目的:探讨傅里叶域光学相干断层扫描仪(FD-OCT)测量角膜上皮厚度参数在圆锥角膜早期诊断中的价值。方法:回顾性对照研究。选取2015-01/2020-10于苏州大学附属第一医院就诊的早期圆锥角膜患者55例62眼,选取同期接受近视激光手术治疗的屈光不正患者110例110眼作为对照组。采用FD-OCT测量获得以瞳孔中心7mm直径范围的角膜上皮层厚度参数、以瞳孔中心9mm直径范围25个分区的角膜上皮层平均厚度值及角膜全层平均厚度值,比较各参数值在早期圆锥角膜组及对照组的差异,对有差异的参数分析诊断价值。结果:早期圆锥角膜组Min-e值较对照组显著减小,***值、Max-Min-e值、Max-e值较对照组显著增大(均Pe值对早期圆锥角膜诊断效能较高(AUC=0.937、0.928),Max-e值、Min-e值诊断效能中等(AUC=0.871、0.797)。25个分区角膜上皮厚度分析,早期圆锥角膜组内环颞下区(eIT1)角膜上皮层厚度变薄最明显,诊断效能最高(AUC=0.827)。各区域角膜全层厚度分析,早期圆锥角膜组各个分区的角膜全层平均厚度值较对照组明显变薄,内环颞下区(CIT1)平均厚度值在25个分区中具有较高的诊断效能(AUC=0.903)。结论:角膜上皮层厚度参数***值、Max-Min-e值、Max-e值、Min-e值、内环颞下区角膜上皮层平均厚度值以及内环颞下区角膜平均厚度值可用于鉴别早期圆锥角膜。

BACKGrOUND Perioperative fluorouracil plus leucovorin,oxaliplatin,and docetaxel(FLOT)improves prognosis in locally advanced gastric cancer(LAGC).Neutrophil-to-lymphocyte(NLr),lymphocyte-tomonocyte(LMr),and platelet-to-lymphocyte(PLr)ratios are prognostic biomarkers but not predictive *** To assess blood ratios’(NLr,LMr and PLr)potential predictive response to FLOT and survival outcomes in resectable LAGC *** This was a multicentric retrospective study investigating the clinical potential of NLr,LMr,and PLr in resectable LAGC patients,treated with at least one preoperative FLOT cycle,from 12 Portuguese *** were compared through non-parametric Mann-Whitney *** operating characteristic curve analysis defined the cut-off values as:High PLr>141 for progression and>144 for mortality;high LMr>3.56 for T stage regression(TSr).Poisson and Cox regression models the calculated relative risks/hazard ratios,using NLr,pathologic complete response,TSr,and tumor regression grade(TrG)as independent variables,and overall survival(OS)as the dependent *** This study included 295 patients(mean age,63.7 years;59.7% males).NLr was correlated with survival time(r=0.143,P=0.014).PLr was associated with systemic progression during FLOT(P=0.022)and mortality(P=0.013),with high PLr patients having a 2.2-times higher risk of progression[95% confidence interval(CI):0.89-5.26]and 1.5-times higher risk of mortality(95%CI:0.92-2.55).LMr was associated with TSr,and high LMr patients had a 1.4-times higher risk of achieving TSr(95%CI:1.01-1.99).OS benefit was found with TSr(P=0.015)and partial/complete TrG(Pents without TSr and with no evidence of pathological response had 2.1-times(95%CI:1.14-3.96)and 2.8-times(95%CI:1.6-5)higher risk of *** Higher NLr is correlated with longer survival *** LMr patients have a higher risk of decreasing T stage,whereas high PLr patients have higher odds of progressing under FLOT a

AIM:To study the effect of celecoxib(CXB) on diethylnitrosamine activation through the regulation of cytochrome P450 in a hepatocarcinogenesis ***:Six-week-old male Sprague-dawley rats were randomly divided into fi ve groups,a non-treated group(NT) ,a diethylnitrosamine-treated group(DeN) ,a DeN+CXB-treated group(DeN+CXB) ,and CXB 8 d-treated and CXB 32 d-treated *** effects of celecoxib on the enzymatic activities of CYP1A1,2A,2B1/2,and 2e1 were assessed in hepatic microsomes 24 h after DeN *** in CYP1A1 and CYP2B1/2 protein expression were also *** rate of DeN metabolism was measured by the production of the deethylation metabolite acetaldehyde,and the denitrosation metabolite ***:DeN+CXB administration produced a significant increase in the enzymatic activities of CYP2B1/2 and 1A1,whereas it did not change the activities of CYP2A and 2e1,compared to that of the DeN *** treatment for eight days did not produce a signif icant effect on enzymatic activity when compared to the NT group;however,when it was administered for prolonged times(CXB 32 d group) ,the enzymatic activities were increased in a similar pattern to those in the DeN+CXB *** observed increase in the enzymatic activities in the DeN+CXB group was accompanied by an increase in the CYP2B1/2 protein levels;no changes were observed in the levels of *** vitro,CXB increased the denitrosation of DeN,a pathway of metabolic *** addition of SKF-525A,a preferential inhibitor of CYP2B,abrogated the denitrosation of ***:These results suggest that the mechanism of action of CXB involves enhancement of the detoxif ication of DeN by an increasing denitrosation via CYP2B1/2.

Groundwater pollution by fluoride is one of the prime concerns of the world population due to its toxicity,which results in adverse health *** this paper,we review the current scenario of the fluoride contamination of groundwater in various countries across the globe and its impact on human *** the last decade,several newly affected regions have been reported all over the world,with more than 100 countries affected by fluoride contamination in groundwater(concentration exceeding the acceptable limit of 1.5 mg/L defined by the World Health Organization).Fluoride poisoning is mainly due to the unsupervised ingestion of products for dental and oral hygiene and over-fluoridated drinking *** is estimated that more than 200 million people from different countries are affected by fluoriderelated groundwater issues and health *** highest among these are from the countries in Africa(38),Asia(28),and europe(24),followed by countries like South America(5),North America(3),and Australia(2).The source of fluoride in drinking water is primarily geogenic,together with forage,grasses and grains,tea,and anthropogenic *** countries affected were correlated with climatic zones and geological factors to gain insights into geospatial *** analyses show that most of the fluoride pollution-prone zones are located in high-grade metamorphic terranes with granitoid or alkaline intrusions,geothermal hot springs,and volcanic regions with arid or semi-arid climatic *** study also finds that children across the globe are more vulnerable than adults to fluoride *** review finds that,although there are efficient fluoride removal techniques,including filters with next-generation nanomaterials,to date,there has not been a single technique developed that can claim to be a practically robust solution for fluoride removal from drinking ***,we suggest developing next-generation filters that can retain essential miner

一株鼠冠状病毒的全基因组序列特征分析

目的确定一株新型鼠冠状病毒全基因组序列特征及系统进化特征。方法采用高通量测序在内蒙地区采集的鼠肠道组织发现与已知冠状病毒差异较大的冠状病毒,拼接获得部分基因组序列,采用PCr验证扩增病毒基因组并采用rACe方法扩增基因组末端序列。采用MeGA软件进行多序列比对并构建系统发育树。结果本研究中发现的鼠冠状病毒经扩增获得其全基因组长度为27674 bp,命名为NMr-13,包括两端非编码区和8个开放阅读框,依次为5′UTr-OrF1ab-S-OrF3-e-M-OrF6-N-OrF8-3′UTr,其中OrF8仅在部分鼠类Alpha冠状病毒存在,功能尚不清楚,并且缺乏许多鼠类Alpha冠状病毒存在的NS2基因。同源性分析结果显示该病毒株属于Alpha冠状病毒属成员,与美国发现的FiCoV/UMN2020病毒株具有最高同源性,核苷酸同源性为91.3%,其次与我国浙江省发现的Lucheng/Lijiang-170,Lucheng/ruian-83和Lucheng/Lijiang-71病毒株核苷酸同源性较高,分别为79.5%,80.6%和81.0%。系统发育分析也显示NMr-13与FiCoV/UMN2020具有最近的亲缘关系。重组分析未发现重组现象。结论本研究发现了一株鼠冠状病毒,丰富了鼠类中Alpha冠状病毒的多样性,为了解国内鼠冠状病毒的分子遗传特征及分子进化提供了参考依据。

目的:探究赖氨酸特异性组蛋白去甲基化酶1(lysine-specific demethylase 1,LSD1)抑制剂对非小细胞肺癌(non-small cell lung cancer,NSCLC)A549细胞侵袭、迁移及上皮间质转化的影响。方法:将体外培养的人非小细胞肺癌A549细胞分为对照组和LSD1抑制剂组,在细胞进入对数生长期时,向实验组细胞中分别加入不同浓度的LSD1抑制剂,培养24 h。CCK-8法用于细胞增殖检测,划痕实验用于细胞迁移检测,Transwell小室法用于细胞侵袭检测,裂解细胞提取总蛋白后通过蛋白质免疫印迹技术(Western blot,WB)检测A549细胞中上皮细胞标志物e型钙黏蛋白(e-cadherin)、间充质细胞标志物N型钙黏蛋白(N-cadherin)和波形蛋白(Vimentin)的表达水平。结果:与对照组相比,LSD1抑制剂组细胞增殖率、穿膜细胞数和划痕愈合率显著降低(Pe-cadherin表达量升高,N-cadherin和Vimentin的表达量均降低(P<0.05)。结论:LSD1抑制剂可抑制非小细胞肺癌A549细胞的侵袭、迁移及上皮间质转化。

铁电陶瓷材料在外场加载下的畴变所引起的材料结构变化,是导致材料性能衰变和破坏的主要原因,raman光谱技术是一种研究铁电材料畴变和微结构变化的无损伤性及原位微区的观测方法。采用传统固相法合成Zr/Ti原子比为53/478的掺镧锆钛酸铅(PLZT)铁电陶瓷材料,采用X射线衍射仪和扫描电子显微镜及Precision_LC铁电测试系统分别对试样进行结构形貌表征和铁电物理性能测试,利用自制的应力加载装置与raman光谱仪联用,实现不同压应力场作用下试样的原位raman谱测试,考察和分析raman谱软模e(2TO)和e(3TO+2LO)+B1的峰强和峰位随散射偏振方向的变化规律。结果表明,不同压应力场下raman软模e(2TO)和e(3TO+2LO)+B1的峰强均随散射偏振角度呈现正弦式的变化规律,在60°偏振角度上软模峰强最大,在150°偏振角度上软模峰强最小。随着压应力场的增加,在0°和60°偏振角度获得的软模峰强随应力场的增加呈现明显的下降趋势,而在90°和150°偏振角度获得的软模峰强基本不变。压应力场变化对PLZT陶瓷的raman软模e(2TO)和e(3TO+2LO)+B1的峰位均不产生影响。

OBJeCTIVe: To verify the action of non-pharmacological interventions, whole body vibration exercise (WBVe) and auriculotherapy (AT) on the management of knee osteoarthritis (KOA) analyzing the handgrip strength (HS). MeTHODS: One hundred twelve participants with KOA were allocated in (a) WBVe group with peak-to-peak displacement of 2.5 to 7.5 mm, frequency from 5 up to 14 Hz, acceleration peak from 0.12 up to 2.95 g (2 d/weekly for 5 weeks),(b) AT group, points of both ears (Kidney, Knee-correspondent point and Shenmen) were stimulated with seeds,(c) WBVe + AT group and (d) respective control groups. HS was assessed in all the participants, in acute and cumulative responses. reSULTS: The intervention with WBVe alone and combined with AT improved (P e response, the HS. CONCLUSION: WBVe alone or combined with AT might promote biological effects that interfere with the HS in individuals with KOA.
