Utility of plasma D-dimer for diagnosis of venous thromboembolism after hepatectomy

BACKGROUND Venous thromboembolism(VTE)is a potentially fatal complication of hepatectomy.The use of postoperative prophylactic anticoagulation in patients who have undergone hepatectomy is controversial because of the risk of postoperative bleeding.Therefore,we hypothesized that monitoring plasma D-dimer could be useful in the early diagnosis of VTE after hepatectomy.AIM To evaluate the utility of monitoring plasma D-dimer levels in the early diagnosis of VTE after hepatectomy.METHODS The medical records of patients who underwent hepatectomy at our institution between January 2017 and December 2020 were retrospectively analyzed.Patients were divided into two groups according to whether or not they developed VTE after hepatectomy,as diagnosed by contrast-enhanced computed tomography and/or ultrasonography of the lower extremities.Clinicopathological factors,including demographic data and perioperative D-dimer values,were compared between the two groups.Receiver operating characteristic curve analysis was performed to determine the D-dimer cutoff value.Univariate and multivariate analyses were performed using logistic regression analysis to identify significant predictors.RESULTS In total,234 patients who underwent hepatectomy were,of whom(5.6%)were diagnosed with VTE following hepatectomy.A comparison between the two groups showed significant differences in operative time(529 vs 403 min,P=0.0274)and blood loss(530 vs 138 mL,P=0.0067).The D-dimer levels on postoperative days(POD)1,3,5,7 were significantly higher in the VTE group than in the non-VTE group.In the multivariate analysis,intraoperative blood loss of>275 mL[odds ratio(OR)=5.32,95%confidence interval(CI):1.05-27.0,P=0.044]and plasma D-dimer levels on POD 5≥21μg/mL(OR=10.1,95%CI:2.04-50.1,P=0.0046)were independent risk factors for VTE after hepatectomy.CONCLUSION Monitoring of plasma D-dimer levels after hepatectomy is useful for early diagnosis of VTE and may avoid routine prophylactic anticoagulation in the postoperative period.

自1970年代以来,新自由主义伴随着全球化的进程从欧美向全球蔓延,日本的城市政策也深受其影响。本文首先提出一个假说,即日本新自由主义的城市政策造成了东京的极化与分化现象。通过国土尺度的分析,笔者观察到自2000年新自由主义成为日本主流城市政策后,东京的单极化趋势明显加速;城市尺度的空间与政策分析结果则反映了东京内部的城市分化日益显著。本文的分析检验并支撑了本文所提假说的有效性,同时证明了东京都政府和各区政府的城市政策与区域绅士化之间存在很强的相关性。本文呼吁,城市规划相关政策应从新自由主义式转向区域协作型,并从大规模城市再开发导向转变为渐进式城市更新导向。在后新自由主义的当下,本文为中国规划理论的范式转型提供了一种路径思考。

Porphyromonas gingivalis,a periodontal pathogen,impairs post-infarcted myocardium by inhibiting autophagosome–lysosome fusion

While several previous studies have indicated the link between periodontal disease (PD) and myocardial infarction (MI), theunderlying mechanisms remain unclear. Autophagy, a cellular quality control process that is activated in several diseases, includingheart failure, can be suppressed by Porphyromonas gingivalis (P.g.). However, it is uncertain whether autophagy impairment byperiodontal pathogens stimulates the development of cardiac dysfunction after MI. Thus, this study aimed to investigate therelationship between PD and the development of MI while focusing on the role of autophagy. Neonatal rat cardiomyocytes(NRCMs) and MI model mice were inoculated with wild-type P.g. or gingipain-deficient P.g. to assess the effect of autophagyinhibition by P.g. Wild-type P.g.-inoculated NRCMs had lower cell viability than those inoculated with gingipain-deficient P.g. Thisstudy also revealed that gingipains can cleave vesicle-associated membrane protein 8 (VAMP8), a protein involved in lysosomalsensitive factor attachment protein receptors (SNAREs), at the 47th lysine residue, thereby inhibiting autophagy. Wild-type P.g.-inoculated MI model mice were more susceptible to cardiac rupture, with lower survival rates and autophagy activity thangingipain-deficient P.g.-inoculated MI model mice. After inoculating genetically modified MI model mice (VAMP8-K47A) with wildtype P.g., they exhibited significantly increased autophagy activation compared with the MI model mice inoculated with wild-typeP.g., which suppressed cardiac rupture and enhanced overall survival rates. These findings suggest that gingipains, which arevirulence factors of P.g., impair the infarcted myocardium by cleaving VAMP8 and disrupting autophagy. This study confirms thestrong association between PD and MI and provides new insights into the potential role of autophagy in this relationship.

Electromechanically reconfigurable plasmonic photodetector with a distinct shift in resonant wavelength

Plasmonic photodetectors have received increasing attention because their detection properties can be designed by tailoring their metal structures on surfaces without using any additional components.Reconfiguration of the plasmonic resonant state in a photodetector is relevant for various applications,including investigating in situ adaptive detection property changes,depending on the situation,and performing single-pixel spectroscopy in geometrically limited regions.However,the spectral responsivity change with conventional reconfiguration methods is relatively small.Here,we propose a plasmonic photodetector that reconfigures its spectral responsivity with electromechanical deformation instead of bias tuning.The photodetector consists of a gold plasmonic grating formed on an n-type silicon cantilever,and the spectral responsivity is reconfigured by electromechanically scanning at an incident angle to the grating on the cantilever.The photodetector exhibits peak shifts in spectral responsivity in a wavelength range from 1250 to 1310 nm after electromechanical reconfiguration.Finally,for potential future applications,we demonstrate near-infrared spectroscopy using the photodetector.This photodetector has the potential to be adopted as a near-infrared spectrometer in industrial silicon imaging systems because its structure enables subbandgap photodetection on silicon by a Schottky junction.

Enzymatic catalysis of 2,6-dimethoxyphenol by laccases and products characterization in organic solutions

2,6-Dimethoxyphenol (DMP) as a substrate was widely used in determination of laccase activity. It is surprising, however, that its catalyzed oxidation products have not been completely determined until now. Studies were thus conducted on Rhus laccase (RL) and immobilized Rhus laccase (IRL)-catalyzed oxidation reactions of 2,6-dimethoxyphenol in water-organic solvent systems. These reactions pro- ceeded well in water-(im)miscible organic solvent systems pre-saturated with water. Only one product, 3,3′,5,5′-tetramethoxy-1,1′biphenyl-4,4′-diol (TMBP), was produced by RL catalysis, and it was thor- oughly characterized by FT-IR, NMR, GC-MS, etc. A simple enzymatic mechanism of this reaction is proposed.

Alterations in cellular metabolism may contribute to tumor proliferation and survival.Upregulation of the facilitative glucose transporter(GLUT)plays a key role in promoting cancer.GLUT5 mediates modulation of fructose utilization,and its overexpression has been associated with poor prognosis in several cancers.However,its metabolic regulation remains poorly understood.Here,we demonstrated elevated GLUT5 expression in human cholangiocarcinoma(CCA),using RNA sequencing data from samples of human tissues and cell lines,as compared to normal liver tissues or a cholangiocyte cell line.Cells exhibiting highexpression of GLUT5 showed increased rates of cell proliferation and ATP production,particularly in a fructose-supplemented medium.In contrast,GLUT5 silencing attenuated cell proliferation,ATP production,cell migration/invasion,and improved epithelialemesenchymal transition(EMT)balance.Correspondingly,fructose consumption increased tumor growth in a nude mouse xenograft model,and GLUT5 silencing suppressed growth,supporting the tumor-inhibitory effect of GLUT5 downregulation.Furthermore,in the metabolic pathways of fructolysis-Warburg effect,the expression levels of relative downstream genes,including ketohexokinase(KHK),aldolase B(ALDOB),lactate dehydrogenase A(LDHA),and monocarboxylate transporter 4(MCT4),as well as hypoxia-inducible factor 1 alpha(HIF1A),were altered in a GLUT5 expression-dependent manner.Taken together,these findings indicate that GLUT5 could be a potential target for CCA therapeutic approach via metabolic regulation.

Comparing salinity tolerance in embryonic and larval development of two species of water strider, Aquarius paludum and Gerris latiabdominis （Hemiptera： Gerridae）

Water strider Aquarius paludum （Fabricius） is a cosmopolitan species colonizes mainly freshwater but occasionally brackish habitats throughout the Palearctic and Oriental regions. Water strider Gerris latiabdominis （Miyamoto） is a common species in Japan lives in temporary habitats as freshwater paddy fields. These two species often occur syntopically. We investigated differences in the developmental response to brackish water during embryonic and larval stages between the two species. Eggs were exposed to 0-1.8% NaC1 solutions within 24 h of oviposition. Larvae of G. latiabdominis were exposed to salinities of 0, 0.5%, and 0.9% from the first instar until adult emergence. Limits of NaC1 concentration for hatching were 1.3% and 1.0% for A. paludum and G. latiabdominis, respectively. The hatching rate of G. latiabdominis was lower than that ofA. paludum at salinities 〉0.9%. The period of embryonic development of G. latiabdominis was more prolonged than that ofA. paludum at a given salinity. Although the salinity tolerance of G. latiabdominis was lower than that ofA. paludum, our results suggest G. latiabdominis has the physiological capacity to expand into brackish waters. High and low salinity tolerances of A. paludum and G. latiabdominis, respectively, reflect the relatively wide range of habitat salinities utilized by A. paludum and the relatively restricted habitats preferred by G. latiabdominis. The high salinity tolerance ofA. paludum could be an important factor contributing to their cosmopolitan distribution because high tolerance to salinity means the possibility of them to be dispersed via ocean or sea to other continents and islands.

Deviation of congruent composition in Fe-Pd system

The first-principles calculation of the disordered Ll0 phase boundary for the Fe-Pd system is attempted by combining the FLAPW electronic structure total energy calculations and the cluster variation method via the cluster expansion method. The lattice vibration effects are taken into account based on the Debye-Gruneisen model within quasi-harmonic approximation. The transition temperature is reproduced with very high accuracy. However, the experimental congruent composition of disordered L10 phase that significantly deviates from 1.＂ 1 stoichiometry is not reproduced. Fulther calculations are attempted based on the phenomenological Lennard-Jones type pair potential, which is capable of introducing both tetragonality of the Ll0 ordered phase and additional configurational freedom because of the magnetic spins. The prelimi- nary calculations indicate that the tetragonality enhances the stability of the L10 ordered phase and the magnetic contributions also change the transition temperature. Despite these findings, the shift of the congruent composition still remains as a subject that needs further research. The electronic origin of the shifting of the congruent composition is briefly discussed.

Enzymatic synthesis of bioactive compounds by Rhus laccase from Chinese Rhus vernicifera

A simple one step synthesis of pinoresinol and its derivatives-active components of Du-Zhong(Eu-commia ulmoides) -from coniferyl alcohol and p-courmaryl alcohol with higher yields was achieved by Rhus laccases(RL) catalysis in water miscible organic solvents. Biomacromolecules dehydro-genative polymers(DHP) were only synthesized by fungal laccases,not by RL. The structures and the reaction mechanism were discussed to promote the understanding of the function of laccases in the process of lignin biosynthesis.
