The objectives of the cur.ent study wer. to evaluate the estr.us r.sponse and pr.gnancy r.te following timed ar.ificial insemination (TAI) with fr.zen-thawed semen in cows. The study was car.ied out in cows at differ.nt villages of KwaZulu-Natal (KZN;n = 160) and Limpopo pr.vinces (L;n = 171). Cows wer. selected r.ndomly as pr.sented by the far.er., r.gar.less of par.ty, age, br.ed and body weight following pr.gnancy diagnosis. The cows wer. gr.uped accor.ing to br.ed type and body condition scor. (BCS) on a scale of 1-5. Selected cows wer. inser.ed a contr.lled intr.vaginal dr.g r.lease (CIDr.sup>®) and r.moved on day 8, followed by administr.tion of pr.staglandin. Heat was obser.ed on day 9 with the aid of heat mount detector. (HMD) that wer. placed on the individual cow’s tail head. Cows on heat wer. then inseminated twice at 12 hour. inter.al. Pr.gnancy diagnosis was per.or.ed by an ultr.sound scanner.and r.ctal palpation 90 days after.TAI. Data wer. analyzed using SAS 2006. Estr.us r.sponses wer. 100% in KZN and 99% in Limpopo. The lowest pr.gnancy r.te was r.cor.ed in Br.hman and Bonsmar. type cows with BCS ≤ 2.5 r.gar.less of pr.vince. Inter.stingly, Nguni type cows with BCS ≤ 2.5 had higher.aver.ge pr.gnancy r.te of 59.5% in Limpopo and 53.5% in KZN. However. cows with BCS ≥ 3 had better.pr.gnancy r.te r.gar.less of br.ed type and pr.vince. In conclusion, village cows can be synchr.nized successfully and inseminated with fr.zen-thawed semen. However. pr.gnancy r.tes ar. low in cows with lower.body condition. Village Nguni type cows wer. not affected by body condition scor.ng as they had higher.and similar.pr.gnancy r.te as those that had body condition of ≥3.

Assessment of the Susceptibility of Boar.Semen to Oxidative Str.ss

The pr.sent study was conducted to assess boar.sper. susceptibility to oxidative str.ss gener.ted by hydr.gen per.xide (H2O2). Semen was collected in r.plicates fr.m thr.e exper.mental lar.e white boar. using the gloved-hand technique. Semen ejaculates fr.m thr.e boar. wer. tr.ated with differ.nt concentr.tions of H2O2 for.thr.e hour.. SYBr.14 and Pr.pidium Iodide (PI) Live/ Dead assay kit was used to deter.ine cell viability, and Yo-pr.-1 and PI apoptosis kit was used to deter.ine cell death, namely, apoptosis. Boar.sper. motility obtained using computer.aided sper. analysis (CASA) was between 90% and 100% with mor. than 98% viability with 0% apoptotic cells. In H2O2 tr.ated boar.sper. cells, r.pid (r.P) and pr.gr.ssive motility (PM) incr.ased. Also, H2O2 tr.atment induced a high positive cor.elation with apoptosis but high negative cor.elation with viability. Hydr.gen per.xide decr.ased boar.semen total motility (TM) by 10%. In addition, most of the boar.sper. cells became apoptotic and lost 55% of viability under.oxidative str.ss induced by H2O2. This study illustr.ted that boar.semen was mor. susceptible to oxidative str.ss induced by H2O2.
