Object.ve: t. det.rmine if current.peer-reviewed lit.rat.re support. t.e exist.nce of a relat.onship bet.een t.e administ.at.on of diet.ry supplement. cont.ining 5-Aminolevulinic Acid (5-ALA) and sleep. Met.ods: Concept.al analysis of a syst.mat.cally defined group of dat. is based on publicat.ons in referred journals. Const.nt.comparat.ve analyses were used t. analyze t.e dat.. Det.iled analyses were used t. ident.fy t.ends t.at.led t. qualifying relat.onships bet.een t.e use of 5-ALA and heme. In addit.on, t.e relat.onship among heme, melat.nin, and sleep were explored. Since t.ere were no published dat. relat.d t. 5-ALA and sleep, an analysis of what.is known about.5-ALA and heme was undert.ken. Result.: Current.y, wit.in t.e scope of t.is review, no lit.rat.re available direct.y connect.ng 5-ALA and sleep was found. Research indicat.s t.at.5-ALA has an indirect.relat.onship wit. int.a-cellular energy product.on, which could result.in a wide range of effect. fromcellular t. endocrine t. neurologic t. behavioral. In addit.on, t.ere is evidence t.at.5-ALA may influence t.ypt.phan and serot.nin levels. Conclusion: While t.ere are no st.dies on 5-ALA and sleep were found, current.research suggest. several pot.nt.al mechanisms by which 5-ALA could influence sleep. t.e mechanisms suggest.t.e pot.nt.al for 5-ALA t. assist.in t.e adjust.ent.of a person’s circadian cycle, endocrine funct.on, or neurologic funct.on in order t. allow for bet.er sleep in a nat.ral way. In doing so, it.may provide for a safer alt.rnat.ve t. current.y available sleep medicat.on. Furt.er research is needed t. explore t.is possibilit..

Object.ve: t. det.rmine if t.ere is a relat.onship bet.een t.e administ.at.on of t.e diet.ry supplement.cont.ining 5-Aminolevulinic Acid (5-ALA) and sleep. Met.ods: A double-blind, randomized parallel-group st.dy was conduct.d. It.was a 4-mont. st.dy of 40 part.cipant. bet.een t.e ages of 40 and 70. Males and females were recruit.d equally. t.ere were 20 in each group who had exist.ng sleep disorders. t.e t.ol used t. measure part.cipant.sleep improvement.included t.e Pit.sburgh Insomnia Rat.ng Scale-20 Quest.on (PIRS-20). Improved sleep is reflect.d when t.e t.t.l PIRS-20 score is lower. Result.: Improvement.in sleep in t.e group t.king 50 mg 5-ALA, compared t. cont.ols, was significant. t.e mean change, from baseline t.rough week 6, was -5.67 unit. less on t.e sleep scale t.an t.e cont.ol group wit. a p value of 0.001. t.e mean change from week 6 t. week 10 when t.e part.cipant.was no longer t.king t.e supplement.was 4.55 unit. higher t.an t.e cont.ol group wit. a p value of 0.062, which is of borderline significance. Conclusions: t.ere is a relat.onship bet.een t.e administ.at.on of diet.ry supplement. cont.ining 5-ALA and sleep. t.e result. of t.is st.dy suggest.t.at.5-ALA is associat.d wit. improved sleep. t.e mechanism for sleep improvement.needs t. be explored. Furt.er research is warrant.d.

Problem: 5-ALA has been used for many years at.relat.vely high dose amount. in single doses for phot.dynamic t.erapy and immunofluorescence of t.mors. An analysis of compiled dat. relat.ng t. safet. and any side-effect. about.t.e use of 5-ALA at.low doses has not.yet.been published. Purpose: t.is report.analyzes dat. about.t.e safet. of t.e use of 5-Aminolevulinic Acid (5-ALA) in low doses as a supplement.over an ext.nded period of t.me. Met.ods: t.is invest.gat.on is a syst.mat.c analysis of t.e current.lit.rat.re ((Medline, and SBI) and snowballing t.chniques) relat.d t. t.e safet. and efficacy of 5-ALA in animals and humans. Clinical t.ials in progress using 5-ALA were also analyzed. Const.nt.comparat.ve analyses were used t. synt.esize t.e findings. Result.: t.e safet. of low-dose 5-ALA as a supplement.has been demonst.at.d by animal and human st.dies. t.e result. suggest.t.at.none of t.e invest.gat.ons document.t.e presence of sympt.ms or abnormal laborat.ry result. of clinical significance. t.e minor laborat.ry changes document.d were judged not.clinically significant.

t.e ext.act.from cult.re of effect.ve microorganisms (ECEM) is a secondary met.bolit. produced in symbiot.c cult.re of a mixt.re of yeast. and lact.c bact.ria followed by phot.synt.et.c bact.ria. Met.bolomics st.dies have demonst.at.d t.at.ECEM cont.ined several act.ve ingredient. associat.d wit. ant.-inflammat.on t.rough int.ract.ons wit. nuclear fact.r-κB. Funct.onal genomics using DNA microarrays revealed t.at.ECEM had dist.nct.ve ant.-inflammat.ry and immunost.mulat.ry act.ons in RAW264.7 cells. ECEM inhibit.d t.e LPS-upregulat.d expression of one group of pro-inflammat.ry genes (iNos, Il6 and Cox2) but.not.anot.er group genes (t.f, Ccl7 and Dusp2);it.also upregulat.d t.e expression of Cd1d1, which part.cipat.s in ant.gen present.t.on on lipids, result.ng in act.vat.on of nat.ral killer t.(NKt. cells relat.d t. infect.on defense and cancer. Bact.rial glycosylat.d lipid and ot.er relat.d complex lipids in ECEM are t.ought.t. t.igger t.is upregulat.on of Cd1d1 expression. t.ese result. suggest.t.at.ECEM funct.ons in t.e immune response in bot. inflammat.on and ant.gen present.t.on and t.at.it.cont.ins component. import.nt.for t.ese funct.ons. t.ese novel effect. of ECEM could be invaluable for developing funct.onal drinks or healt. beverages.

Our previous research showed t.at.aliphat.c amines were put.in order of high react.vit. as “et.ylamine > ammonia > t.but.lamine > diet.ylamine” on t.e aromat.c nucleophilic subst.t.t.on of 1-dimet.lamino-2,4-bis(t.ifluoroacet.l)-napht.alene 1 in acet.nit.ile. t.e DFt.calculat.on st.dy (B3LYP/6-31G* wit. solvat.on model) for t.e react.ons of 1 wit. above four amines rat.onally explained t.e difference of each amines react.vit. based on t.e energies of t.eir Meisenheimer complexes 3 which are assumed t. formed as t.e react.on int.rmediat.s in t.e course of t.e react.on giving t.e corresponding N-N exchange product. 2. Int.amolecular hydrogen bond bet.een amino prot.n in 1-amino group and carbonyl oxygen in 2-t.ifluoroacet.l group st.bilizes Meisenheimer complexes 3 effect.vely, and accelerat.s t.e subst.t.t.on react.on from 1 t. 2. Our calculat.on result. also predict.d t.at.t.e above order of amines is also t.ue if less polar t.luene is used as a solvent.inst.ad of acet.nit.ile even t.ough more enhanced condit.ons are required.
