 3-d structure of zinc fingers （ZFs） bound to dNA in 1991 （Pavletich and Pabo, 1991）. This structure suggested that tandemly arranged ZFs act as functionally independ- ent modules with each finger binding three bases. However, the combination of fingers with experimentally defined base specificities into arrays revealed that ZFs are not function- ally independent. Instead, they were found to behave in a context-dependent, and often unpredictable fashion requir- ing iterative cycles of selection to get ZF arrays with desired specificity （Joung and Sander, 2013）.

The continuous emergence of new severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)variants meansthere is a need to explore additional strategies to develop broad-spectrum vaccines or therapeutics for individuals remaining at risk of coronavirus disease 2019(COVId-19).Neutralizing monoclonal antibody(mAb)that binds to theconserved S2 subunit of the SARS-CoV-2 spike(S)protein alone,or in combination with mAb that binds to the receptor-binding domain(RBd)of S protein,might be effective in eliciting protection from infection by a variety of SARS-CoV2 *** high-throughput single-cell immunoglobulin sequencing of B cells from COVId-19-convalescent donors,we identified a high-affinity S2-specific mAb-39,that could inhibit original SARS-CoV-2 strain,Omicron BA.1,BA.2.86,BA.4,BA.5,and EG.5.1 S protein-mediated membrane fusion,leading to the neutralization of these ***,mAb-39 could also improve the neutralizing activity of anti-RBd antibody against the highlyneutralization-resistant Omicron *** docking and point mutation analyses revealed that mAb-39 recognized epitopes within the conserved upstream region of the heptad repeat 2(HR2)motif of the S2 ***,these findings demonstrate that targeting the conserved upstream region of the HR2 motif(e.g.,using mAbs)provides anovel strategy for preventing the infection of SARS-CoV-2 and its variants.

Sperm cells of flowering plants are non-motile and thus require transportation to the egg apparatus via the pollen tube to execute double fertilization. during its journey, the pollen tube interacts with various sporophytic cell types that support its growth and guide it towards the surface of the ovule. The final steps of tube guidance and sperm delivery are controlled by the cells of the female gametophyte. during fertilization, cell-cell communication events take place to achieve and maximize reproductive success. Additional layers of crosstalk exist, including self-recognition and specialized processes to prevent self-fertilization and consequent inbreeding. In this review, we focus on intercellular communication between the pollen grain/pollen tube including the sperm cells with the various sporophytic maternal tissues and the cells of the female gametophyte. Polymorphic-secreted peptides and small proteins, especially those belonging to various subclasses of small cysteine-rich proteins （CRPs）, reactive oxygen species （ROS）/NO signaling, and the second messenger Ca2＋, play center stage in most of these processes.

Halitosis:the multidisciplinary approach

Halitosis, bad breath or oral malodour are all synonyms for the same pathology. Halitosis has a large social and economic impact. For the majority of patients suffering from bad breath, it causes embarrassment and affects their social communication and life. Moreover, halitosis can be indicative of underlying diseases. Only a limited number of scientific publications were presented in this field until 1995. Ever since, a large amount of research is published, often with lack of evidence. In general, intraoral conditions, like insufficient dental hygiene, periodontitis or tongue coating are considered to be the most important cause （85%） for halitosis. Therefore, dentists and periodontologists are the first-line professionals to be confronted with this problem. They should be well aware of the origin, the detection and especially of the treatment of this pathology. In addition, ear-nose-throat-associated （10%） or gastrointestinal/ endocrinological （5%） disorders may contribute to the problem. In the case of halitophobia, psychiatrical or psychological problems may be present. Bad breath needs a multidisciplinary team approach： dentists, periodontologists, specialists in family medicine, ear- nose-throat surgeons, internal medicine and psychiatry need to be updated in this field, which still is surrounded by a large taboo. Multidisciplinary bad breath clinics offer the best environment to examine and treat this pathology that affects around 25% of the whole population. This article describes the origin, detection and treatment of halitosis, regarded from the different etiological origins.
