Comparison of Three Experimental Models for Rat Osteoarthritis Induction

Background: Osteoarthritis is a slowly progressive and debilitating disease with high prevalence in adult population. Knee is one of the joints most affected by this disorder. There are several models for animals’ osteoarthritis induction, however it is not identified any paper that compares these techniques. The present study was aimed to define the most appropriate model for rats osteoarthritis induction. Material and Methods: 40 Wistar rats were distributed into 4 groups of 10 animals each: normality group (NG);meniscectomy group (MG);quinolone group (QG) and iodoacetate group (IG). Radiographic images of the rat’s knees were analyzed as well as the amount of chondrocytes in the epiphyseal and articular cartilage. Results: In the radiographic analysis, there was a low correlation between the raters. Regarding the amount of chondrocytes in the epiphyseal cartilage, it was noticed that the IG and QG groups had fewer chondrocytes than NG, in contrast to MG that reported similar results to normality (p > 0.05). There was no significant difference between IG and QG groups (p > 0.05). Regarding the amount of chondrocytes in articular cartilage, it was noticed that the IG group showed fewer chondrocytes than NG (p 0.05). There was no significant difference between QG and MG groups (p > 0.05). Conclusion: Intraarticular injection of iodoacetate in rats is the model with greatest effect on reduction of chondrocytes amount.

Partitioning between primary and secondary metabolism of carbon allocated to roots in four maize genotypes under water deficit and its effects on productivity

Plants may respond to drought by altering biomass allocation to shoots and roots or by changing the metabolic activities in these organs. To determine how drought changes the partitioning of carbon allocated to growth and secondary metabolism in maize roots and how it affects photosynthesis(A) and productivity in maize, we evaluated leaf gas exchange, yield componentes, root morphology, and primary and secondary metabolites including total soluble sugars(TSS), starch(S), phenolics(PHE), and lignin(LIG). Data were collected from pot-grown plants of four maize genotypes: BRS 1010 and 2B710(sensitive genotypes) and DKB390 and BRS1055(tolerant genotypes) under two soil water tensions: field capacity(FC,-18 kP a) and water deficit(WD,-138 kP a). WD was applied at the pre-flowering stage for 12 days and then the water supply was restored and maintained at optimum levels until the end of the cycle. For genotype BRS 1055 under FC, the greatest A did not result in greater grain biomass(DGB) because the accumulated photoassimilates had already filled the cells, and thus the excessive TSS synthesized in leaves was allocated to roots in large amounts. However, the sharp decrease in A caused by WD imposition in this genotype did not affect the influx pressure of leaf TSS, which was due largely to conversion of primary metabolites to PHE compounds to increase the length of fine roots. In leaves of DKB390 under WD, both S and TSS were reduced, whereas PHE were increased to prevent excessive water loss and xylem cavitation. Under WD, both BRS1010 and2B710 genotypes displayed reduced allocation of biomass to shoots and roots and LIG content in leaves, as well as lower A and DGB values. In BRS1010 this response was coupled to S decrease in leaves and TSS increase in roots, whereas in 2B710 there was a concomitant S increase in roots.

A Research of the Environmental and Social Effects of the Adoption of Biotechnological Practices for Soybean Cultivation in Argentina

In recent years, the excessive and unregulated implementation of biotechnological practices has placed a large number of farmers in a precarious position throughout our country. Here we review and we analyzed results produced in field experiments installed along the Argentinean Pampas to determine environmental and social effects of the adoption of biotechnology practices associated with direct sowing (DS) of soybean. The use of machinery for farm work, the perfection of handling techniques, the incorporation of improved seeds, and the use of fertilizers and pesticides together influenced a significant increase in the productivity of the land, manual labors and capital. Despite positive economic results of this agricultural renewal also brought about consequences that proved to be detrimental to producers. By the end of the 1980s numerous sectors were in states of crisis. The major findings of our research are: 1) exhaustion of resources;2) erosion problems brought on by inappropriate land use and soil compaction by increased area under DS system;3) loss of biodiversity in the ecosystem;4) crisis among family-owned farms who cannot access to new improved technologies due to profitability, obsolete machinery and the impossibility of access to credit;5) reduction in smaller farms;6) increase in specialized production, which implies the subordination of farming production to the dynamic of capital and finally more direct relationships between the entities generating technology and primary productions. The results of our study on adoption of biotechnology practice associated to DS of soybean give the following results: 1) the deterioration of family income from farms reached 41% per hectare comparing the average values of the 1990s to the 1980s. Farms smaller than 190 hectares, therefore, ceased to be “competitive” in Buenos Aires State, it can be observed a reduction of 24.2% in the number of farms. El Soplao outcrop, an Early Cretaceous amber deposit recently discovered in northern Spain （Cantabria）, has been shown to be the largest site of amber with arthropod inclusions that has been found in Spain so far. Relevant data provided herein for biogeochemistry of the amber, palynology, taphonomy and arthropod bioinclusions complement those previously published. This set of data suggests at least two botanical sources for the amber of El Soplao deposit. The first （type A amber） strongly supports a source related to Cheirolepidiaceae, and the second （type B amber） shows non-specific conifer biomarkers. Comparison of molecular composition of type A amber with Frenelopsis leaves （Cheirolepidiaceae） strongly suggests a biochemical affinity and a common botanical origin. A preliminary palynological study indicates a regional high taxonomical diversity, mainly of pteridophyte spores and gymnosperm pollen grains. According to the preliminary palynological data, the region was inhabited by conifer forests adapted to a dry season under a subtropical climate. The abundant charcoalified wood associated with the amber in the same beds is evidence of paleofires that most likely promoted both the resin production and an intensive erosion of the litter, and subsequent great accumulation of amber plus plant cuticles. In addition, for the first time in the fossil record, charcoalified plant fibers as bioinclnsions in amber are reported. Other relevant taphonomic data are the exceptional presence of serpulids and bryozoans on the surfaces of some amber pieces indicating both a long exposure on marine or brackish-water and a mixed assemblage of amber. Lastly, new findings of insect bioiuclusions, some of them uncommon in the fossil record or showing remarkable adaptations, are reported. In conclusion, a documented scenario for the origin of the El Soplao amber outcrop is provided.

The cotton plant is a plant belonging to the family Malvaceae and its leaves are often used in folk medicine. The present study aimed to evaluate the mutagenic or antimutagenic activity of the ethanolic extract of Gossypium barbadense L. (EEG) and to analyze its capacity to prevent or repair mutagenic lesions caused by cyclophosphamide (CP). For the Micronucleus test, Swiss mice were divided into 5 groups: negative control;Positive control: treated with 50 mg/kg cyclophosphamide (CP), group receiving EEG 500 mg/kg for 7 days;Group receiving EEG 500 mg/kg for 7 days associated with CP24h before euthanasia;Group who received CP and EEG 500 mg/kg for 7 days. After the treatment period, the animals were euthanized, the bone marrow removed and the blood smear prepared. From this analysis, it was observed that the EEG caused a significant increase in the number of micronucleated erythrocytes, indicating mutagenic activity of the extract. In addition, it was verified that the extract did not present the capacity to prevent, but it presented the capacity of repair of the chromosomal damages caused by CP.

Economic Feasibility of Canola Production in the Region of Campos Gerais, Paraná, Brazil

This study aimed to analyze the economic feasibility of canola production (Brassica napus var oleifera) for the region Campos Gerais-PR. We interviewed eight producers, and used information from four of them for economic analysis of agricultural crops in 2012, 2013 and 2014. The variables analyzed were: grain production cost, economical analysis (hurdle rate, net present value, internal rate of return, payback, lucratively index, profitability index and sensitivity analysis) and vegetable oil production costs. We compared the performance of canola, wheat (Triticum aestivum)—as the main crop in the autumn/winter—and soybean (Glycine max)—by liquidity in the world market. It was concluded that although the economic analysis indicates superior results of canola production compared to the wheat crop and vegetable oil production is more attractive financially than soybean, but the culture of canola is still insignificant in the region of Campos Gerais (PR). The producers interviewed pointed to limit the expansion of culture financing the limited farming, production technology in consolidation, the need for machine adjustments and canola be host of the white mold disease (Sclerotinia sclerotiorun).

Electrochemical Evaluation of Aminoguanidine Hydrazone Derivative with Potential Anticancer Activity:Studies of Glassy Carbon/CNT and Gold Electrodes Both Modified with PAMAM

Aminoguanidine hydrazones (AGHs) are a class of compounds that have interesting pharmacological activities. They are derived from the same chemical group as aminoguanidine, so it has mixed properties (receptor and donor) in the formation of hydrogen bonds. Its anticancer agent properties were recently highlighted, but the molecules of this class have solubility in aqueous solutions that can be considered low. The identification of this class, by a simple, sensitive and low-cost technique, such as electrochemistry, which also allows the evaluation of its solubilization process through agents such as PAMAM dendrimer is the main objective of the work described here. The electrochemical response of the LQM10 (AGH derivative) was evaluated, as well as its behavior in different electrochemical sensors. Electrochemical experiments were performed in buffered (phosphate at pH 7.02 and acetate at 4.5). LQM10 has a reversible oxidation peak with a potential of +0.22 V. It was efficiently detected in different electrodes tested (glass carbon/CNT, glass carbon/CNT/PAMAM), which proves the viability of the electrodes for various analyses and has the determination of the apparent constant association, indicating its interaction with the analysis that is higher in the presence of the PAMAM encapsulating agent. This was corroborated by the results for the modified gold electrode with MUA and PAMAM. The sum of the results shows the possibility of electrochemically evaluating the Aminoguanidine hydrazone derivative, the viability of electrodes employed and the greater solubilization of LQM10 in the presence of the PAMAM dendrimer.

Corticosteroids for severe influenza pneumonia: A critical appraisal

Influenza pneumonia is associated with high number of severe cases requiring hospital and intensive care unit(ICU) admissions with high mortality. Systemic steroids are proposed as a valid therapeutic option even though its effects are still controversial. Heterogeneity of published data regarding study design, population demographics, severity of illness, dosing, type and timing of corticosteroids administered constitute an important limitation for drawing robust conclusions. However, it is reasonable to admit that, as it was not found any advantage of corticosteroid therapy in so diverse conditions, such beneficial effects do not exist at all. Its administration is likely to increase overall mortality and such trend is consistent regardless of the quality as well as the sample size of studies. Moreover it was shown that corticosteroids might be associated with higher incidence of hospital-acquired pneumonia and longer duration of mechanical ventilation and ICU stay. Finally, it is reasonable to conclude that corticosteroids failed to demonstrate any beneficial effects in the treatment of patients with severe influenza infection. Thus its current use in s evere influenza pneumonia should be restricted to very selected cases and in the setting of clinical trials.
