试验旨在确定饲喂3种玉米粉加工副产品对泌乳荷斯坦奶牛采食量、产奶量、瘤胃发酵和消化率的影响。试验1为3种玉米粉加工副产品按15%日粮干物质(DM)饲喂平均体重(625±81kg)、泌乳(116±33d)的28头荷斯坦奶牛,测定对干物质采食量和产奶量的影响。试验2为同样的粮饲喂平均体重(677±41kg)、泌乳(144±5d)的4头装有瘤胃套管的经产荷斯坦奶牛,确定对瘤胃发酵和消化率的影响。试验采用4×4拉丁方设计,在4个21d的周期,奶牛和处理随机分配。15%玉米粉加工副产品代替粗料和精料部分配合日粮,处理包括:①0%副产品(对照组),②含可溶物干酒粕(DDGS组),③脱水玉米胚饼粉(胚芽组),④高蛋白干酒粕(HPDDG组)。每天记录采食量,各个周期的第19～21d,收集奶样,分析主要成分。试验2的第21天,饲喂后在24h内的10个时间点收集瘤胃液体。结果表明,在试验1中,胚芽组(24.3kg·d-1)和DDGS组(23.8kg·d-1)的干物质采食量较高,而DDGS组(22.4kg·d-1)与对照组(22.9kg·d-1)和HPDDG组(22.4kg·d-1)无显著差异。产奶量与DM采食量相对应,胚芽组(32.1kg·d-1)和DDGS组(30.9kg·d-1)的干物质采食量较高,而DDGS组(22.4kg·d-1)与对照组(30.6kg·d-1)和HPDDG组(30.3kg·d-1)无显著差异。乳脂量、乳蛋白量和3.5%乳脂肪校正乳(FCM)相似,分别为1.1±0.1、0.9±0.03、31.7±1.3kg·d-1。HPDDG组(15.9mg·d-1·l-1)和胚芽组(15.5mg·d-1·l-1)的乳中尿素氮比对照组(15.0mg·d-·1l-1)和DDGS组(14.9mg·d-1·l-1)的相应值要高。试验2的处理组之间的DM采食量和产奶量无差别,平均为26.1±2.3和28.3±3.9kg·d-1。瘤胃pH(6.26±0.08)和挥发性脂肪酸总浓度(125.3±4.2mM)相似。对照组的乙酸盐浓度比DDGS组、胚芽组和HPDDG组高(81.7,75.8、75.0、78.4mM)。处理组之间的丙酸盐和丁酸盐浓度无差别,分别为27.8±1.2和14.3±0.9mM。对照组、胚芽组和HPDDG组的醋酸盐与丙酸盐的比率比DDGS组的要高(3.02、2.88、2.91,2.62)。处理组之间的干物质、有机物和中性洗涤纤维消化率相似,分别为63.5±2.7%、67.3±2.2%和43.5±4.2%。试验1的奶牛产奶量受干物质采食量的影响,而主要乳成分的产量则不受影响。试验结果表明,配合成的奶牛日粮玉米粉加工副产品可占日粮干物质的15%,同时能保持或提高奶牛的干物质采食量以及产奶量和乳成分的产量。

试验选取内洛尔公牛127头,分为低剩余饲料摄入量(RFI)和高剩余饲料摄入量,通过3个选择线和5个试验来确定RFI和胴体品质、肉品质之间的关系。对生产性能、胴体性状和背最长肌进行评估。包括RFI的固定效应、选择线、RFI等级和选择线之间的相互作用作为混合模型的主要变量,屠宰年龄作为协变量,并在试验过程中随机安排,通过T检验比较。平均饲料采食量差异为0.962 kg·d^(-1),低RFI组动物消耗量为0.479kg·d^(-1),高RFI组消耗量为0.483kg·d^(-1),高于预期水准。不同RFI组间各检测指标无差异,RFI对胴体、肉质无不利影响。低RFI可提高内洛尔公牛饲料效率和肉品质。

Identification in lupin Seed of a Serine- Endopeptidase Activity Cleaving between Twin Arginine Pairs and Causing limited Proteolysis of Seed Storage Proteins

The occurrence of twin-arginine motifs （-R-R-） in the amino acid sequences of animal pro-proteins frequently defines the cleavage site（s） for their structural/functional maturation. No information is available on the presence and possible biological meaning of these motifs in the seed storage proteins. In this work, a novel endopeptidase activity with cleavage specificity to twin-arginine pairs has been detected in mature dry lupinus albus seeds. The endopeptidase was tested with a number of endogenous and exogenous protein substrates, which were selected according to the pres- ence of one or more twin-arginine residue motifs in their amino acid sequences. The observed hydrolysis patterns were limited and highly specific. Partial proteolysis led to stable polypeptide fragments that were characterized by 1- and 2-D electrophoresis. Selected polypeptides were submitted to N-terminal amino acid sequencing and mass spectrometry anal- yses, These approaches, supported by bioinformatic analysis of the available sequences, allowed the conclusion that the polypeptide cleavage events had occurred at the peptide bonds comprised between twin-arginine residue pairs with all tested protein substrates. The endopeptidase activity was inhibited by 4-（2-AminoEthyl）Benzene-Sulphonyl Fluoride hy- drochloride （AEBSF）, leupeptin, and serine proteinase protein inhibitors, while it was not affected by pepstatin, trans- EpoxysuccinyI-l-leucylamido（4-guanidino）butane （E64）, and ethylenediaminetetraacetic acid （EDTA）, thus qualifying the Arg-Arg cleaving enzyme as a serine endopeptidase. The structural features of storage proteins from lupin and other legume seeds strongly support the hypothesis that the occurrence of an endopeptidase activity cleaving -R-R- bonds may be functional to facilitate their degradation at germination and possibly generate polypeptide fragments with specific biological activity.

Urinary corticosterone and normetanephrine levels after voluntary wheel and forced treadmill running in the db/db mouse

Exercise is recommended for the treatment of type 2 diabetes because of its benefits on body weight and glycemic control. our recent work using the db/db mouse, a model that mimics the phenotype of type 2 diabetes, demonstrated that forced treadmill training exerted detrimental effects on obesity, hyperglycemia and insulin resistance. We investigated whether this response is explained by increased corticosterone and norepinephrine secretion, measured as urinary byproducts, since these hormones are known to alter glucose homeostasis. Male db/db mice and lean littermates serving as controls, were assigned to sedentary, voluntary wheel, and forced treadmill training groups for a period of 5 weeks. After 5 weeks of treadmill running, db/db mice remained hyperglycemic compared to sedentary db/db mice and were hyperinsulinemic compared to db/db voluntary runners. Urine glucose and corticosterone levels were also highest in db/db treadmill runners compared to all groups. Urine normetanephrine levels, although lower in db/db mice compared to control mice, were increased after treadmill running. our results indicate that treadmill running leads to perturbations in plasma levels of hormones associated with glucose homeostasis. A greater stress response may be invoked by treadmill training, worsening glycemic control in this model of type 2 diabetes.

Use of Panoramic Radiographs to Detect Signs of osteoporosis in Edentulous

objective: The objective is to analyze the importance of panoramic and carpal radiographs for evaluation of edentulous individuals in different ages, demonstrating the contribution of different dental radiographic techniques in detecting patients with signs of osteoporosis. Methods: The study was developed using panoramic and carpal digital radiographs of 30 edentulous women (age ranged 50 - 90 years). The panoramic radiographs were performed to measure the thickness of the mental index—MI and gonial index—GI, and evaluation of the morphology of the mandibular cortical shape (Klemetti classification). The carpal radiographs were performed to measure the cortical thickness of the 2nd, 3rd and 4th metacarpals (metacarpal index). Statistics analyzed the relationship of these indices with the patient’s age and the contribution of dental radiographs in detecting patients at risk for osteoporosis. Results: The average age of subjects was 68.43 years. In the analysis of cortical thickness of the panoramic radiographs, the mean values of mental index (MI) and gonial index (GI) for the age group 50 to 59 years were considered normal values (≥3.1 mm and ≥1.2 mm , respectively). To analyze the morphology of the cortex, the C3 group corresponded to the majority of cases (43.33%). The higher the age range, more morphological changes were observed (sum of the characteristics of groups C2 and C3). For the hand-wrist radiographs, the highest values of the metacarpal index (BMI) were found in the youngest age group (50 - 59 years). Conclusions: There was positive correlation between age and indices of quantitative assessment (IM, IG, BMI) and qualitative (analysis Klemetti) on panoramic and carpal radiographs. Age is a risk factor for the onset of osteoporosis.

objective: To measure white matter (WM) and gray matter (GM) atrophy and lesion load in a large population of patients with multiple sclerosis (MS) using a fully automated, operatorindependent,multiparametric segmentation method. Methods: The study population consisted of 597 patients with MS and 104 control subjects. The MRI parameters were abnormal WM fraction (AWM-f), global WM-f (gWM-f), and GM fraction (GM-f). Results: Significant differences between patients with MS and control subjects included higher AWM-f and reduced gWM-f and GM-f. MRI data showed significant differences between patients with relapsing-remitting and secondary progressive forms of MS. Significant correlations between MRI parameters and between MRI and clinical data were found. Conclusions: Patients with multiple sclerosis have significant atrophy of both white matter (WM) and gray matter (GM); secondary progressive patients have significantly more atrophy of both WM and GM than do relapsing-remitting patients and a significantly higher lesion load (abnormal WM fraction); lesion load is related to both WM and even more to GM atrophy; lesion load and WM and GM atrophy are significantly related to Expanded Disability Status Scale score and age at onset (suggesting that the younger the age at disease onset, the worse the lesion load and brain atrophy); and GM atrophy is the most significant MRI variable in determining the final disability.
