Dosimetric Advantages of Volumetric Modulated Arc Therapy Based Coronal Arc Delivery Technique in Brain Stereotactic Radiosurgery: A Feasibility Study

The feasibility of a volumetric modulated arc therapy (VMAT) based coronal arc (cARC) technique for treating a single brain metastasis or lesion proximal to the brainstem or optic chiasm was evaluated. Coplanar (Cp) and non-coplanar (NCp) treatment plans to an anthropomorphic head/neck phantom scanned head-first supine were compared to a cARC plan with the phantom rotated vertically. A set of planning target volumes (pTVs) were contoured centrally between the brainstem and optic chiasm (“Ant pTVs”) and posterior to brainstem (“post pTVs”). Dosimetric indices such as conformity index (C.I.), gradient measure (G.M.), and dose volume histograms (DVHs) were compared for Cp, NCp and cARC techniques. The TG101 guidelines for organs-at-risk (OARs), and 95% of pTV receiving at least 100% of the prescription dose (D95 = 100%) were used as plan objectives. Reductions in D50 and D30 to the brainstem of 85.1% ± 3.9% and 87.6% ± 3.2%, respectively were seen for “post pTVs”, and 51.1% ± 17.8% and 85.6% ± 6.0% respectively for “Ant pTVs” using cARC versus Cp (p ≤ 0.01). For chiasm, reductions of D50 and D30 were 61.7% ± 3.2% and 44.2% ± 8.9% for “Ant pTVs”, by 69.3% ± 8.0% and 74.3% ± 8.2% for “post pTVs” (p ≤ 0.01). Comparing cARC to NCp led to similar dosimetric improvements. The conformity index (C.I.) was measured to be 1.101 ± 0.038, 1.088 ± 0.054, and 1.060 ± 0.040 for cARC, Cp and NCp respectively (p ≤ 0.01). The overall GM in cm was 0.581 ± 0.097, 0.708 ± 0.064, and 0.476 ± 0.050 for cARC, Cp and NCp respectively (p ≤ 0.01). The mean distance gradient fall-off (in cm) was 0.249 ± 0.038 (cARC), 0.749 ± 0.107 (Cp), and 0.621 ± 0.068 (NCp) at the center slice in anterior-posterior direction of the target volume (p ≤ 0.01). The objective of this study is to compare the dosimetric indices of cARC with Cp and NCp techniques. In conclusion, cARC can provide improved dosimetry as compared to Cp and NCp for lesion proximal to the brainstem or optic chiasm.

Serotonin uptake rates in platelets from angiotensin II-induced hypertensive mice

Angiotensin II (Ang II) is a critical component of the reninangiotensin system that contributes to hypertension. Although platelets in blood from hypertensive subjects have an abnormal biological profile, it is unclear if circulating Ang II influences platelet aggregation or thrombus formation. One of the abnormalities presented to the platelets during hypertension is an elevated plasma concentration of serotonin (5-HT) caused by reduced 5-HT uptake secondary to loss of the 5-HT transporter (SERT) on the platelet plasma membrane. In the current study, we evaluated in vivo platelet function after 7 days of subcutaneous Ang II infusion to establish hypertension in mice and additionally assessed the biology of isolated platelets exposed to Ang II in vitro. The administration of Ang II elevated systolic blood pressure, but markers of platelet activation including p-selectin and pEJon/A staining were not changed. However, the aggregation response to collagen was reduced in isolated platelets from Ang II-infused mice, which also showed reduced 5-HT uptake by SERT. In vitro exposure of isolated platelets to Ang II also resulted in a loss of surface SERT associated with a reduced aggregation response to collagen. These abnormalities were reversed by increasing concentra tions of the Ang II receptor antagonist, valsartan. Interestingly, SERT KO mice failed to fully develop hypertension in response to Ang II infusion and isolated platelets from these animals were insensitive to the anti-aggregatory influence of Ang II. Thus, Ang II blunts the aggregation responses of platelets and the mechanism underlying this action may involve a loss of SERT on the platelet plasma membrane. The latter event depletes intracellular 5-HT in platelets, an event that is associated with reduced aggregation. Influence of the coupling agent on microstructure and dielectric properties of ceramic-polymer composites is systematically studied using CaCu_(3)Ti_(4)O_(12)(CCTO)as the filler,trichloro-(1H,1H,2H,2H-perfluorooctyl)-silane(Cl_(3)-silane)as coupling agent,and p(VDF-CTFE)88/12 mol.%copolymer as the *** is demonstrated that Cl_(3)-silane molecules can be attached onto CCTO surface using a simple *** experimental results show that coating CCTO with Cl_(3)-silane can improve the microstructure uniformity of the composites due to the good wettability between Cl_(3)-silane and p(VDF-CTFE),which also significantly improves the electric breakdown field of the *** is found that the composites using CCTO coated with 1.0 wt.%Cl_(3)-silane exhibit a higher dielectric constant with a higher electric breakdown *** the composites with 15 vol.%CCTO that is coated with 1.0 wt.%Cl_(3)-silane,an electric breakdown field of more than 240 MV/m is obtained with an energy density of more than 4.5 J/cm^(3).It is also experimentally found that the dielectric constant can be used to easily identify the optimized content of coupling agent.

Coactivation Does Not Contribute to Fatigue-Induced Decreases in Isokinetic Forearm Flexion and Extension Torque

Constitutive renal Rel/nuclear factor-κB expression in Lewis polycystic kidney disease rats

AIM： To determine the temporal expression and pattern of Rel/nuclear factor （NF）-κB proteins in renal tissue in polycystic kidney disease （pKD）. METHODS： The renal expression of Rel/NF-κB proteins was determined by immunohistochemistry, immunofuorescence and immunoblot analysis in Lewis polycystic kidney rats （LpK, a genetic ortholog of human nephronopthsis-9） from postnatal weeks 3 to 20. At each timepoint, renal disease progression and the mRNA expression of NF-κB-dependent genes （TNFa and CCL2） were determined. NF-κB was also histologically assessed in human pKD ***： progressive kidney enlargement in LpK rats was accompanied by increased renal cell proliferation and interstitial monocyte accumulation （peaking at weeks 3 and 10 respectively）, and progressive interstitial fibrosis （with a smooth muscle actin and Sirius Red deposition significantly increased compared to Lewis kidneys from weeks 3 to 6 onwards）. Rel/NF-κB proteins （phosphorylated-p105, p65, p50, c-Rel and RelB） were expressed in cystic epithelial cells （CECs） of LpK kidneys as early as postnatal week 3 and sustained until late-stage disease at week 20. From weeks 10 to 20, nuclear p65, p50, RelB and cytoplasmic IκBa protein levels, and TNFa and CCL2 expression, were upregulated in LpK compared to Lewis kidneys. NF-κB proteins were consistently expressed in CECs of human pKD. The DNA damage marker γ-H2AX was also identifed in the CECs of LpK and human polycystic kidneys. CONCLUSION： Several NF-κB proteins are consistently expressed in CECs in human and experimental pKD. These data suggest that the upregulation of both the canonical and non-canonical pathways of NF-κB signaling may be a constitutive and early pathological feature of cystic renal diseases.
