Wet adhesive hydrogel cardiac patch loaded with anti-oxidative,autophagy-regulating molecule capsules and MSCs for restoring infarcted myocardium

Hydrogel patch-based stem cell transplantation and microenvironment-regulating drug delivery strategy is promising for the treatment of myocardial infarction(MI).However,the low retention of cells and drugs limits their therapeutic efficacies.Here,we propose a prefixed sponge carpet strategy,that is,aldehyde-dextran sponge(ODS)loading anti-oxidative/autophagy-regulating molecular capsules of 2-hydroxy-β-cyclodextrin@resveratrol(HP-β-CD@Res)is first bonded to the rat’s heart via capillary removal of interfacial water from the tissue surface,and the subsequent Schiff base reaction between the aldehyde groups on ODS and amino groups on myocardium tissue.Then,an aqueous biocompatible hydrazided hyaluronic acid(HHA)solution encapsulating mesenchymal stem cells(MSCs)is impregnated into the anchored carpet to form HHA@ODS@HP-β-CD@Res hydrogel in situ via click reaction,thus prolonging the in vivo retention time of therapeutic drug and cells.Importantly,the HHA added to outer surface consumes the remaining aldehydes to contribute to nonsticky top surface,avoiding adhesion to other tissues.The embedded HP-β-CD@Res molecular capsules with antioxidant and autophagy regulation bioactivities can considerably improve cardiac microenvironment,reduce cardiomyocyte apoptosis,and enhance the survival of transplanted MSCs,thereby promoting cardiac repair by facilitating angiogenesis and reducing cardiac fibrosis.

The repair of cartilage injury caused by osteoarthritis(OA)has long plagued clinicians.Mesenchymal stem cell(MSC)-derived small extracellular vesicles(sEVs)show great potential in cartilage regeneration and immunoregulation to realise cell-free therapy.However,unprotected sEVs are prone to be washed away by the flow of fluids and degraded in vivo.In this work,we successfully revealed that human Wharton’s jelly of umbilical cord-derived MSC(hWJMSC)sEVs promote proliferation,inhibit apoptosis,and attenuate the inflammation of chondrocytes in vitro.In light of the long period required for cartilage regeneration,we synthesised an injectable and adhesive aldehyded sodium alginate crosslinked acylhydrazide-modified hyaluronic acid(ALG-CHO/HHA)hydrogel loaded with sEVs to boost the reparative effect of the sEVs.The hydrogel-bearing sEVs was injected into cartilage defects where they adhered to the cartilage surface.In this configuration,the sEVs were delivered in a sustainable manner with the degradation of the hydrogel at the injury sites,thus contributing to highly efficient cartilage repair by regulating the regenerative and immune microenvironment.The ALG-CHO/HHA-sEV platform meets the clinical demand for long-lasting repair with a single injection.Thus,this work provides a new idea and theoretical basis for the clinical application of sEVs in the treatment of OA.

Tea eggs-inspired high-strength natural polymer hydrogels

Natural polymer(NP)hydrogels are an irreplaceable class of biomaterials owing to their identified biosafety;however,the intrinsic poor mechanical strengths severely limit their applications as structural tissue engineering scaffolds.Inspired by the stiffening albumen gel of tea eggs,a traditional Chinese snack,high-strength NP hydrogels are constructed by simply soaking in aqueous solution of tea polyphenols(TP),an active ingredient extracted from tea.The TP-treated representative NP hydrogels exhibit considerably enhanced multifaceted mechanical properties with maximum 19-/30-,8.4-,6.1-,72-fold increases in tensile/compressive strengths,Young’s modulus,elongation at break and facture toughness,respectively,compared with pristine hydrogels,primarily due to the hydrogen bonding interactions between TP and NP chains.The TP-treated NP hydrogels can resist different large deformations,which cannot be achieved by their original species at all.In aqueous solution,the TP-treated NP hydrogels can still maintain robust mechanical performances,in spite of somewhat decline in strengths with release of TP,which just favorably affords increased water contents,antibacterial and antioxidant activities.GelMA-TP hydrogel is shown to facilitate wound healing in a full-thickness skin defect model.Importantly,the weak 3D printed GelMA scaffolds are significantly strengthened by TP treatment,broadening the possibility for customizing individualized bioscaffolds.

Effects of a Dry-Mass Conserving Dynamical Core on the Simulation of Tropical Cyclones

The accurate forecasting of tropical cyclones(TCs)is a challenging task.The purpose of this study was to investigate the effects of a dry-mass conserving(DMC)hydrostatic global spectral dynamical core on TC simulation.Experiments were conducted with DMC and total(moist)mass conserving(TMC)dynamical cores.The TC forecast performance was first evaluated considering 20 TCs in the West Pacific region observed during the 2020 typhoon season.The impacts of the DMC dynamical core on forecasts of individual TCs were then estimated.The DMC dynamical core improved both the track and intensity forecasts,and the TC intensity forecast improvement was much greater than the TC track forecast improvement.Sensitivity simulations indicated that the DMC dynamical core-simulated TC intensity was stronger regardless of the forecast lead time.In the DMC dynamical core experiments,three-dimensional winds and warm and moist cores were consistently enhanced with the TC intensity.Drier air in the boundary inflow layer was found in the DMC dynamical core experiments at the early simulation times.Water vapor mixing ratio budget analysis indicated that this mainly depended on the simulated vertical velocity.Higher updraft above the boundary layer yielded a drier boundary layer,resulting in surface latent heat flux(SLHF)enhancement,the major energy source of TC intensification.The higher DMC dynamical core-simulated updraft in the inner core caused a higher net surface rain rate,producing higher net internal atmospheric diabatic heating and increasing the TC intensity.These results indicate that the stronger DMC dynamical coresimulated TCs are mainly related to the higher DMC vertical velocity.

本文报道了一种高强度聚丙烯酸基黏附水凝胶的简便制备方法.在丙烯酸(AA)单体中溶解氢键单体2-乙烯基-4,6-二氨基-1,3,5三嗪(VDT),再加入去离子水、交联剂聚乙二醇二丙烯酸酯(PEGDA)和热引发剂过硫酸铵(APS)并混合均匀,热引发聚合即可得到氢键和化学键共交联的聚丙烯酸基水凝胶.交联剂PEGDA的加入使得水凝胶的韧性增强,疏水氢键单体VDT的引入提高了水凝胶强度和溶胀稳定性.在不需要任何外界刺激条件下,该水凝胶可对多种基材实现黏附,对猪皮和铁片基材的最大黏附强度分别达到105和99 kPa.此双交联水凝胶可迅速黏附和修补破损的膀胱,72 h内未发生模拟尿液的泄露,这种新型的高强度聚丙烯酸基黏附水凝胶有望作为一种强韧的医疗"创可贴"应用于急救领域.
