First characterization of endophytic Corynespora cassiicola isolates with variant cassiicolin genes recovered from rubber trees in Brazil

Corynespora cassiicola is the causal agent of Corynespora Leaf Fall(CLF)*** is one of the most important fungal diseases of rubber trees in Asia and Africa but disease outbreaks have not been reported in South ***,a small cysteine-rich glycoprotein secreted by the pathogenic *** isolate CCP,was previously identified as a potential disease effector in rubber ***,the cassiicolin-encoding gene(Cas1)was characterized and shown to be expressed in the early phase of *** this study,we investigated whether previously undetected strains of *** are present in South American rubber plantations by examining the fungal endophyte population found in asymptomatic rubber tree *** isolates were identified as *** encoding new forms of the cassiicolin precursor protein(Cas3 and Cas4)were identified from these *** of four isolates were able to induce symptoms on the cultivar they were isolated from in a detached leaf assay,with different kinetics and *** isolate had the same pathogenicity profile as the pathogenic isolate CCP;the other two isolates developed symptoms late during the course of infection,suggesting saprotrophic ***,no Cas3 or Cas4 transcripts could be detected upon inoculation with the endophytic isolates,whereas the reference gene Cas1 was expressed upon inoculation with the CCP *** work demonstrated that *** is present in South America in an endophytic form and that it may evolve from an endophytic to a saprophytic or even potentially pathogenic life style.

A global analysis of the introduction pathways and characteristics associated with non-native fish species introduction, establishment, and impacts

Background The invasion success of introduced freshwater fishes is influenced by many factors, including ecological, species, and socioeconomic characteristics. Most studies that document the importance of these characteristics are conducted at local scales and/or focus on a single step of the invasion process. In this study, we aim to determine the species characteristics, ecological characteristics, and socioeconomic characteristics of non-native freshwater fish invasions. Our assessment was done at the global scale and considers all three steps of the invasion(i.e., introduction, establishment, and impact). For this purpose, we applied generalized linear models to 20 variables collected for 307 non-native species and modeled them as a function of ecological characteristics(i.e., environmental features), species traits(i.e., functional and morphological), and socioeconomic characteristics(i.e., human use and introduction pathways). We considered the number of countries in which each species was introduced, established, or had ecological impacts as a proxy of invasion step success. We also explored the specifics of species introduced through the aquaculture and the ornamental fish trade *** We found that non-native freshwater fishes with broad diets, high parental care, and multiple introduction pathways are the most widely introduced and established worldwide. The number of countries with impacts reported was best explained by the type of introduction pathway(i.e., aquaculture or fisheries). Moreover, among non-native species introduced through aquaculture, those belonging to Cypriniformes and having broad diets were the most widely introduced and established species. Consolidation of a Femoral Neck Fracture in Sickle Cell Disease Patient after Surgical Abstention: A Beneficial and Favorable Outcome

Femoral neck fracture is classically a traumatic pathology in the elderly but it can also occur in young people during a road traffic accident or occasionally when the femoral neck has been weakened by a pathological process. In sickle cell patients, the hip is often subjected to aseptic necrosis. The authors report the case of the child NL, aged 5 years and 4 months in whom a fracture of the femoral neck classified Delbet 3 was found as well as his sickle cell status discovered and in whom treatment by surgical abstention was carried out with a favorable evolution. This association of factors of the femoral head necrosis or nonunion is unusual and confirms the osteogenetic potential of a child. However, monitoring is necessary because other anomalies may reveal themselves.

Clinical Epidemiology and Surgical Treatment of Spontaneous Perforations of the Terminal Ileum: A Multicentre Study in Cameroon

Background: Perforations of the terminal ileum are a frequent clinical situation and a therapeutic challenge. Surgical treatment is controversial. Several surgical modalities have been proposed and postoperative morbidity and mortality remain high. The aim of this study was to review our experience in the surgical management of perforations of the terminal ileum. Patients and methods: This is a descriptive cross-sectional study with retrospective data collection from January 1, 2017 to December 31, 2021, in five hospitals in Cameroon. Patients’ demographic, clinical presentation, surgical findings and 30-days postoperative outcomes data were collected. Results: We collected 34 files. The sex ratio was 1.4 and the average age was 20.28 years. The average consultation time was 9.1 days. Abdominal pain was present in all our patients. Peritoneal irritation was present in 46 cases (88.5%). The mean time to treatment after admission was 17.5 hours. The perforation was unique in 40 cases (76.9%) and located between 6 and 10 cm from the ileocecal valve in 29 cases (55.8%). Typhoid was the most suspected etiology intraoperatively in 38 cases (73.1%). A simple suture was performed in 18 patients (34.6%), resection with anastomosis in 11 patients (21.2%) and an ileostomy in 5 patients (9.6%). The average length of hospitalization was 21.74 days. Postoperative morbidity and mortality were 32.7% and 17.3% respectively. Conclusion: Perforations of the terminal ileum are common and affect young people. Surgical procedures are varied. Reducing consultation and care times and respecting treatment principles could improve postoperative morbidity and mortality, which remain high.

Rapid prototyping of a polymer MEMS dropletdispenser by laser-assisted 3D printing

In this work, we introduce a polymer version of a previously developed silicon MEMS drop deposition tool for surfacefunctionalization that consists of a microcantilever integrating an open fluidic channel and a reservoir. The device isfabricated by laser stereolithography, which offers the advantages of low-cost and fast prototyping. Additionally,thanks to the ability to process multiple materials, a magnetic base is incorporated into the cantilever for convenienthandling and attachment to the holder of a robotized stage used for spotting. Droplets with diameters ranging from∼50 μm to ∼300 μm are printed upon direct contact of the cantilever tip with the surface to pattern. Liquid loading isachieved by fully immersing the cantilever into a reservoir drop, where a single load results in the deposition of morethan 200 droplets. The influences of the size and shape of the cantilever tip and the reservoir on the printing outcomeare studied. As a proof-of-concept of the biofunctionalization capability of this 3D printed droplet dispenser,microarrays of oligonucleotides and antibodies displaying high specificity and no cross-contamination are fabricated,and droplets are deposited at the tip of an optical fiber bundle.

Is hyperexcitability really guilty in amyotrophic lateral sclerosis?

Amyotrophic lateral sclerosis （ALS） is a lethal disorder char- acterized by the gradual degeneration of brainstem and spinal motoneurons as well as lateral cortico-spinal tracts. The onset generally occurs during the adult age except for some juvenile aggressive forms. Until recently, the vast majority of the cases （90%） were deemed sporadic. Mutations in the SOD1 gene have been for a long time the only ones reported in familiar forms of ALS. However, the recent implication of new genes of little known function cast a new view on this disease. C9ORF72, for example, is now recognized to account for 30% of the familial cases. Overall, ALS has been linked to 20 different genes, many also associated with other degenerative diseases （frontotemporal dementia, Alzheimer or ataxia）. Some of these genes are in- volved in RNA maturation （FUS, TARBD）. Clinical observation of human patients and mouse models suggests that all motor pools and motoneurons are not equally affected （see Kanning et al., 2010）. The disease usually starts in motor pools controlling the limbs or in the bulbar area before expanding to other motor pools, with the exception of a few resistant ones （Onuf＇s and oculomotor nuclei）. Within a vulnerable motor pool, motoneu- tons subtypes also exhibit differential vulnerability and follow an orderly degeneration; starting with the motoneurons innervating fast-contracting fatigable motor units （IZF motoneurons） and followed by the ones innervating fast-contractile fatigue-resistant motor units （FR motoneurons）. The motoneurons in- nervating slow motor units （S motoneurons） appear resistant to the disease. Although the mechanisms leading to the orderly degeneration are not known, many hypotheses have been raised （Ilieva et al., 2009）.

The development and application of systems strategies to biology and disease are transforming medical research and clinical practice in an unprecedented rate. In the foreseeable future, clinicians, medical researchers, and ultimately the consumers and patients will be increasingly equipped with a deluge of personal health information, e.g., whole genome sequences, molecular profiling of diseased tissues, and periodic multi-analyte blood testing of biomarker panels for disease and wellness. The convergence of these practices will enable accurate prediction of disease susceptibility and early diagnosis for actionable preventive schema and personalized treatment regimes tailored to each individual. It will also entail proactive participation from all major stakeholders in the health care system. We are at the dawn of predictive, preventive, personalized, and participatory （P4） medicine, the fully implementation of which requires marrying basic and clinical researches through advanced systems thinking and the employment of high-throughput technologies in genomics, pro- teomics, nanofluidics, single-cell analysis, and computation strategies in a highly-orchestrated discipline we termed translational systems medicine.

Redistribution and magnitude of stresses around horse shoe and circular excavations opened in anisotropic rock

In this paper numerical analysis of underground structures, taking account the transverse isotropy system of rocks, was done using CAST 3M code by varying the shape of excavation and the coefficient of earth pressure k. Numerical results reveal that the anisotropy behavior, the shape of hole and the coefficient of earth pressure k have significant influence to the mining induced stress field and rock deformations which directly control the stability of underground excavation design. The magnitude of horizontal stress obtained for the horse shoe shape excavation(25.2 MPa for k = 1; 52.7 MPa for k = 2)is lower than the magnitude obtained for circular hole(26.4 MPa for k = 1; 59.5 MPa for k = 2).Therefore, we have concluded that the horse shoe shape offers the best stability and the best design for engineer. The anisotropy system presented by rock mass can also influence the redistribution of stresses around hole opened. Numerical results have revealed that the magnitude of redistribution of horizontal stresses obtained for transverse isotropic rock(12.1 MPa for k = 0.5; 25.2 MPa for k = 1 and52.7 MPa for k = 2) is less than those obtained in the case of isotropic rock(27.6 MPa for k = 1;48.6 MPa for k = 2 and 90.81 MPa for k = 2). The more the rock has the anisotropic behavior, the more the mass of rock around the tunnel is stable.
