Utilization of RNA sequencing to investigate olfactory dysfunction in chronic rhinosinusitis without nasal polyps:A pilot study

Objectives:Prior research on olfactory dysfunction in chronic rhinosinusitis(CRS)has focused on patients with polyps and suggests that direct inflammation of the olfactory cleft mucosa plays a contributory *** purpose of this study was to evaluate gene expression in superior turbinate mucosal specimens,comparing normosmic and dysosmic CRS patients without polyps(CRSsNP).Methods:Tissue samples were obtained from the superior turbinates of patients with CRSsNP at the time of endoscopic sinus *** subsequently underwent RNA sequencing and functional analysis to investigate biological pathways associated with differentially expressed genes between dysosmic(n=7)and normosmic(n=4)***:Differential gene expression analysis comparing dysosmic and normosmic CRSsNP patients showed upregulation of 563 genes and downregulation of 327 *** stringent criteria for multiple comparisons,one upregulated gene(Immediate Early Response 3[IER3])had an false discovery rate(FDR)correction adjustedP value considered statistically significant(P<0.001,fold change 2.69).Reactome functional analysis revealed eight biological pathways significantly different between dysosmic and normosmic patients(P<0.05,FDR correction)including IL-4 and IL-13 signaling,IL-10 signaling,and rhodopsin-like ***:RNA sequencing of the superior turbinates in patients with CRSsNP can provide valuable information regarding biological pathways and genes involved in olfactory *** study supports literature suggesting that Type 2 inflammation may play a role in olfactory dysfunction in at least some patients with *** study also prompts questions regarding the role of IL-10,rhodopsin-like receptors,andIER3 in the pathogenesis of olfactory dysfunction.

Comparison of trigeminal lateralization with differing stimulants

Trigeminal-specific stimulants have been shown to activate different receptors preferentially and this likely accounts for variation in sensory *** is unclear whether trigeminal sensitivity is similar across different transient receptor potential(TRP)receptors or if dysfunction of different receptors results in differing patient ***,a prospective cohort study was conducted,consisting of trigeminal lateralization testing with three different stimulants(eucalyptol,isothiocyanate,acetic acid),olfaction testing with Sniffin'Sticks,and measurement of various patient‐reported outcome measures(PROMs).A total of 50 participants were enrolled across the olfactory *** TDI score was 27.1±8.3(range 7.0–39.5)with 38%normosmic and 62%*** trigeminal lateralization scores out of 20 in the overall cohort were 16.18(2.78)for eucalyptol,14.94(3.49)for mustard oil,and 15.28(3.68)for *** showed a significant correlation with threshold scores of Sniffin'Sticks.A significant correlation was found between acetic acid and various *** of the lateralization scores of the trigeminal stimulants correlated to each other significantly and there was no correlation to *** lack of correlation suggests that the measured sensitivity of one type of TRP receptor may not translate to similar sensitivity of the other *** investigations with TRPV1 and TRPA1 agonists are needed to corroborate our findings.

Isolating type-specific phenologies through spectral unmixing of satellite time series

Vegetation phenology is commonly studied using time series of multispectral vegetation indices derived from satellite *** in reflectance among land-cover and/or plant functional types are obscured by sub-pixel mixing,and so phenological analyses have typically sought to maximize the compositional purity of input satellite data by increasing spatial *** present an alternative method to mitigate this‘mixed-pixel problem’and extract the phenological behavior of individual land-cover types inferentially,by inverting the linear mixture model traditionally used for sub-pixel land-cover *** using genetic algorithms,the method takes advantage of the discriminating capacity of calibrated surface reflectance measurements in red,near infrared,and short-wave infrared wavelengths,as well as the Normalized Difference Vegetation Index(NDVI)and the Normalized Difference Water *** simulation,the unmixing procedure reproduced the reflectances and phenological signals of grass,crop,and deciduous forests with high fidelity(RMSE<0.007 NDVI);and in empirical tests,the algorithm extracted the phenological characteristics of evergreen trees and seasonal grasses in a semi-arid *** approach shows potential for a wide range of ecological applications,including detection of differential responses to climate,soil,or other factors among vegetation types.

Executive function impairments in high IQ children and adolescents with ADHD

Objective: To demonstrate that high IQ children and adolescents diagnosed with ADHD tend to suffer from executive function (EF) impairments that: a) can be identified with a combination of standardized measures and normed self-report data;and b) occur more frequently in this group than in the general population. Method: From charts of 117 children and adolescents aged 6 to 17 years with high IQ ( ≥ 120) who fully met DSM-IV diagnostic criteria for ADHD, data on 8 normed measures of executive function (EF) were extracted: IQ index scores for working memory and processing speed, a standardized measure of auditory verbal memory, and 5 clusters of the Brown ADD Scale, a normed, age-graded rating scale for ADHD-related executive function impairments in daily life. Significant impairment was computed for each individual relative to age-appropriate norms for each measure and comparisons were made to base-line rates in the general population. Results: Sixty-two percent of participants were significantly impaired on at least 5 of these 8 markers of EF. Chi-square comparisons of scores from these high IQ participants were significantly different (p < 0.001) from standardization norms for each of the eight EF measures. Conclusions: High IQ children and adolescents with ADHD, despite their cognitive strengths, tend to suffer from significant impairments of executive functions that can be assessed with these measures;incidence of these impairments is significantly greater than in the general population. These results are fully consistent with data on high IQ adults diagnosed with ADHD.

Extended time improves reading comprehension test scores for adolescents with ADHD

Objective: To test the hypotheses that reading comprehension difficulties of adolescent students with ADHD: 1) are related not so much to weak verbal abilities or weak basic reading skills, as to impairments of working memory and processing speed characteristic of ADHD;and 2) that extended time on a test of reading comprehension would yield significantly higher reading comprehension scores than would standard time. Method: Charts of 145 adolescents 13-18 years diagnosed with DSM-IV ADHD and no specific reading disorder after a comprehensive clinical and psycho-educational evaluation, were reviewed to extract 1) word reading and word attack subtest scores from the Woodcock-Johnson Achievement Test or the Wechsler Individual Achievement Test;2) Index scores from WISC-IV or WAIS-III IQ tests;3) scores from the Nelson-Denny Reading Test. Results: Mean index scores for verbal comprehension abilities not including reading were in the high average range, but working memory and processing speed index scores were significantly weaker. Under standard time limits 53% were unable to complete the reading comprehension test and only 42.8% were able to score within 1 SD of their IQ verbal comprehension index (VCI). When allowed extended time, 77.9% were able to score within 1 SD of their VCI. T-test comparisons between standard time and extended time were significant at < 0.001. Conclusions: Allowing extended time for adolescents with ADHD to complete tests involving reading may help to compensate for their impairments of working memory and processing speed, allowing them to score closer to their actual verbal abilities.

An autonomous adaptive low-power instrument platform(AAL-PIP)chain of six stations has been newly established on East Antarctic Plateau along the 40°geomagnetic meridian,to investigate interhemispheric geomagnetically conjugate current systems,waves,and other space weather phenomena in Polar *** six stations,PG0 to PG5,which host low-power magnetometers(Fluxgate and Searchcoil),dual frequency GPS receivers,HF radio experiment,and run autonomously with solar power and two-way satellite communication,are designated at the geomagnetically conjugate(based on the International Geomagnetic Reference Field)locations of the West Greenland geomagnetic chain covering magnetic latitudes from 70°to 80°.We present the development,deployment,and operation of this chain,as well as the data collected by the chain and some preliminary scientific results showing evidence of interhemispheric asymmetries,which are important to better understand Solar Wind–Magnetosphere–Ionosphere(SWMI)coupling in Polar *** investigations focus on magnetic impulse(MI)events,traveling convection vortices(TCVs),and ultra-low frequency(ULF)waves in the coupled southern and northern hemispheres.

We report on frequency doubling of high-energy,high repetition rate ns pulses from a cryogenically gas cooled multi-slab ytterbium-doped yttrium aluminum garnet laser system,Bivoj/DiPOLE,using a type-I phase matched lithium triborate *** achieved conversion to 515 nm with energy of 95 J at repetition rate of 10 Hz and conversion efficiency of 79%.High conversion efficiency was achieved due to successful depolarization compensation of the fundamental input beam.

Hydrogen technologies and fuel cells offer an alternative and improved solution for a decarbonised energy *** cells are electrochemical converters;transforming hydrogen (or energy sources containing hydrogen) and oxygen directly into *** hydrogen fuel cell,invented in 1839,permits the generation of electrical energy with high efficiency through a non-combustion,electrochemical process and,importantly,without the emission ofits point of ***,despite numerous efforts to exploit the obvious attractions of hydrogen technologies and hydrogen fuel cells,various challenges have been encountered,some of which are reviewed ***,however,given the exigent need to urgently seek low-carbon paths for humankind’s energy future,numerous countries are advancing the deployment of hydrogen technologies and hydrogen fuel cells not only for transport,but also as a means of the storage of excess renewable energy from,for example,wind and solar ***,hydrogen is also being blended into the natural gas supplies used in domestic heating and targeted in the decarbonisation of critical,large-scale industrial processes such as steel *** briefly review specific examples in countries such as Japan,South Korea and the People’s Republic of China,as well as selected examples from Europe and North America in the utilization of hydrogen technologies and hydrogen fuel cells.

Joint Contact Forces with Changes in Running Stride Length and Midsole Stiffness

The purpose of this study was to determine if lower extremity joint loading was influenced by stride length or shoe midsole *** subjects completed 10 trials of overground running at an average speed of 4.43 m/s in each of three condi-tions:normal running,running with a stride length(SL)reduced by 10%of normal,and running with a cushioned midsole stiffness(i.e.,mechanical impact reduction of 13.7-10.9 g).Reaction forces calculated from inverse dynamics were summed with muscle forces estimated from a musculoskeletal model using static optimization to obtain joint contact forces at the hip,knee and ankle *** components of the contact forces[axial,anterior-posterior,and medial-lateral(ML)]were examined using parametric statistics(α=0.05).Reducing stride length resulted in significant decreases in absolute peak ankle contact forces in the axial direction(normal:-14.5±1.5 BW;reduced SL:-14.0±1.6 BW)and the ML direction(normal:0.67±0.23 BW;reduced SL:0.61±0.21 BW).Reducing stride length also reduced the peak absolute axial forces at the knee(normal:-10.6±1.3 BW;reduced SL:-9.8±1.2 BW)and the hip(normal:-7.26±2.24 BW;reduced SL:-6.75±2.10 BW).The cushioned shoe did not statistically reduce the peak absolute contact forces from the normal stride condition at any of the *** hoc stress analysis suggested that the observed changes in anterior hip force would increase stress more than any of the other statistically significant *** in stride length appear to decrease some joint contact variables but cushioning in the heel region of the shoe does not.
