Epigenomic mediation after adverse childhood experiences:a systematic review and meta-analysis

Epigenetic mechanisms are potential mediators of the physiological response to abuse by altering the genetic predisposition of the cellular response to the environment,leading to changes in the regulation of multiple organ *** study was established to review the epigenetic mechanisms associated with childhood abuse as well as the long-term deter-minants that these epigenetic changes may have on future *** retrospectively ana-lysed the effect of exposure to adverse childhood experiences(ACEs,specifically those relating to childhood maltreatment)between the ages of 0 and 16years on the human epi-genome,as well as possible clinical *** meeting inclusion and exclusion crite-ria,36 articles were included in this systematic *** of these studies did not find a relationship between childhood maltreatment and DNA *** the remaining 28 studies,nine were genome-wide association studies,whereas the rest were candidate gene studies,mainly studying effects on neuroendocrine,serotoninergic and immunoregulatory ***-analysis of correlation coefficients from candidate gene studies estimated an association of childhood adversity and DNA methylation variation at r=0.291(P<0.0001),and meta-analysis of two epigenome-wide association studies(EWASs)identified 44 differen-tially methylated CpG *** conclusion,childhood maltreatment may mediate epigenetic mechanisms through DNA methylation,thereby affecting physiological responses and con-ferring a predisposition to an increased risk for psychopathology and forensic *** evidence for somatic illnesses is not yet available.

Assessment of aerobic biodegradation of lower-chlorinated benzenes in contaminated groundwater using field-derived microcosms and compound-specific carbon isotope fractionation

Biodegradation of lower chlorinated benzenes(tri-, di-and monochlorobenzene) was assessed at a coastal aquifer contaminated with multiple chlorinated aromatic hydrocarbons. Field-derived microcosms, established with groundwater from the source zone and amended with a mixture of lower chlorinated benzenes, evidenced biodegradation of monochlorobenzene(MCB) and 1,4-dichlorobenzene(1,4-DCB) in aerobic microcosms,whereas the addition of lactate in anaerobic microcosms did not enhance anaerobic reductive dechlorination. Aerobic microcosms established with groundwater from the plume consumed several doses of MCB and concomitantly degraded the three isomers of dichlorobenzene with no observable inhibitory effect. In the light of these results, we assessed the applicability of compound stable isotope analysis to monitor a potential aerobic remediation treatment of MCB and 1,4-DCB in this site. The carbon isotopic fractionation factors(ε) obtained from field-derived microcosms were-0.7‰ ± 0.1 ‰ and-1.0‰ ± 0.2 ‰ for MCB and1,4-DCB, respectively. For 1,4-DCB, the carbon isotope fractionation during aerobic biodegradation was reported for the first time. The weak carbon isotope fractionation values for the aerobic pathway would only allow tracing of in situ degradation in aquifer parts with high extent of biodegradation. However, based on the carbon isotope effects measured in this and previous studies, relatively high carbon isotope shifts(i.e., Δδ13C > 4.0 ‰) of MCB or 1,4-DCB in contaminated groundwater would suggest that their biodegradation is controlled by anaerobic reductive dechlorination.

Metrics for macroscale invasion and dispersal patterns

Aim The ability to quantitatively measure the continuum of macroscale patterns of species invasion is a first step toward deeper understand-ing of their causal *** took advantage of two centuries worth of herbarium data,to evaluate a set of metrics to measure macroscale patterns,allowing cross-species comparisons of inva-sive expansion across large geographic *** We used herbarium specimens to reconstruct county-level invasion histories for two non-native plants(Alliaria petiolata and Lonicera japonica),with distinct spatiotemporal distribution patterns over the past two *** county centroids from species’ini-tial occurrences,we quantified point pattern metrics from multiple disciplines(*** crime analysis,landscape ecology etc.)that are historically used at smaller spatial scales,to evaluate their abil-ity to detect macroscale spatial diffusion and amount of directional *** were further assessed for their ease of use,data requirements,independence from other metrics and intuitiveness of *** Findings We identified four suitable metrics for distinguishing differences in spatial patterns:(i)standard distance,(ii)number of patches,(iii)Euclidean nearest neighbor summary class statistic coefficient of variation and(iv)mean center that when applied to county-level presence data allowed us to determine the directions by which distributions expanded and if distributions increased via outward expansion,infilling and/or jump dispersal *** metrics when compared during the same invasion phase are capable of quantifying macroscale variability among species in their distri-butional and dispersal *** able to quantify differences among species in these patterns is important in understanding the drivers of species dispersal *** metrics therefore repre-sent a simple yet thorough toolset for achieving this goal.

Efficacy and safety of XEN 45 gel stent alone or in combination with phacoemulsification in advanced open angle glaucoma patients: 1-year retrospective study

AIM:To assess the effectiveness of the XEN 45 gel stent,either alone or combined with cataract surgery,in advanced stage open angle glaucoma(OAG)***:Retrospective and single-center study conducted on consecutive OAG patients who underwent a XEN 45 gel stent implantation surgery,between July 2017 and September *** primary efficacy end-point was the mean intraocular pressure(IOP)reduction at the end of the follow-up *** was defined as an IOP reduction of at least 20%and an IOP value≤18 mm Hg without(complete)or with(qualified)hypotensive ***:Seventy-four patients(80 eyes)were included in the *** the overall study sample,XEN implant significantly reduced IOP from 21.0(19.8 to 22.1)mm Hg at baseline to 9.3(8.2 to 10.4),10.7(9.6 to 11.9),13.4(12.2 to 14.7),14.5(13.6 to 15.4),14.7(13.8 to 15.6),and 14.7(13.9 to 15.4)mm Hg at 1 d,1 wk,1,3,6,and 12 mo of follow-up,respectively(P<0.0001 each).In the overall study population,at the end of the study the mean IOP reduction was 27.4%(23.3%to 31.5%).Adjusted IOP reduction was similar in XEN and XEN+phacoemulsification groups[30.0(23.4 to 36.4)mm Hg vs 24.8(18.4 to 31.2)mm Hg,respectively,P=0.2939].At the last follow-up visit,52(65.0%)eyes were considered success,29(36.3%)eyes as complete success and 23(28.7%)as qualified *** number of hypotensive medications was significantly reduced from 2.8(2.7 to 3.0)at baseline to 1.1(0.8 to 1.3),P<***-Meier survival analysis did not find any difference in the success rate between XEN and XEN+PHACO,mean hazard ratio 0.56,95%CI 0.26 to 1.23;P=*** was performed in 7(8.8%)eyes at months 1(n=3);3(n=2);4(n=1)and 11(n=1).Eleven(13.8%)eyes presented adverse ***:XEN implant,either alone or in combination with phacoemulsification,significantly reduced the IOP and the number of hypotensive medications in patients with OAG in advanced stage.

Differential regulation of protein synthesis in skeletal muscle and liver of neonatal pigs by leucine through an mTORC1-dependent pathway

Neonatal growth is characterized by a high protein synthesis rate that is largely due to an enhanced sensitivity to the postprandial rise in insulin and amino acids, especially leucine. The mechanism of leucine＇s action in vivo is not well understood. In this study, we investigated the effect of leucine infusion on protein synthesis in skeletal muscle and liver of neonatal pigs. To evaluate the mode of action of leucine, we used rapamycin, an inhibitor of mammalian target of rapamycin （mTOR） complex-1 （mTORC1）. Overnight-fasted 7-day-old piglets were treated with rapamycin for 1 hour and then infused with leucine （400 μmol·kg^-1·h^-1） for 1 hour. Leucine infusion increased the rate of protein synthesis, and ribosomal protein S6 kinase 1 （S6K1） and eukaryotic initiation factor （elF） 4E-binding protein-1 （4E-BP1） phosphorylation in gastrocnemius and masseter muscles （P 〈 0.05）, but not in the liver. The leucine-induced stimulation of protein synthesis and S6K1 and 4E-BP1 phosphorylation were completely blocked by rapamycin, suggesting that leucine action is by an mTORC1-dependent mechanism. Neither leucine nor rapamycin had any effect on the activation of the upstream mTQRC1 regulators, AMP-activated protein kinase and protein kinase B, in skeletal muscle or liver. The activation of elF2α and elongation factor 2 was not affected by leucine or rapamycin, indicating that these two pathways are not limiting steps of leucine-induced protein synthesis. These results suggest that leucine stimulates muscle protein synthesis in neonatal pigs by inducing the activation of mTORC1 and its downstream pathway leading to mRNA translation.

Knowledge of measured energy expenditure(EE)during walking and running is important for exercise ***,research on the EE comparison and EE predicted equation during walking or running among different ethnicities is *** purpose of the current study was to compare EE to walk or run 1 mile in Caucasian,African American and Asian adults and to develop a regression equation to predict EE to walk or run 1 *** hundred and twenty-four participants were included(71 Caucasians,68 African Americans and 85 Asians)with 3 groups(normal weight walking,overweight walking and running).EE was measured via indirect *** of variance was used to compare EE across *** regression analysis was employed for EE prediction,and the prediction equation was cross-validated.A significant EE difference was found between walking and running among three *** prediction equation was:EE=0.978 Body Weight-4.571 Gender(male=1;female=2)+3.524 Ethnicities(Caucasians=1,African Americans=2,Asians=3)+32.447(standard error of estimate=12.5 kcal·mile^(-1)).The equation was valid through cross-validation,so it is recommended to apply for calculating EE during walking or running 1 mile among Cauca-sians,African Americans and Asians.

Breast cancer can infrequently metastasize to the upper gastrointestinal(GI)tract but the exact incidence is not well established-there is considerable variation between incidence reported from clinical studies and incidence noted in autopsy *** presentation can be very non-specific and often mimics primary gastrointestinal *** alone may not be sufficient to make a diagnosis and misdiagnosis is also common.A high degree of awareness and clinical suspicion is required to establish metastases to the upper GI *** undertook a comprehensive review of the available literature on breast cancer metastases to the esophagus and stomach including the clinical symptoms and presentation,endoscopic features,additional diagnostic imaging modalities,treatment and outcomes.

The unclear role of VEGF in POEMS syndrome: therapeutic implications of neoangiogenesis in a rare plasma cell disorder

POEMS syndrome is a rare paraneoplastic disorder due to an underlying clone of aberrant plasma *** name POEMS is an acronym for some of the major disease manifestations,namely polyneuropathy,organomegaly,endocrinopathy,presence of monoclonal component,and skin *** clinical presentation can be various and could lead to delayed diagnosis and *** is known about the pathogenic mechanism,although the neoangiogenesis due to overproduction of vascular endothelial growth factor(VEGF)by plasma cells seems to play a key *** latest evidence suggests that the blood concentration of this cytokine correlates with the activity of the syndrome:VEGF could then be used as a therapeutic target and a marker to monitor *** case reports have shown the efficacy of this approach,but extended studies are required to better define the use of anti VEGF in patients affected by POEMS syndrome.

This study investigates the development of novel montmorillonite/castor oil blends to formulate sustainable lubricating greases to promote the replacement of petrochemicals industry-derived materials by substances obtained from renewable ***,the effect of the thickener concentration on the rheological,chemical,thermal,tribological properties,and atomic force microscopy(AFM)microstructure of these systems were *** results showed that the C20A nanoclay content could be used to modulate the viscosity values,the linear viscoelastic functions,and tribological properties of these montmorillonite *** general,these gel-like dispersions exhibited remarkable lubricant properties;the samples showed values of the friction coefficient and wear scars similar or lower than those obtained with model bentonite grease.
