Effect of varying normal stress on stability and dynamic motion of a spring-slider system with rate- and state-dependent friction

Incorporating rate and state friction laws, stability of linearly stable （i.e., with stiffness greater than the critical value） spring-slider systems subjected to triggering perturbations was analyzed under variable normal stress condition, and comparison was made between our results and that of fixed normal stress cases revealed in previous studies. For systems associated with the slip law, the critical mag- nitude of rate steps for triggering unstable slips are found to have a similar pattern to the fixed normal stress case, and the critical velocity steps scale with a/（b - a） when k = kcr for both cases. The rate-step boundaries for the variable normal stress cases are revealed to be lower than the fixed normal stress case by 7 %-16 % for a relatively large ct = 0.56 with （b - a）/a ranging from 0.25 to 1, indicating easier triggering under the variable normal stress condition with rate steps. The difference between fixed and variable normal stress cases decreases when the α value is smaller. In the same slip- law-type systems, critical displacements to trigger instability are revealed to be little affected by the variable normal stress condition. When k 〉 kcr（V,）, a spring-slider system with the slowness law is much more stable than with the slip law,suggesting that the slowness law fits experimental data better when a single state variable is adopted. In stick-slip motions, the variable normal stress case has larger stress drops than the constant normal stress case. The variable normal stress has little effect on the range of slip velocity in systems associated with the slowness law, whereas systems associated with the slip law have a slowest slip velocity immensely smaller than the fixed normal stress case, by ~ 10 orders of magnitude.

A biosynthetic gene cluster for the bioactive fungal sesterterpenoids variecolin(1)and variecolactone(2)was identified in Aspergillus aculeatus ATCC 16872.Heterologous production of 1 and 2 was achieved in Aspergillus oryzae by expressing the sesterterpene synthase VrcA and the cytochrome P450 VrcB.Intriguingly,the replacement of VrcB with homologous P450s from other fungal terpenoid pathways yielded three new variecolin analogues(5-7).Analysis of the compounds’anticancer activity in vitro and in vivo revealed that although 5 and 1 had comparable activities,5 was associated with significantly reduced toxic side effects in cancer-bearing mice,indicating its potentially broader therapeutic window.Our study describes the first tests of variecolin and its analogues in animals and demonstrates the utility of synthetic biology for creating molecules with improved biological activities.

The rise of artifcial intelligence(AI)has brought breakthroughs in many areas of medicine.In ophthalmology,AI has delivered robust results in the screening and detection of diabetic retinopathy,age-related macular degeneration,glaucoma,and retinopathy of prematurity.Cataract management is another feld that can beneft from greater AI application.Cataract is the leading cause of reversible visual impairment with a rising global clinical burden.Improved diagnosis,monitoring,and surgical management are necessary to address this challenge.In addition,patients in large developing countries often sufer from limited access to tertiary care,a problem further exacerbated by the ongoing COVID-19 pandemic.AI on the other hand,can help transform cataract management by improving automation,efcacy and overcoming geographical barriers.First,AI can be applied as a telediagnostic platform to screen and diagnose patients with cataract using slit-lamp and fundus photographs.This utilizes a deep-learning,convolutional neural network(CNN)to detect and classify referable cataracts appropriately.Second,some of the latest intraocular lens formulas have used AI to enhance prediction accuracy,achieving superior postoperative refractive results compared to traditional formulas.Third,AI can be used to augment cataract surgical skill training by identifying diferent phases of cataract surgery on video and to optimize operating theater workfows by accurately predicting the duration of surgical procedures.Fourth,some AI CNN models are able to efectively predict the progression of posterior capsule opacifcation and eventual need for YAG laser capsulotomy.These advances in AI could transform cataract management and enable delivery of efcient ophthalmic services.The key challenges include ethical management of data,ensuring data security and privacy,demonstrating clinically acceptable performance,improving the generalizability of AI models across heterogeneous populations,and improving the trust of end-users.

We aim to evaluate the incidence of incontinence following laser endoscopic enucleation of the prostate(EEP)comparing en-bloc(Group 1)versus 2-lobe/3-lobe techniques(Group 2).We performed a retrospective review of patients undergoing EEP for benign prostaticenlargement in 12 centers between January 2020 and January 2022.Data were presented as median and interquartile range(IQR).Univariable and multivariable logistic regression analysis was performed to evaluate factors associated with stress urinary incontinence(SUI)and mixed urinary incontinence(MUI).There were 1711 patients in Group 1 and 3357 patients in Group 2.Patients in Group 2were significantly younger(68[62–73]years vs 69[63–74]years,P=0.002).Median(interquartile range)prostate volume(PV)wassimilar between the groups(70[52–92]ml in Group 1 vs 70[54–90]ml in Group 2,P=0.774).There was no difference in preoperativeInternational Prostate Symptom Score,quality of life,or maximum flow rate.Enucleation,morcellation,and total surgical time weresignificantly shorter in Group 1.Within 1 month,overall incontinence rate was 6.3%in Group 1 versus 5.3%in Group 2(P=0.12),and urge incontinence was significantly higher in Group 1(55.1%vs 37.3%in Group 2,P<0.001).After 3 months,the overall rate ofincontinence was 1.7%in Group 1 versus 2.3%in Group 2(P=0.06),and SUI was significantly higher in Group 2(55.6%vs 24.1%in Group 1,P=0.002).At multivariable analysis,PV and IPSS were factors significantly associated with higher odds of transient SUI/MUI.PV,surgical time,and no early apical release technique were factors associated with higher odds of persistent SUI/MUI.

Pteropine orthoreovirus(PRV)is an emerging zoonotic respiratory virus with the potential to spill over from fruit bats to humans.PRV belongs to the family Reoviridae,whose name is derived from an abbreviation of"respiratory enteric orphan".This family of viruses was initially not believed to cause diseases in humans and therefore was named an"orphan virus".PRV infections in humans are characterized by symptoms ranging from mild influenza-like illness to severe acute respiratory distress.

Some features of effective radius and variance of dust particles in numerical simulations of the dust climate on Mars

Airborne dust is an important constituent in the Martian atmosphere because of its radiative interaction with the atmospheric circulation.Dust size is one crucial factor in determining this effect.In reality dust sizes are varied;however,in numerical modeling of dust processes,dust size has usually been described by choice of a particular size distribution function,or by use of fixed values of effective radius(ER)and effective variance(EV).In this work,we present analytical expressions that have been derived to specify ER and EV for Nbin dust schemes,based on a model-calculated dust mixing ratio.Numerical simulations based on this approach thus would consider the effects of variable ER on the atmospheric radiation and their interaction.Results have revealed some interesting features of the dust distribution parameters,such as seasonal and spatial variation of ER and EV,which are generally consistent with some previous observational and modeling studies.Compared with the usual approach of using a fixed ER,simulation results from the present approach suggest that the variability of ER can have significant effects on the simulated thermal field of the Martian atmosphere.

One of the major obstacles to the application of potassium-ion batteries in large-scale energy storage is the lack of safe and effective electrolytes.KNH_(2),a new potassium-ion solid electrolyte has been developed in this study.Its ionic conductivity reaches 4.84×10^(-5)S cm^(-1)at 150°C and can reach3.56×10^(-4)S cm^(-1)after mechanochemical treatment.The result from electron paramagnetic resonance(EPR) measurement shows that the increment of ionic conductivity is dependent on the concentration of nitrogen defects in the KNH_(2) electrolyte.To the best of our knowledge,this is the first report that adopts inorganic amide as an electrolyte for potassium-ion battery and initiates the search for a new amidebased solid electrolyte for an all-solid-state potassium-ion battery.

Emerging roles of interferon-stimulated genes in the innate mmune response to hepatitis C virus infection

Infection with hepatitis C virus （HCV）, a major viral cause of chronic liver disease, frequently progresses to steatosis and cirrhosis, which can lead to hepatocellular carcinoma. HCV infection strongly induces host responses, such as the activation of the unfolded protein response, autophagy and the innate immune response. Upon HCV infection, the host induces the interferon （IFN）-mediated frontline defense to limit virus replication. Conversely, HCV employs diverse strategies to escape host innate immune surveillance. Type I IFN elicits its antiviral actions by inducing a wide array of IFN-stimulated genes （ISGs）. Nevertheless, the mechanisms by which these ISGs participate in IFN-mediated anti-HCV actions remain largely unknown. In this review, we first outline the signaling pathways known to be involved in the production of type I IFN and ISGs and the tactics that HCV uses to subvert innate immunity. Then, we summarize the effector mechanisms of scaffold ISGs known to modulate IFN function in HCV replication. We also highlight the potential functions of emerging ISGs, which were identified from genome-wide siRNA screens, in HCV replication. Finally, we discuss the functions of several cellular determinants critical for regulating host immunity in HCV replication. This review will provide a basis for understanding the complexity and functionality of the pleiotropic IFN system in HCV infection. Elucidation of the specificity and the mode of action of these emerging ISGs will also help to identify novel cellular targets against which effective HCV therapeutics can be developed.

Effect of Rorrico, extracted from group of Chinese medicines, on influenza A and H1N1 infections

OBJECTIVE: To investigate the therapeutic and preventive effects of Rorrico on influenza, especially influenza A viral infection, including swine flu(H1N1)in humans.METHODS: Eighty-nine subjects were recruited in Hong Kong and Macao, and divided into treatment group(TG) and prevention group(PG) based on their influenza A and swine flu symptoms. All subjects were prescribed Rorrico or placebo, and monitored by a Chinese medicine practitioner. Blood samples were collected before and after 7-day Rorrico or placebo treatment for laboratory investigations.RESULTS: After treatment, there were some full recoveries and obvious relief of onset symptoms in the TG. Blood test results showed that Rorrico produced(a) no adverse effects on subjects' renal andliver functions, muscle enzyme and hematological status,(b) no up-regulation of pro-inflammatory cytokines tumour necrosis factor-a and interleukin-18 in both TG and PG,(c) mild yet statistically significant elevation of plasma mannose-binding lectin(MBL) in PG.CONCLUSION: Rorrico has no up-regulating effect on the participants' immune response, or, equally likely, the immuno-modulatory effects of Rorrico do not non-specifically or unnecessarily promote inflammation when not required. It is possible that oral administration of Rorrico can promote hepatic synthesis of MBL in healthy PG subjects, thereby conferring increased protection against infection.
