BACKGROUND:Although insulinomas are very rare tumors, they are the most common pancreatic neuroendocrine *** incidence in general population is 1-4 per 1 000 000 yearly but the incidence is higher in autopsy studies. The present study aims to assess the externalities of a ski resort,that is planned to be built at Tzoumerka mountains,in *** research relies on the Contingent Valuation method,which indirectly estimates the economic value of environmental and other nonmarket assets,based on the expressed preferences of members of a social *** investigation was based on primary *** core idea of questionnaire’s design,is subject to the use of a double WTP(willingness to pay)question,aiming at the monetary assessment of the attitude of the residents,regarding the upcoming infrastructure project,taking into account both its positive and its negative *** on the attitude of the survey respondents,they were asked to decide if they wish to offer an amount of money either to promote or to prevent the construction of the ski *** and non-parametric models were then applied in order to calculate in economic terms the expressed opinion of the *** primary question to be answered is whether the willingness to build the ski resort holds a higher economic value than the desire to prevent *** to the results of the survey,the vast majority of residents(92.4%)supported the idea of building the ski resort,projecting their apparent desire for the tourist and economic development of the *** reinforcement of the construction idea of the project was estimated on average at €378,*** very low proportion of the participants who disagreed with the project(5%),did not allow the economic assessment of its prevention,leaving room for further research into the application of Contingent Valuation method and its contribution in cost-benefit analysis and decision making,regarding upcoming projects.

Arabidopsis CPR5 regulates ethylene signaling via molecular association with the ETR1 receptor

The plant hormone ethylene plays various functions in plant growth, development and response to environmental stress. Ethylene is perceived by membranebound ethylene receptors, and among the homologous receptors in Arabidopsis, the ETR1 ethylene receptor plays a major role. The present study provides evidence demonstrating that Arabidopsis CPR5 functions as a novel ETR1 receptor-interacting protein in regulating ethylene response and signaling. Yeast split ubiquitin assays and bifluorescence complementation studies in plant cells indicated that CPR5 directly interacts with the ETR1 receptor. Genetic analyses indicated that mutant alleles of cpr5 can suppress ethylene insensitivity in both etr1-1 and etr1-2, but not in other dominant ethylene receptormutants. Overexpression of Arabidopsis CPR5 either in transgenic Arabidopsis plants, or ectopically in tobacco,significantly enhanced ethylene sensitivity. These findings indicate that CPR5 plays a critical role in regulating ethylene signaling. CPR5 is localized to endomembrane structures and the nucleus, and is involved in various regulatory pathways, including pathogenesis, leaf senescence, and spontaneous cell death. This study provides evidence for a novel regulatory function played by CPR5 in the ethylene receptor signaling pathway in Arabidopsis.

PCSK9 and carbohydrate metabolism:A double-edged sword

Proprotein convertase subtilisin/kexin type 9(PCSK9) plays a paramount role in the degradation of lowdensity lipoprotein(LDL) receptors(LDLR) on the hepatic cells surface and subsequently affects LDL particles catabolism and LDL cholesterol(LDL-c) levels. The anti-PCSK9 monoclonal antibodies lead to substantial decrease of LDL-c concentration. PCSK9(which is also expressed in pancreatic delta-cells) can decrease LDLR and subsequently decrease cholesterol accumulation in pancreatic beta-cells, which impairs glucose metabolism and reduces insulin secretion. Thus, a possible adverse effect of PCSK9 inhibitors on carbohydrate metabolism may be expected by this mechanism, which has been supported by the mendelian studies results. On the other hand, clinical data have suggested a detrimental association of PCSK9 with glucose metabolism. So, the inhibition of PCSK9 may be seen as a double-edged sword regarding carbohydrate metabolism. Completed clinical trials have not shown a detrimental effect of PCSK9 inhibitors on diabetes risk, but their short-term duration does not allow definite conclusions.

Role of fetuin A in the diagnosis and treatment of joint arthritis

Osteoarthritis is a slowly progressive disease which includes the intervention of several cytokines, macrophage metalleinoproteinases reaction, leading to the degradation of the local cartilage but also having an impact on the serum acute phase proteins(APPs). Subsequently, biomarkers seem to be essential to estimate its progression and the need for any surgical intervention such as total arthroplasty, but also can be used as therapeutic agents. Recently, among APPs, fetuin A drew attention regarding its possible anti-inflammatory role in animal models but also as a therapeutic agent in the inflammatory joint disease in clinical trials. In contrast with other APPs such as C-reactive protein, fetuin A appears to be lower in the serum of patients with degenerative joint disease in comparison with the healthy ones, and also acts as an antagonist of the anti-proliferative potential of transforming growth factor-b(TGF-b) cytokines. Because of its lower serum levels in arthritis, an unregulated binding of TGF-b and bone morphogenetic proteins takes place leading to further arthritic lesions. The purpose of the present review is to assess the current evidence regarding the multipotent role of the alpha-2-HSglycoprotein or as also known Fetuin-a in animal models but also as a biomarker of the degenerative joint arthritis in clinical trials.

Quality and Sensory Properties of Reduced Fat Yoghurt Made with Addition of <i>β</i>-Glucans

β-Glucans, isolated from Pleurotus citrinopileatus mushroom, were used in the manufacture of yoghurt from reduced fat cow milk with added proteins. β-Glucans were added to the milk in the form of a paste containing 1.21% (on a dry weight basis) of β-glucans, such that the final concentrations in the yoghurt were 0.3%, 0.4% and 0.5%. Control yoghurt was also manufactured from the same milk without the addition of β-glucans. The resultant yoghurts were examined for their physicochemical and microbial characteristics, gel formation and sensory properties and compared to the control at 1, 7, 14 and 21 days of storage. Results showed that the microbial population, pH, titratable acidity, ash and syneresis were not affected by the addition of different levels of glucans. The lowest fat, total solids and protein contents were recorded in the yoghurt containing 0.5% glucan. The yoghurt containing 0.3% β-glucan had the same viscosity as the control and the yoghurt containing 0.5% β-glucan had the lowest viscosity. Yoghurt with β-glucans received slightly higher scores for flavour, appearance and texture than the control. It was concluded that the incorporation of 0.3% or 0.4% β-glucans isolated from edible mushrooms, into the yoghurt does not affect the final product, which is similar or sensorially even better than the control.

Impact of Switch to Fosamprenavir and Addition of Lovaza<sup>®</sup>for Treatment of Hypertriglyceridemia in HIV-Infected Subjects on Antiretroviral Therapy

Background: Managing hypertriglyceridemia in HIV-infected patients often requires multiple pharmacologic strategies. Many protease inhibitors (PIs), one of 6 classes of drugs used to treat HIV, have been associated with hypercholesterolemia and drug interactions. For this study, we examined a dual strategy to manage hypertriglyceridemia in HIV-infected patient taking PIs: 1) switching patients to fosamprenavir (FPV), a PI with fewer drug interactions, and 2) adding prescription fish oil (LOVAZA?), which has been shown to reduce triglycerides. Methods: This multicenter, 24-week study enrolled 36 patients virologically suppressed (HIV-1 RNA <50 copies/mL) on PI-containing therapy with screening triglyceride levels of 200 - 1200 mg/dL and LDL cholesterol levels ≤160 mg/dL. At baseline, patients were switched to ritonavir (RTV)-boosted fosamprenavir (FPV 1400 mg/RTV 100 mg QD) and any lipid-lowering agents were stopped. At Week 6, LOVAZA 4 g QD was added. Results: Five patients prematurely discontinued due to adverse events (2), non-compliance, lost-to-follow up, and protocol violation. Median triglyceride concentration was 303 mg/dL at screening, 262 mg/dL at baseline, 290 mg/dL at Week 6 (+8% from baseline), and 218 mg/dL at Week 24 (–30% from Week 6). At Week 24, 39% (12/31) of patients had triglycerides <200 mg/dL. Among patients reaching Week 24, 100% (31/31) and 90% (28/31) had HIV-1 RNA <400 and <50 copies/mL, respectively. Conclusions: In this study, a switch to FPV/RTV followed by LOVAZA decreased median triglyceride levels and modestly increased the percentage of patients with triglyceride levels <200 mg/dL while maintaining virologic suppression in HIV-infected subjects with hypertriglyceridemia. Our data suggest that baseline PI may affect the likelihood of achieving triglycerides <200 mg/dL after 18 weeks on study. A larger study would be needed to understand the relative contributions of choice of protease inhibitor and LOVAZA to triglyceride concentration

Background. Mutational activation of KRAS and BRAF protooncogenes contributes to the development of many hu man cancers. Current research on gynecological cancer and specifically in cervical and endometrial cancer is focused on the mechanisms of their mutational activation. Objectives. In view of the paucity of data on their mutation frequency and the status of BRAF in these two types of gynecological cancer, we performed a systematic molecular study in 114 clinically and histologically well- defined malignant tumors of uterine cervix and endometrium and correlated the mutation status of KRAS and BRAF with the age at diagnosis and with tumor grade, stage or histological type. Methods. Direct sequence analysis of the PCR products of KRAS and BRAF genes was used to screen for known activating mutations. Results. In 67 cases of endometrial cancer, six KRAS mutations (8.9% ) were found, four at codon 12 (5.9% ) and two at codon 13 (2.9% ),while no mutation was detected at codon 61. Most of the mutations occurred in surgical stage I and in the endometrioid adenocarcinoma subtype. We also detected three KRAS point mutations (6.3% ) in the 47 cervical cancer samples, two at codon 12 (4.2% ) and one at codon 13 (2.1% ), while there was no mutation at codon 61. On the contrary,nomutation was identified inBRAF exon 15 for either endometrial or cervical cancer samples at position V600, which represents the most frequently mutated site of BRAF in human cancer. There was no association between KRAS mutations with either histological type, tumor grade or stage. Interestingly, however, KRAS mutation status in endometrial cancer was strongly associated with increased age at diagnosis (P < 0.001). Conclusions. Our data document (a) the absence of BRAF mutations in cervical and endometrial cancer, despite the mutation status of KRAS, (b) suggest that KRAS mutations reflect an early event in endometrial carcinogenesis and (c) imply that BRAF activation is involving alternative pathway
