ATP synthases in chloroplasts （cpATPase） and mitochondria （mtATPase） are responsible for ATP production during photosynthesis and oxidative phosphorylation, respectively. Both enzymes consist of two multi- subunit complexes, the membrane-bound coupling factor O and the soluble coupling factor 1. During cpATPase biosynthesis, several accessory factors facilitate subunit production and orchestrate complex assembly. Here, we describe a new auxiliary protein in Arabidopsis thaliana, which is required for cpATPase accumulation. AtCGLD11 （CONSERVED IN THE GREEN LINEAGE AND DIATOMS 11） is a protein without any known functional domain and shows dual localization to chloroplasts and mitochondria. Loss of AtCGLDll function results in reduced levels of cpATPase and impaired photosynthetic performance with lower rates of ATP synthesis. In yeast two-hybrid experiments, AtCGLD11 interacts with the 13 subunits of the cpATPase and mtATPase. Our results suggest that AtCGLD11 functions in F1 assembly during cpATPase biogenesis, while its role in mtATPase biosynthesis may not, or not yet, be essential.

Typical load profile-supported convolutional neural network for short-term load forecasting in the industrial sector

Adsorption and Desorption of Cry1Ab Proteins on Differently Textured Paddy Soils

In recent years, selected cry genes from Bacillus thuringiensis（Bt） encoding the production of Cry proteins（Bt toxins） have been engineered into crop plants（Bt-crops）. Through the cultivation of Bt crops and the application of Bt pesticides, Cry proteins could be introduced into arable soils. The interaction between the proteins and soils was analyzed in this study to investigate the affinity of Cry proteins in paddy soil ecosystems. Four Paddy soils were selected to represent different soil textures. Cry proteins were spiked in soils, and the amount of protein adsorbed was measured over 24 h. Desorption of Cry1Ab proteins from paddy soils was performed by washing with sterile Milli-Q water（H_2O_（MQ））, and subsequently extracted with an extraction buffer. The paddy soils had a strong affinity for Cry1Ab proteins. Most of the Cry1Ab proteins added（＆gt; 98%） were rapidly adsorbed on the paddy soils tested. More Cry1Ab proteins were adsorbed on non-sterile soils than on sterile soils. Less than 2% of the adsorbed Cry1Ab proteins were desorbed using H2 OMQ, while a considerable proportion of the adsorbed proteins could be desorbed with the buffer, ranging from 20% to 40%.The amount of proteins desorbed increased with the increases in the initial amount of Cry1Ab proteins added to the paddy soils. The concentration of Cry1Ab proteins desorbed from the paddy soils was higher for sterile soils than non-sterile ones. Our results indicate that Bt toxins released via the cultivation of Bt crops, the application of Bt pesticides can be adsorbed on paddy soils, and soil texture could impose an impact on the adsorption capability.

Low-loss optical waveguides made with a high-loss material

For guiding light on a chip,it has been pivotal to use materials and process flows that allow low absorption and *** on subwavelength gratings,here,we show that it is possible to create broadband,multimode waveguides with very low propagation losses despite using a strongly absorbing *** perform rigorous coupled-wave analysis and finite-difference time-domain simulations of integrated waveguides that consist of pairs of integrated high-index-contrast *** showcase this concept,we demonstrate guiding of visible light in the wavelength range of 550-650 nm with losses down to 6dB/cm using silicon gratings that have a material absorption of 13,000 dB/cm at this wavelength and are fabricated with standard silicon photonics *** approach allows us to overcome traditional limits of the various established photonics technology platforms with respect to their suitable spectral range and,furthermore,to mitigate situations where absorbing materials,such as highly doped semiconductors,cannot be avoided because of the need for electrical driving,for example,for amplifiers,lasers and modulators.

Direct Measurement of CO_2 Retention in Arable Soils with pH Above 6.5 During Barometric Process Separation Incubation

The barometric process separation(BaPS)technique is a well-established incubation method to simultaneously measure gross nitrification and respiration rates in *** application,however,is still critical in soils with pH above ***,a substantial part of microbial CO_2 production is retained in soil solution(CO_2,aq)due to shifts in the carbonate *** may lead to substantial errors in gas balance ***,utilization of the BaPS technique is only reliable if the critical term is adequately *** present an easy,inexpensive,and direct method,the sterilization-CO_2-injection(SCI)method,to measure CO_2 retention during soil *** soil was incubated in the BaPS system,and defined volumes of CO_2 were injected to stepwise increase CO_2partial pressure(p CO_2)inside the chamber and to analyse the physicochemical equilibration *** exemplary agricultural soils from Northeast China and Southwest Germany were used for method establishment,presenting pH values between 4.4 and 7.6 and carbonate contents between 0% and 3.9%.We observed that in soils with pH>6.5,70%–90% of the injected CO_2 was taken up by the soil until the equilibrium inside the chamber was *** expected,in soils with low pH(<6.5),measured CO_2 retention was ***_2 retention patterns were sensitive to incubation temperature with tri-fold dissolution capacity at 5~?C compared to 25?C,but insensitive to variations in soil water *** resulting soil-specific relationship between p CO_2 and CO_2,aq concentration allowed the quantification of CO_2,aq concentration as a function of headspace p CO_2.

Over 100 Million Years of Enzyme Evolution Underpinning the Production of Morphine in the Papaveraceae Family of Flowering Plants

Phylogenomic analysis of whole genome sequences of five benzylisoquinoline alkaloid(BIA)-producing species from the Ranunculales and Proteales orders of flowering plants revealed the sequence and timing of evolutionary events leading to the diversification of these compounds.(S)-Reticuline is a pivotal intermediate in the synthesis of many BIAs and our analyses revealed parallel evolution between the two orders,which diverged122 million years ago(MYA).Berberine is present in species across the entire Ranunculales,and we found co-evolution of genes essential for production of the protoberberine *** benzophenanthridine class,which includes the antimicrobial compound sanguinarine,is specific to the Papaveraceae family of Ranunculales,and biosynthetic genes emerged after the split with the Ranunculaceae family110 MYA but before the split of the three Papaveraceae species used in this study at77 *** phthalideisoquinoline noscapine and morphinan class of BIAs are exclusive to the opium poppy *** estimation of paralogous pairs indicates that morphine biosynthesis evolved more recently than 18 MYA in the Papaver *** the preceding 100 million years gene duplication,neofunctionalization and recruitment of additional enzyme classes,combined with gene clustering,gene fusion,and gene amplification,resulted in emergence of medicinally valuable BIAs including morphine and noscapine.

Structure-Function Analysis of Arabidopsis thaliana Histidine Kinase AHK5 Bound to Its Cognate Phosphotransfer Protein AHP1

The multi-step phosphorelay （MSP） system defines a key signal transduction pathway in plants and many eukaryotes. In this system, external stimuli first lead to the activation of a histidine kinase, followed by transfer of a phosphoryl group from the receiver domain of the kinase （HKRD） to downstream, cytosolic phosphotransfer proteins （HPs）. In order to establish the determinants of specificity for this signaling relay system, we have solved the first crystal structure of a plant HKRD, AHK5RD, in complex with one of its cognate HPs, AHP1. AHP1 binds AHK5RD via a prominent hydrogen bond docking ridge and a hydrophobic patch. These features are conserved among all AHP proteins, but differ significantly from other structurally characterized prokaryotic and eukaryotic HPs. Surface plasmon resonance experiments show that AHK5RD binds to AHP1-3 with similar, micromolar affinity, consistent with the transient nature of this signaling complex. Our correlation of structural and functional data provide the first insight, at the atomic level as well as with quantitative affinity data, into the molecular recognition events governing the MSP in plants.
